Malignancy of the larynx: (Experimentation on Animal, Construction of Biologic Neo-Larynx and Rehabilitation of the Laryngectomee-20 Years Experience).

Squamous cell carcinoma is by far the commonest malignancy of the larynx and I would confine my present paper mainly to this along with the management and post-operative rehabilitation after total laryngectomy. High survival rate in majority of the cases, if diagnosed and treated early and adequately, provokes and encourages the interested laryngologist to consider not only about performing effective surgery, including on those cases where radiotherapy is inadequate or has failed, but also resorting to rehabilitative surgical procedures after total laryngectomy with or without neck dissection,

22-kHz ultrasonic vocalization in rats as an index of anxiety but not fear: behavioral and pharmacological modulation of affective state.

Ultrasonic vocalization (USV) was found useful for differentiating fear and anxiety in rats. These affective states were established through a Pavlovian conditioning procedure. Danger stimulus, preceding unavoidable tail shock, elicited acute fear. Intertrial situational cues evoked anxiety. A safety signal (SS) indicating the omission of shock inhibited fear. Sustained 22-kHz USV characterized anxiety and was present between trials. A signal of danger resulted in immediate inhibition of vocalization, while a SS reversed this effect. These results are discussed in the context of three theories

Neural correlates of frog calling: production by two semi-independent generators.

The anterior preoptic nuclei of the isolated brainstem of male, Northern leopard frogs (Rana p. pipiens) were stimulated electrically and neural correlates of mating calling recorded from the rhombencephalic mating calling pattern generator. Lesions of discrete areas of the brainstem showed that the mating calling generator is separable into two generators, the pretrigeminal nucleus and the classical pulmonary respiration generator (which is approximately co-extensive with the motor nuclei IX-X). Each of these still can produce pulses when isolated from the other. Their interaction changes the

Voice production in a MRI-based subject-specific vocal fold model with parametrically controlled medial surface shape.

The goal of this study was to investigate how realistic changes in medial surface shape, as occur in human phonation, affect voice production. In a parametric magnetic resonance imaging-based three-dimensional vocal fold model, the superior and inferior portions of the medial surface were systematically manipulated to produce different medial surface contours similar to those observed in previous excised larynx and in vivo canine larynx experiments. Voice simulations were performed to investigate the differences in the resulting voice production. The results showed that both superior-medial bu

Acquired idiopathic laryngeal paralysis as a prominent feature of generalised neuromuscular disease in 39 dogs.

A retrospective evaluation of 17 dogs treated surgically for idiopathic acquired laryngeal paralysis demonstrated a marked variability in outcome, with many dogs continuing to exhibit weakness and exercise tolerance. In a subsequent prospective study, 22 consecutive affected dogs were tested for neurological deficits by neurological examination, electrodiagnostic evaluation, and blood analysis to measure thyroxine and thyroid-stimulating hormone and to detect any evidence of myasthenia gravis. Clinical neurological deficits and/or electrodiagnostic abnormalities were found in each case. There

Laryngeal reinnervation.

Laryngeal reinnervation refers to any of a number of surgical procedures intended to restore neural connections to the larynx, which have usually been lost from some type of trauma (eg, surgical). The nerve function(s) to be restored may be those of the recurrent laryngeal nerve or its subdivisions, those of the superior laryngeal nerve, or both, and they may be motor or sensory. Several different donor nerves are available and have been described. The technique used may be direct end-to-end anastomosis (neurorrhaphy), direct implantation of a nerve ending into a muscle, the nerve-muscle pedic

Ultrastructural and histochemical study of the neuromuscular junctions in the denervated intrinsic laryngeal muscle of the cat.

Morphological changes and acetylcholinesterase (AchE) activity in the neuromuscular junctions (NMJ) of the normal and denervated posterior cricoarytenoid muscle (PCA muscle) of the cat were studied. Two days after denervation, the nerve terminals at the NMJ had almost disappeared. Six weeks after denervation, intensity of AchE activity at the former junctional site (FJS) was unchanged histochemically. At this stage, primary synaptic clefts were distorted and the Schwann's cells covered the FJS. Fourteen weeks after denervation, AchE activity at the FJS had decreased in contrast to that of the

Developmental delays and reduced pup ultrasonic vocalizations but normal sociability in mice lacking the postsynaptic cell adhesion protein neuroligin2.

Mutations in neurexin and neuroligin genes have been associated with neurodevelopmental disabilities including autism. Autism spectrum disorder is diagnosed by aberrant reciprocal social interactions, deficits in social communication, and repetitive, stereotyped patterns of behaviors, along with narrow restricted interests. Mouse models have been successfully used to study physiological and behavioral outcomes of mutations in the trans-synaptic neurexin-neuroligin complex. To further understand the behavioral consequences of Neuroligin2 (NLGN2) mutations, we assessed several behavioral phenoty

Estradiol and song affect female zebra finch behavior independent of dopamine in the striatum.

Female songbirds display preferences for certain song characteristics, but the neural and hormonal mechanisms mediating these preferences are not fully clear. The present study sought to further explore the role of estradiol, as well as assess potential roles of dopaminergic systems, on behavioral responses to song. Adult female zebra finches were treated with estradiol and exposed to tutored or untutored song or silence. Behavior was quantified and neurochemistry of the nucleus accumbens and striatum was examined with high performance liquid chromatography. As a control, the responses of thes

The effects of click rate on the auditory brainstem response of bottlenose dolphins.

