Enhancing oxygen utilization and mitigating oxidative stress in Tibetan chickens for adaptation to high-altitude hypoxia.

Tibetan chicken (TBC) is one of the native poultry species that is well adapted to the high-altitude environment of the Qinghai-Tibet Plateau. To elucidate the genetic mechanisms underlying adaptation, the transcriptomes of five tissues (heart (HE), lung (LU), liver (LI), ovary (OV), and abdominal fat (AB)) were compared between TBCs and Roman chickens (RMCs) inhabiting the plateau for one year. Moreover, weighted gene co-expression network analysis (WGCNA) was applied to detect tissue-associated modules and hub genes. A total of 1105, 239, 400, 483, and 275 differentially expressed genes (DEG

pH-Isomerizable Acylhydrazone-Based Ionizable Lipids for Spleen-Targeted mRNA Vaccines.

Effectively and safely amplifying systemic immunity against heterogeneous and immunosuppressive cancers remains highly challenging. Here, we report a library of acylhydrazone-based ionizable lipids (AHzILs) that exploit pH-responsive E/Z isomerization to dynamically tune the membrane properties of mRNA-encapsulated lipid nanoparticles (LNPs), thereby achieving spleen targeting, efficient endosomal escape, and robust immune activation. These isomerizable mRNA vaccines acquired a protein corona enriched in mannose-binding protein A and vitronectin, promoting preferential uptake by antigen-presen

Downregulated expression of dual-specificity phosphatase-1 in multiple myeloma as a predictor of poor survival outcomes.

OBJECTIVES: Multiple myeloma (MM) is an incurable hematological malignancy, Dual-specificity phosphatase-1 (DUSP1) plays a crucial role in the initiation and progression of various tumors. Here, we aim to elucidate the role of DUSP1 in MM. METHODS: DUSP1 mRNA expression was analyzed based on public datasets, and protein expression was determined by immunohistochemistry. The association between DUSP1 and clinicopathological characteristics, as well as its impact on survival, were investigated. Protein-protein interaction and gene set enrichment analysis were performed. RESULTS: Low DUSP1 expr

Tumor Microenvironment Lactate: Is It a Cancer Progression Marker, Immunosuppressant, and Therapeutic Target?

The "Warburg effect" is a term coined a century ago for the preferential use of glycolysis over aerobic respiration in tumor cells for energy production, even under aerobic conditions. Although this is a less efficient mechanism of generating energy from glucose, aerobic glycolysis, in addition to the canonical anaerobic glycolysis, is an effective means of lactate production. The abundant waste product, lactate, yielded by the dual glycolysis in a tumor, has been discovered to be a major biomolecule that drives cancer progression. Lactate is a metabolic energy source that, via cell membrane l

Tanshinone IIA Promotes Hippocampal Neurogenesis in ApoE-/- Mice Through cAMP/PKA/CREB/BDNF Signaling Pathway.

New cells are generated from neural stem cells (NSCs) in the subgranular zone (SGZ) of the dentate gyrus of the hippocampus throughout life to shape brain function. Apolipoprotein E (ApoE) deficiency impairs hippocampal dentate gyrus development by affecting the neural progenitor pool over time. Impaired adult hippocampal neurogenesis has been reported in human ApoE4 and ApoE-knockout (ApoE-/-) mice. The ApoE gene is the strongest genetic risk factor for late-onset Alzheimer's disease (AD). Our previous studies indicated that tanshinone IIA (tan IIA) has a broad range of pharmacological action

Iron oxide-mediated enhancement of extracellular electron transfer and symbiosis in consortium of electroactive bacteria and microalgae for wastewater treatment.

This study explores the role of α-Fe₂O₃ in improving extracellular electron transfer (EET) and symbiotic interactions between electroactive Shewanella oneidensis MR-1, its gene-deficient mutants (ΔmtrC, ΔomcA, and ΔcymA), and microalgae (Chlorella vulgaris). The iron oxide facilitates the efficient transfer of electrons generated by MR-1 to microalgal photosystem via the pathway of CymA-MtrC-OmcA to α-Fe₂O₃. This process enhances the removals of TOC, TN, and NH₄⁺-N in the MR-1 bacterial-algal consortium by 9.99%, 12.32%, and 52.25% respectively via OmcA regulation while boosting phosphorus rem

Antigen reactivity defines tissue-resident memory and exhausted T cells in tumours.

CD8 + T cells are a key weapon in the therapeutic armamentarium against cancer. While CD8 + CD103 + T cells with a tissue-resident memory T (T RM ) cell phenotype have been favourably correlated with patient prognoses 1-6 , the tumour microenvironment also contains dysfunctional exhausted T (T EX ) cells that exhibit a myriad of T RM -like features, leading to conflation of these two populations. Here, we deconvolute T RM and T EX cells within the intratumoural CD8 + CD103 + T cell pool across human cancers, ascribing markers and gene signatures that distinguish these CD8 + populations and ena

Metabolic response of tellurite resistant Bacillus altitudinis strain 3W19 highlights the potential as a model organism for bioremediation.

