Lithium-induced neuroprotection in bipolar disorder: Translational insights into cytoskeletal and proteomic mechanisms.

The pharmacotherapy of lithium plays mainstay in the modulation of bipolar disorder (BD) and controls other neuropsychiatric conditions such as prophylactic and mood episodes. However, its neurotrophic properties embrassing hypothetically converged synaptic regulation to BD remains elusive. Several studies intricate evidence of lithium targetting 'microtubular associated protein-2 (MAP2)' to endure neuroprotection. Synthesis of multiple single nucleiotide gene polymorphisms of Snap-25 gene is considered a milestone in MAP2 alteration. In fact, upregulated NAA (N-acetyl-aspartate), oxidative st

Region-specific response of central microglial cells to sciatic nerve demyelination through sensory and motor pathways.

Peripheral nerve injury can cause changes in microglial cells on the spinal dorsal and ventral horns. This region-specific response implies that central microglial cells could be activated through both sensory and motor pathways. In order to further determine how peripheral nerve injury activates central microglial cells through neural pathways, the sciatic nerve was selected as the target for neural tract tracing and demyelination. Firstly, we used cholera toxin subunit B (CTB) to map the central sensory and motor territories of the sciatic nerve. Secondly, we applied lysophosphatidylcholine …

Food restriction, refeeding, and gastric fill fail to affect emesis in musk shrews.

Nausea and emesis are common side effects of gastrointestinal disease. Reports indicate that ghrelin and endocannabinoids, agents that stimu … minent effects on food intake, neural controls for feeding behavior do not significantly affect the neural pathways for emesis. These results may have implications for how we treat patients who experience a constellation of side effects, including nausea and emesis, since stimulating appetite may not necessarily inhibit emetic pathways.

Role of Reward Dysregulation in the Association Between Depression and Obesity: A Systematical Review.

OBJECTIVE: Depression and obesity represent major public health challenges, and their comorbidity suggests underlying shared mechanisms. A comprehensive understanding of the common cognitive-behavioral and neurobiological pathways, particularly those involving reward processing, is essential for developing effective clinical and public health interventions. This systematic review synthesizes evidence regarding the overlap of behavioral and neural substrates or correlates of reward processing and manifestations in depression and obesity. RESULTS: The findings indicate that comorbid depression

Effects of Pseudoloma neurophilia infection on the brain transcriptome in zebrafish (Danio rerio).

Laboratory zebrafish are commonly infected with the intracellular, brain-infecting microsporidian parasite Pseudoloma neurophilia. Chronic P. neurophilia infections induce inflammation in meninges, brain and spinal cord, and have been suggested to affect neural functions since parasite clusters reside inside neurons. However, underlying neural and immunological mechanisms associated with infection have not been explored. Utilizing RNA-sequencing analysis, we found that P. neurophilia infection upregulated 175 and downregulated 45 genes in the zebrafish brain, compared to uninfected controls. F

Cortical Paired Associative Stimulation Influences Response Inhibition: Cortico-cortical and Cortico-subcortical Networks.

BACKGROUND: The ability to stop a suboptimal response is integral to decision making and is commonly impaired across psychiatric disorders. Cortical paired associative stimulation (cPAS) is a form of transcranial magnetic stimulation in which paired pulses can induce plasticity at cortical synapses. Here we used cPAS protocols to target cortico-cortical and cortico-subcortical networks by using different intervals between the paired pulses in an attempt to modify response inhibition. METHODS: A total of 25 healthy volunteers underwent four cPAS sessions in random order 1 week apart: right inf

Rubrocerebellar Feedback Loop Isolates the Interposed Nucleus as an Independent Processor of Corollary Discharge Information in Mice.

