Tactical Diagnostic Strategies at Native California Ornamental Nurseries for Preventing Phytophthora Havocs in Wildlands.

Native plant restoration efforts in California are continuously challenged by Phytophthora-induced diseases, particularly through nursery introductions, resulting in significant losses and distribution of pathogens into restoration areas. Effective diagnostic strategies can help manage Phytophthora, yet determining the most suitable method, considering specificity, sampling time, resource availability, and expertise, remains uncertain. This study compares the effectiveness of five diagnostic methods: irrigation leachate baiting, ImmunoStrip® for Phytophthora, culture-based isolations, recombin

ADAM17-dependent Autocrine and Paracrine Signaling Promotes Pancreatic Premalignant Progression.

BACKGROUND & AIMS: A Disintegrin and Metalloproteinase 17 (ADAM17) is a membrane-bound sheddase that regulates the release of multiple signaling molecules, including inflammatory mediators and epidermal growth factor receptor (EGFR) ligands. Ligand-driven EGFR activation is essential for pancreatic acinar cell transdifferentiation into metaplastic ducts, which progress to neoplasia in the presence of oncogenic KrasG12D. The aim of this study is to understand how ADAM17 in the tumor and myeloid cells contribute to the initiation and progression of pancreatic tumors. METHODS: KRASG12D-driven pa

Characterization of a bronchoscopically induced transgenic lung cancer pig model for human translatability.

There remains a need for animal models with human translatability in lung cancer (LC) research. Findings in pigs can have a substantial impact owing to their similar anatomy and physiology to humans. Here we present a bronchoscopically induced LC model in Oncopigs carrying inducible KRASG12D and TP53R167H mutations. A total of 12 Oncopigs underwent 29 injections via flexible bronchoscopy: 18 adenovirus-Cre recombinase gene (AdCre) inductions were performed endobronchially (n = 6) and transbronchially (n = 12), and 11 control injections were performed without AdCre. Oncopigs underwent serial ch

Aldh1l1-Cre/ERT2 Drives Flox-Mediated Recombination in Peripheral and CNS Infiltrating Immune Cells in Addition to Astrocytes During CNS Autoimmune Disease.

INTRODUCTION: The transgenic murine Cre/loxP system is deployed to investigate the role of central nervous system (CNS) cell-specific gene alterations in both healthy conditions and models of neurologic disease. The Aldh1l1-Cre/ERT2 line is widely used to target astrocytes with high coverage and specificity within the CNS. Specificity outside the CNS, however, has not been well-characterized, and Aldh1l1-Cre/ERT2-mediated recombination within the spleen has been reported. In many CNS diseases, infiltrating immune cells from the periphery drive or regulate pathogenesis. We tested whether flox-m

Isotype-selective roles of hepatic acetyl-CoA carboxylases in a mouse model of fatty liver disease.

OBJECTIVES: Acetyl-CoA carboxylase enzymes ACC1 and ACC2 promote liver fat storage. Accordingly, ACC inhibition represents a strategy to reverse fatty liver disease and related disorders. Human and rodent studies show that targeting both ACC isotypes can reverse some fatty liver phenotypes, but also result in unwanted metabolic phenotypes including hypertriglyceridemia. The objective of this study was to determine whether liver-selective genetic inhibition of ACC1 or ACC2 individually can reverse fatty liver disease phenotypes without adverse metabolic phenotypes in a mouse model of fatty live

Advances in Nematode Diagnostics: A Comparative Review of Traditional and Modern Identification Techniques

Nematodes, the most diverse group of invertebrate metazoans in the phylum Nematoda, inhabit a wide range of ecological conditions but remain largely understudied, with less than 3% of species explored. They play a vital role in organic matter recycling, and impact plant and animal health. Due to the lack of distinct identification features, taxonomical knowledge about nematodes, is limited. Traditional morphological classification, based on De Man’s indices, faces limitations due to insufficient variations and a shortage of trained taxonomists. While protein-based approaches can assist in iden

Exploring Origin-Dependent Susceptibility of Smooth Muscle Cells to Aortic Diseases Through Intersectional Genetics.

BACKGROUND: The developmental diversity among smooth muscle cells (SMCs) plays a crucial role in segment-specific aortic diseases. However, traditional genetic approaches are inadequate for enabling in vivo analysis of disease susceptibility associated with cellular origin. There is an urgent need to build genetic technologies that target different developmental origins to investigate the mechanisms of aortopathies, thereby facilitating the development of effective therapeutics. METHODS: To address this challenge, we developed an advanced dual recombinase-mediated intersectional genetic syste

Investigating the relationships of structural and functional neural networks of primary visual cortex with engineered AAVs and chemogenetic-fMRI techniques.

Rationale: Neural networks are crucial for brain function, but the relationship between functional and structural networks remains unclear, hindering disease understanding and treatment. This study employs AAVs, MRI reporter gene (AQP1), and chemogenetic-fMRI to explore the relationship between structural and functional connectivity in the mouse primary visual cortex (V1), offering a novel approach to study abnormal brain functions. Methods: The rAAV-PHP.eB-DIO vector encoding AQP1 was used to cross the blood-brain barrier and infect the brain, enabling in vivo diffusion-weighted imaging (DWI-

Magel2 in hypothalamic POMC neurons influences the impact of stress on anxiety-like behavior and spatial learning associated with a food reward in male mice.

