Selection of controls in case-control studies on maternal medication use and risk of birth defects.

BACKGROUND: In case-control studies on teratogenic risks of maternal drug use during pregnancy, the use of normal or malformed controls may lead to recall-bias or selection bias. This can be avoided by using controls with a genetic disorder. However, researchers are hesitant to use these as controls because it is unknown whether their selection is independent of exposure status. The aim of this study is to investigate whether first trimester drug use among mothers of children with genetic disorders is representative for the "general pregnant population". METHODS: From a birth defects registry

Assigning Targetable Molecular Pathways to Transdiagnostic Subgroups Across Autism and Related Neurodevelopmental Disorders.

Significant genetic, behavioural and neuroanatomic heterogeneity is common in autism spectrum- and related- neurodevelopmental disorders (NDDs). This heterogeneity constrains the development of effective therapies for diverse patients in precision medicine paradigms. This has led to the search for subgroups of individuals having common etiologic factors/biology (e.g., genetic pathways), thus creating potential uniformity in prognosis and/or treatment response. Despite NDDs having a strong genetic component, only ~15-20% of individuals will present with a specific rare genetic variant considere

[Cognitive, emotional and behavioral impact of an uncertain outcome after study of BRCA1/2: review of the literature].

BACKGROUND: Recent advances in oncogenetics have enabled the development of tests for predisposition to breast and ovarian cancers. Where no mutation has been identified in the BRCA1 or 2 genes, the proband (first person tested in a family with a genetic risk) can receive an uncertain outcome: negative inconclusive or identification of a variant of unknown clinical significance. From the demonstration of such outcomes, their psychological impact has been studied among women concerned. OBJECTIVE: The purpose of this article is to summarize the results of studies about the impact of delivering

Updated global prevalence and ethnic diversity of von Willebrand disease based on population genetics analysis.

von Willebrand disease (VWD) is known as the most common inherited bleeding disorder. Despite this, some uncertainty remains regarding its prevalence and the distribution of different phenotypes worldwide and across ethnic groups. By leveraging genetic epidemiology approaches that enable population-scale estimates, the prevalence of VWD and its diverse phenotypes can be assessed more precisely. We estimated the global VWD prevalence and its distribution across distinct ethnic groups using the largest and most widely used publicly available genetic database. Available exome/genome sequencing da

Rheumatoid arthritis increases risk of atherosclerosis: A Mendelian randomization study.

Understanding of the relationship between rheumatoid arthritis (RA) and atherosclerosis (AS) remains limited by the shortcomings of previous observational studies. This study investigated the causal relationship between RA and AS risks using Mendelian randomization (MR) analysis. Genetic data related to RA and AS were sourced from the genome-wide association studies database and FinnGen Biobank gene database, respectively. Standard screening metrics were applied and palindromic single nucleotide polymorphisms were processed. The initial analysis employed the inverse-variance weighted method fo

Dental anomalies in syndromes displaying hypertrichosis in the clinical spectrum.

Hypertrichosis and dental anomalies may occur alone or in combination in the spectrum of many syndromes. To identify genetic entities characterized by hypertrichosis and dental anomalies, a search was performed in the Mendelian Inheritance in Man database with the terms "hypertrichosis" or "hirsutism" and "tooth" or "dental abnormalities." Nondependent androgen metabolism disturbances were classified as hypertrichosis. Genetic entities with hypertrichosis and dental anomalies were included in the study. Additional searches were performed in the PubMed and Orphanet databases, when necessary, in