Understanding cerebellar contributions to motor coordination requires deeper insight into how the output structures of the cerebellum, the cerebellar nuclei, integrate their inputs and influence downstream motor pathways. The magnocellular red nucleus (RNm), a brainstem premotor structure, is a major target of the interposed nucleus (IN), and has also been described in previous studies to send feedback collaterals to the cerebellum. Because such a pathway is in a key position to provide motor efferent information to the cerebellum, satisfying predictions about the use of corollary discharge in

Have we been asking the right questions when assessing response inhibition in go/no-go tasks with fMRI? A meta-analysis and critical review.

The popular go/no-go paradigm is supposed to ensure a reliable probing of response inhibition mechanisms. Functional magnetic resonance imaging (fMRI) studies have repeatedly found a large number of structures, usually including a right lateralized parieto-frontal network and the pre-supplementary motor area (pre-SMA). However, it is unlikely that all these regions are directly related to the mechanism that actively suppresses the motor command. Since most go/no-go designs involve complex stimulus identification/detection processes, these activations may rather reflect the engagement of differ

An early attentional bias to BEGIN-stimuli of the smoking ritual is accompanied with mesocorticolimbic deactivations in smokers.

RATIONALE: Biased processing of drug-associated stimuli is believed to be a crucial feature of addiction. Particularly, an attentional bias seems to contribute to the disorder's maintenance. Recent studies suggest differential effects for stimuli associated with the beginning (BEGIN-smoking-stimuli) or the terminal stage of the smoking ritual (END-smoking-stimuli), with the former but not the later evoking high cue-reactivity. OBJECTIVE: The current study investigated the neuronal network underlying an attentional bias to BEGIN-smoking-stimuli and END-smoking-stimuli in smokers and tested the

Lasting secondary antiepileptogenesis induced by cingulate kindling.

PURPOSE: To test the validity of a hypothesis that kindling-induced enduring kindling inhibition of the homotopic secondary site is not a primate species-specific effect but occurs also in feline species. METHODS: Five mature male cats were kindled at the anterior (AC) and posterior (PC) cingulate cortex followed by secondary-site kindling and primary-site retest. RESULTS: Kindling was characterized by a marked seizure-stage instability and a progressive afterdischarge threshold elevation in all the animals. A protracted nonconvulsive seizure stage was followed by convulsive evolution and ra

Neural substrates for reflex salivation induced by taste, mechanical, and thermal stimulation of the oral region in decerebrate rats.

In order to investigate the neural mechanisms of reflex salivary secretion, experiments were carried out on anesthetized, decerebrate rats f … ulation, but gave only a slight response to VC stimulation. These results suggest that two distinct neural pathways exist which mediate reflex salivation in the lower brain stem of the rat, i.e., the taste pathway via the NTS and the nociceptive pathway via the trigeminal sensory nuclei.

Theoretical evaluation of the Rosenblueth hypothesis.

Rosenblueth's hypothesis states that atrioventricular (AV) nodal conduction delay and Wenckebach periodicity of AV transmission are not due to overall decremental conduction within the AV node but are due to a single step delay which is caused by a special element or layer of the AV nodal tissue. This paper discusses some theoretical considerations which allow detailed evaluation of the original hypothesis. Two artificial conduction structures which incorporate the Rosenblueth phenomenon are presented and tested by theoretical experiments that consider the potential of these structures to prod

[Results of static perimetry in subclinical optic neuritis in multiple sclerosis].

Static perimetry of the central visual field was performed using Harms apparatus and Friedmann's analyser in 20 M.S. patients with subclinical optic neuritis at least for one eye. Sub-clinical optic neuritis was defined as delayed or absent pattern reversal visual evoked potentials (VEPs) associated with normal routine ophthalmological examination (visual acuity, optic fundi, colour vision, kinetic perimetry). Results in patients were compared to data obtained in 22 control subjects, matched for age. A highly significant loss of mean retinal sensitivity was observed in the 20 central degrees o

Ets1 is required for proper migration and differentiation of the cardiac neural crest.