INTRODUCTION: Prader-Willi syndrome (PWS) results from a lack of expression in several paternally inherited, imprinted contiguous genes. Among the genes inactivated in PWS, the Magel2 gene is considered a significant contributor to the etiology of the syndrome. The loss of the Magel2 gene causes abnormalities in growth and fertility and increased adiposity with altered metabolism in adulthood, which aligns with some of the pathologies observed in PWS. Given that anxiety is a prominent phenotypic behavior in PWS, we investigate the role of the Magel2 gene, particularly in hypothalamic POMC neur

Nox3 expression and function in retinal ganglion cells and Amacrine cells.

NADPH oxidase 3 (Nox3), expressed in the endolymphatic duct and sac within the vestibule of the inner ear, is essential for otoconia formation. Mice with functionally deficient Nox3 exhibit a "head-tilt" phenotype. Recently, we reported that Nox3 expression in the cochlea is induced by aging, cisplatin treatment, and intense noise exposure, contributing to the primary source of reactive oxygen species (ROS) and causing acquired sensorineural hearing loss. However, its expression and function outside the inner ear remain poorly characterized. To explore novel Nox3 functions, we used Nox3-Cre;td

Rapid, ultrasensitive, and specific RPA-THz system for pathogenic microorganism detection.

Pathogenic microorganisms responsible for infectious diseases pose a significant global threat to human health. Existing detection methods, such as qPCR and ELISA, fail to simultaneously meet the requirements for high sensitivity, high specificity, and rapid detection. This study presents an innovative approach for the rapid, specific, and highly sensitive detection of pathogenic microorganisms, particularly Escherichia coli O157:H7 (E. coli O157:H7) and varicella-zoster virus (VZV), by combining recombinase polymerase amplification (RPA) with terahertz time-domain spectroscopy (THz-TDS). Lysozyme 1 Inflamed CCR2+ Macrophages Promote Obesity-Induced Cardiac Dysfunction.

BACKGROUND: Macrophages are key players in obesity-associated cardiovascular diseases, which are marked by inflammatory and immune alterations. However, the pathophysiological mechanisms underlying macrophage's role in obesity-induced cardiac inflammation are incompletely understood. Our study aimed to identify the key macrophage population involved in obesity-induced cardiac dysfunction and investigate the molecular mechanism that contributes to the inflammatory response. METHODS: In this study, we used single-cell RNA-sequencing analysis of Cd45+CD11b+F4/80+ cardiac macrophages to explore t

CRISPR technology combined with isothermal amplification methods for the diagnosis of Candida albicans infection.

Since Candida albicans, a type of fungus, causes severe infections that pose a significant threat to human health, its rapid detection is critical in clinical antifungal therapy. Traditional fungal diagnostic approaches are largely based on the culture method. This method is time-consuming and laborious, taking about 48-72 h, and cannot identify emerging species, making it unsuitable for critically ill patients with bloodstream infections, sepsis, and so on. Other antigen or nucleic acid amplification-based methods were also found to be unsuitable for Point-of-Care Testing (POCT) diagnosis due

Type selective ablation of postnatal slow and fast fatigue-resistant motor neurons in mice induces late onset kinetic and postural tremor following fiber-type transition and myopathy.

Animals on Earth need to hold postures and execute a series of movements under gravity and atmospheric pressure. VAChT-Cre is a transgenic Cre driver mouse line that expresses Cre recombinase selectively in motor neurons of S-type (slow-twitch fatigue-resistant) and FR-type (fast-twitch fatigue-resistant). Sequential motor unit recruitment is a fundamental principle for fine and smooth locomotion; smaller-diameter motor neurons (S-type, FR-type) first contract low-intensity oxidative type I and type IIa muscle fibers, and thereafter larger-diameter motor neurons (FInt-type, FF-type) are recrui

Schistosomiasis diagnosis: Challenges and opportunities for elimination.

OVERVIEW: The roadmap adopted by the World Health Organization (WHO) for eliminating neglected tropical diseases aims to eliminate schistosomiasis, as a public health concern, by 2030. While progress has been made towards reducing schistosomiasis morbidity control in several sub-Saharan African countries, there is still more that needs to be done. Proper surveillance using accurate diagnostics with acceptable sensitivity and specificity is essential for evaluating the success of all efforts against schistosomiasis. Microscopy, despite its low sensitivity, remains the gold standard approach for

A rapid and sensitive one-pot platform integrating fluorogenic RNA aptamers and CRISPR-Cas13a for visual detection of monkeypox virus.

The recent global upsurge in Monkeypox virus (MPXV) outbreaks underscores the critical need for rapid and precise diagnostic solutions, particularly in resource-constrained settings. The gold standard diagnostic method, qRT-PCR, is hindered by its time-consuming nature, requirement for nucleic acid purification, expensive equipment, and the need for highly trained personnel. Traditional CRISPR/Cas fluorescence assays, relying on trans-cleavage of ssDNA/RNA reporters labeled with costly fluorophores and quenchers, pose challenges that limit their widespread application, especially for point-of-