Contaminated environments can pose new challenges when new contaminants appear and can select organisms with new genetic and metabolic strategies. The increased presence of Te(IV) in the environment is becoming more important. This highlights how underexplored the investigation of how bacteria molecularly respond to less common environmental contaminants, such as tellurite when compared to other metals/ metalloids. Understanding what tools an organism uses from its genetic pool when responding to a new contaminant requires a multiple-technique approach, such as metabolic tests and differential

Role of thymus in health and disease.

The thymus is a primary lymphoid organ, essential for the development of T-cells that will protect from invading pathogens, immune disorders, and cancer. The thymus decreases in size and cellularity with age referred to as thymus involution or atrophy. This involution causes decreased T-cell development and decreased naive T-cell emigration to the periphery, increased proportion of memory T cells, and a restricted, altered T-cell receptor (TCR) repertoire. The changes in composition and function of the circulating T cell pool as a result of thymic involution led to increased susceptibility to

Genetic Engineering of Talaromyces marneffei to Enhance Siderophore Production and Preliminary Testing for Medical Application Potential.

Siderophores are compounds with low molecular weight with a high affinity and specificity for ferric iron, which is produced by bacteria and fungi. Fungal siderophores have been characterized and their feasibility for clinical applications has been investigated. Fungi may be limited in slow growth and low siderophore production; however, they have advantages of high diversity and affinity. Hence, the purpose of this study was to generate a genetically modified strain in Talaromyces marneffei that enhanced siderophore production and to identify the characteristics of siderophore to guide its me

ASCORBATE PEROXIDASE6 delays the onset of age-dependent leaf senescence.

Age-dependent changes in reactive oxygen species (ROS) levels are critical in leaf senescence. While H2O2-reducing enzymes such as catalases and cytosolic ASCORBATE PEROXIDASE1 (APX1) tightly control the oxidative load during senescence, their regulation and function are not specific to senescence. Previously, we identified the role of ASCORBATE PEROXIDASE6 (APX6) during seed maturation in Arabidopsis (Arabidopsis thaliana). Here, we show that APX6 is a bona fide senescence-associated gene. APX6 expression is specifically induced in aging leaves and in response to senescence-promoting stimuli

Analysis of 16S rRNA genes reveals reduced Fusobacterial community diversity when translocating from saliva to GI sites.

Fusobacterium nucleatum is a Gram-negative oral commensal anaerobe which has been increasingly implicated in various gastrointestinal (GI) disorders, including inflammatory bowel disease, appendicitis, GI cancers. The oral cavity harbors a diverse group of Fusobacterium, and it is postulated that F. nucleatum in the GI tract originate from the mouth. It is not known, however, if all oral Fusobacterium translocate to the GI sites with equal efficiencies. Therefore, we amplified 16S rRNA genes of F. nucleatum and F. periodonticum, two closely related oral species from matched saliva, gastric asp

Distinct iron homeostasis in C57BL/6 and Balb/c mouse strains.

C57BL/6 (BL6) and Balb/c mice exhibit prototypical Th1- and Th2-dominant immune predispositions, respectively. Iron is a proinflammatory metal ion; however, limited information is documented on the differences in iron homeostasis between BL6 and Balb/c strains. The objective of this study was to investigate the extent to which strain-level differences in these mice dictates the regulation of iron homeostasis during physiologic and inflammatory conditions. At basal levels, Balb/c mice displayed significantly higher levels of iron in systemic circulation and tissue compared to BL6 mice. Moreover

Bayesian GWAS with Structured and Non-Local Priors.

MOTIVATION: The flexibility of a Bayesian framework is promising for GWAS, but current approaches can benefit from more informative prior models. We introduce a novel Bayesian approach to GWAS, called Structured and Non-Local Priors (SNLPs) GWAS, that improves over existing methods in two important ways. First, we describe a model that allows for a marker's gene-parent membership and other characteristics to influence its probability of association with an outcome. Second, we describe a non-local alternative model for differential minor allele rates at each marker, in which the null and altern

Mechanisms of Oncogenesis by HTLV-1 Tax.

The human T-cell lymphotropic virus type 1 (HTLV-1) is the etiological agent of adult T-cell leukemia/lymphoma (ATLL), a neoplasm of CD4+CD25+ T cells that occurs in 2-5% of infected individuals after decades of asymptomatic latent infection. Multiple HTLV-1-encoded regulatory proteins, including Tax and HTLV-1 basic leucine zipper factor (HBZ), play key roles in viral persistence and latency. The HTLV-1 Tax oncoprotein interacts with a plethora of host cellular proteins to regulate viral gene expression and also promote the aberrant activation of signaling pathways such as NF-κB to drive clon

Huntingtin protein maintains balanced energetics in mouse cardiomyocytes.

Huntingtin (HTT) is a multifunctional protein crucial for proper embryogenesis and nervous system development. Mutation of a single allele in gene coded this protein results in the Huntington׳s disease (HD). There is growing evidence of cardiovascular system pathologies coexisting with the neurological symptoms in HD patients. Thus, this study aims to establish the role of huntingtin protein in cardiomyocytes cellular energy and nucleotides metabolism. We used HTT KO mice embryonic stem cells (ESC) obtained with CRISPR method, wild type mice ESC treated by CRISPR with Scramble control sequence