Rate manipulations can be used to study adaptation processes in the auditory nerve and brainstem. For this reason, rate effects on the click-evoked auditory brainstem response (ABR) have been investigated in many mammals, including humans. In this study, click-evoked ABRs were obtained in eight bottlenose dolphins (Tursiops truncatus) while varying stimulus rate using both conventional averaging and maximum length sequences (MLSs), which allow disentangling ABRs that overlap in time and thus permit the study of adaptation at high rates. Dolphins varied in age and upper cutoff frequency of hear

Nociceptive vocalization response in guinea pigs modulated by opioidergic, GABAergic and serotonergic neurotransmission in the dorsal raphe nucleus.

The dorsal raphe nucleus (DRN) is involved in the control of several physiological functions, including nociceptive modulation. This nucleus is one of the main sources of serotonin to the CNS and neuromodulators such as opioids and GABA may be are important for its release. This study evaluated the influence of serotonergic, GABAergic and opioidergic stimulation, as well as their interactions in the DRN, on vocalization nociceptive response during a peripheral noxious stimulus application in guinea pigs. Morphine (1.1 nmol), bicuculline (0.50 nmol) and alpha-methyl-5-HT (1.6 nmol) microinjecti

Response properties of auditory activated cells in the occipital cortex of the blind mole rat: an electrophysiological study.

Previous studies have demonstrated that despite its blindness, the subterranean blind mole rat (Spalax ehrenbergi) possesses a noticeable lateral geniculate nucleus and a typical cyto-architectural occipital cortex that are reciprocally connected. These two areas, as revealed by the metabolic tracer 2-deoxyglucose, are activated by auditory stimuli. Using single unit recordings, we show that about 57% of 325 cells located within the occipital cortex of anesthetized mole rats responded to at least one of the following auditory stimuli--white noise, pure tones, clicks, and amplitude modulated to

Post-transcriptional regulation of zenk expression associated with zebra finch vocal development.

In the male zebra finch, highly variable juvenile song and stereotyped adult song induce mRNA expression of the immediate early gene zenk in telencephalon. However, the functional consequences of this behavior-driven gene expression remain unknown. Here we characterize the developmental expression of zenk mRNA and protein in two forebrain song regions (HVC, the higher vocal center, and RA, the robust nucleus of the archistriatum). In HVC, singing results in similar percentages of cells producing zenk mRNA and zenk protein at different stages of vocal development. Similarly, song behavior at al

Sensory nerve-derived signaling coordinates oropharyngeal structural organization that supports suckling and vocalization in neonatal mice.

Proper oropharyngeal function is essential for suckling, feeding, and speech, and relies on the coordinated development of the palate, oropharyngeal musculature, and the cranial nerves that control them. Disruption of this integration leads to severe neonatal complications. However, how neuromuscular architecture is developmentally coordinated to support oropharyngeal function remains unclear. Using single-cell and spatial transcriptomics, we identify trigeminal nerve-derived GDF11 as a crucial regulator of soft palatal muscle architecture that acts through cranial neural crest-derived perimys

Juvenile manifestation of ultrasound communication deficits in the neuroligin-4 null mutant mouse model of autism.

Neuroligin-4 (Nlgn4) is a member of the neuroligin family of postsynaptic cell adhesion molecules. Loss-of-function mutations of NLGN4 are among the most frequent, known genetic causes of heritable autism. Adult Nlgn4 null mutant (Nlgn4(-/-)) mice are a construct valid model of human autism, with both genders displaying a remarkable autistic phenotype, including deficits in social interaction and communication as well as restricted and repetitive behaviors. In contrast to adults, autism-related abnormalities in neonatal and juvenile Nlgn4(-/-) mice have not been reported yet. The present study

Yolk testosterone levels and offspring phenotype correlate with parental age in a precocial bird.

Parents, and particularly mothers, can influence their offspring's development in non-genetic ways. Maternal effects can occur during the mothering phase as well as during the embryonic phase. Prenatal maternal effects in birds can be mediated by yolk steroid hormones that influence subsequent offspring development. Studies have focused mainly on the influence of laying females' living conditions on yolk hormonal contents, and rarely on the effects of individual characteristics. Here, we investigated prenatal influence of parent age on yolk steroid levels and on offspring phenotype. We compare

Hippocampal lesions impair spatial memory performance, but not song--a developmental study of independent memory systems in the zebra finch.

Songbirds demonstrate song- and spatial-learning, forms of memory that appear distinct in formal characteristics and fitting the descriptions and criteria of procedural and episodic-like memory function, respectively. As in other vertebrates, the neural pathways underlying these forms of memory may also be dissociable, and include the corresponding song circuit and hippocampus (HP). Whether (or not) these two memory systems interact is unknown. Interestingly, the HP distinguishes itself as a site of immediate early gene expression in response to song and as a site of estrogen synthesis, a ster

Induction of physical dependence upon ethanol and the associated behavioral changes in rats.

This paper reports findings relative to a simple, rapid and reproducible technique for the induction of physical dependence upon ethanol in the rat. The dependence was induced by intragastric intubation of 20% (w/v) ethanol solutions at 9-15 g/kg in 3-5 fractional doses daily for 4 days, maintaining blood ethanol concentrations above a threshold level sufficient to sustain observable sedation throughout the entire period of intubation. Two phases were distinguished during the withdrawal period: 1. Prodromal detoxication, characterized by a spectrum of signs and responses of diminishing severit