Defects in cardiac neural crest lead to congenital heart disease through failure of cardiac outflow tract and ventricular septation. In this report, we demonstrate a previously unappreciated role for the transcription factor Ets1 in the regulation of cardiac neural crest development. When bred onto a C57BL/6 genetic background, Ets1(-/-) mice have a nearly complete perinatal lethality. Histologic examination of Ets1(-/-) embryos revealed a membranous ventricular septal defect and an abnormal nodule of cartilage within the heart. Lineage-tracing experiments in Ets1(-/-) mice demonstrated that c

Network model of pathology spread recapitulates neurodegeneration and selective vulnerability in Huntington's Disease.

Huntington's Disease (HD), an autosomal dominant genetic disorder caused by a mutation in the Huntingtin gene (HTT), displays a stereotyped topography in the human brain and a stereotyped progression, initially appearing in the striatum. Like other degenerative diseases, spatial topography of HD is divorced from where implicated genes are expressed, a dissociation whose mechanistic underpinning is not currently understood. Cell autonomous molecular factors characterized by gene expression signatures, including proteolytic and post translational modifications, play a role in vulnerability to di

The posterior vermis of the cerebellum selectively inhibits 10-Hz sympathetic nerve discharge in anesthetized cats.

We studied the changes in inferior cardiac sympathetic nerve discharge (SND) and mean arterial pressure (MAP) produced by aspiration or chemical inactivation (muscimol microinjection) of lobule IX (uvula) of the posterior vermis of the cerebellum in baroreceptor-denervated and baroreceptor-innervated cats anesthetized with urethane. Autospectral analysis was used to decompose SND into its frequency components. Special attention was paid to the question of whether the experimental procedures affected the rhythmic (10-Hz and cardiac-related) components of SND. Aspiration or chemical inactivation

Task-dependent intracortical inhibition is impaired in focal hand dystonia.

We tested whether task-dependent modulation of inhibition within the motor cortex is impaired in patients with dystonia. Paired-pulse transcranial magnetic stimulation (TMS) at an interstimulus interval of 2 msec was used to measure the effect of two different tasks on short ISI intracortical inhibition (SICI) in dystonic and normal subjects. In two experiments, SICI of the fourth dorsal interosseus (4DIO) and abductor pollicis brevis (APB) muscles were measured before and at the end of the training task. In the first experiment, subjects performed a nonselective task consisting of abducting t

Magnetic transcranial brain stimulation: painless evaluation of central motor pathways. Normal values and clinical application in spinal cord diagnostics: upper extremities.

Painless transcranial magnetic stimulation has been performed on 53 healthy adults in order to obtain normal values of central motor pathway conduction times to cervical segments innervating upper limb muscles. Central motor latency was 5.1 msec when recording from the biceps brachii and 5.2 msec when recording from the abductor pollicis brevis or abductor digiti minimi muscles. The plexus motor latency as the conduction time between the cervical motor root and the proximal nerve trunks in the axillary region was 2.2 msec and 2.6 msec when recorded from the abductor pollicis brevis and abducto

Spinal mechanisms of the analgesic action of electroconvulsive shock.

The present study investigated the spinal systems involved in the analgesic action of electroconvulsive shock (ECS). To identify such systems complete spinal transections and discrete lesions within the dorsal half of the spinal cord were performed. Complete spinal transection eliminated ECS analgesia totally, demonstrating that the observed analgesic effect is attributable to neural conduction. Lesions within the region of the dorsolateral funiculus (DLF) caused a pronounced, but incomplete, attenuation of ECS analgesia. Larger lesions of the dorsal aspects of the spinal cord including both t

Cerebellar output regulation by the climbing and mossy fibers with and without the inferior olive.

The activity of the olivocerebellar complex and the structures related in series with it have been studied using the complementary action of harmaline and 3-acetylpyridine to isolate the two principal inputs to the cerebellar Purkinje cells. The activities of the various nuclei as well as the entire brain have been simultaneously monitored using the [14C]2-deoxy-glucose method under the various combined effects of the pharmacological agents. (1) Tremogenic doses of harmaline increased the frequency of discharge in selected parts of the olivocerebellar system, increasing climbing fiber input an
