Microscopic and molecular evidence in support of rodent as a reservoir for dissemination of Leishmaniasis.

Leishmaniasis is a worldwide public health problem and vector-borne disease. It is caused by a diverse group of protozoan parasites that belong to the genus Leishmania and transmitted to humans through a bite of an infected female sand fly. Leishmaniasis has attained epidemic proportion in Khyber Pakhtunkhwa and raises serious concern over its management. The present research work was conducted in cutaneous leishmaniasis (CL) prevalent village named Surgul of district Kohat, Khyber Pakhtunkhwa with a focus to investigate whether rodent can act as a source for dissemination of leishmanial speci

The Neuroimmunology of Chronic Pain: From Rodents to Humans.

Chronic pain, encompassing conditions, such as low back pain, arthritis, persistent post-surgical pain, fibromyalgia, and neuropathic pain disorders, is highly prevalent but remains poorly treated. The vast majority of therapeutics are directed solely at neurons, despite the fact that signaling between immune cells, glia, and neurons is now recognized as indispensable for the initiation and maintenance of chronic pain. This review highlights recent advances in understanding fundamental neuroimmune signaling mechanisms and novel therapeutic targets in rodent models of chronic pain. We further d

Morphological, Molecular, and Pathological Appraisal of Hymenolepis nana (Hymenolepididae) Infecting Laboratory Mice (Mus musculus).

Hymenolepis nana, typically a parasite found in conventionally established mouse colonies, has zoonotic potential characterized by autoinfection and direct life cycle. The objective of this study was to determine the rate of parasite infection in laboratory mice. The hymenolepidide cestode infected 40% of the 50 mice sampled. The rate of infection in males (52%) was higher than in females (28%). Morphological studies on the cestode parasite showed that worms had a globular scolex with four suckers, a retractable rostellum with 20-30 hooks, and a short unsegmented neck. In addition, the remaini

Circulation of Tick-Borne Spirochetes in Tick and Small Mammal Communities in Santa Barbara County, California, USA.

A diversity of Borrelia burgdorferi sensu lato (Johnson, Schmid, Hyde, Steigerwalt & Brenner) (Spirochaetales: Spirochaetaceae) genomospecies, including the Lyme disease agent, Borrelia burgdorferi sensu stricto (s.s.), have been identified in the western United States. However, enzootic transmission of B. burgdorferi s.l. in small mammals and ticks is poorly characterized throughout much of the region. Here we report prevalence of B. burgdorferi s.l. in small mammal and tick communities in the understudied region of southern California. We found B. burgdorferi s.l. in 1.5% of Ixodes species t

Yersinia pestis: the Natural History of Plague.

The Gram-negative bacterium Yersinia pestis is responsible for deadly plague, a zoonotic disease established in stable foci in the Americas, Africa, and Eurasia. Its persistence in the environment relies on the subtle balance between Y. pestis-contaminated soils, burrowing and nonburrowing mammals exhibiting variable degrees of plague susceptibility, and their associated fleas. Transmission from one host to another relies mainly on infected flea bites, inducing typical painful, enlarged lymph nodes referred to as buboes, followed by septicemic dissemination of the pathogen. In contrast, drople

Disseminated Mycobacterium ulcerans Infection in Wild Grasscutters (Thryonomys swinderianus), Côte d'Ivoire.

Buruli ulcer is an infectious disease provoking chronic, disabling skin ulcers in mammals and humans. Buruli ulcer is caused by Mycobacterium ulcerans, an environmental mycobacterium synthesizing a toxin called mycolactone responsible for the pathogenicity. The reservoirs and the modes of transmission of M. ulcerans remain elusive, limiting the prophylaxis capabilities in rural areas in endemic countries. In Australia, several studies have demonstrated the probable role of possums as reservoirs. In Côte d'Ivoire, some studies have speculated on the potential role of grasscutters in the transmi

Immobilization of American Beaver (Castor canadensis) with Nalbuphine, Medetomidine, and Azaperone.

Thirty-two American beavers (Castor canadensis) were immobilized with a mixture of nalbuphine, medetomidine, and azaperone (NalMedA) for tail transmitter placement and health assessments prior to translocation. Inductions and reversals were very smooth, but regardless of the dose administered, which ranged from 0.02 to 0.06 mL/kg, many beavers reacted to mild stimuli such as being lifted out of the cage, drawing blood from the tail, expressing anal glands for sex determination, and turning on isoflurane to deepen anesthesia before placement of tail transmitters. On a scale from 1 to 5, a sedat

Diversity of Toxoplasma gondii strains shaped by commensal communities of small mammals.

Commensal rodent species are key reservoirs for Toxoplasma gondii in the domestic environment. In rodents, different T. gondii strains show variable patterns of virulence according to host species. Toxoplasma gondii strains causing non-lethal chronic infections in local hosts will be more likely to persist in a given environment, but few studies have addressed the possible role of these interactions in shaping the T. gondii population structure. In addition, the absence of validated techniques for upstream detection of T. gondii chronic infection in wild rodents hinders exploration of this iss

The Role of Mammalian Reservoir Hosts in Tick-Borne Flavivirus Biology.

Small-to-medium sized mammals and large animals are lucrative sources of blood meals for ixodid ticks that transmit life-threatening tick-borne flaviviruses (TBFVs). TBFVs have been isolated from various organs obtained from wild-caught Myodes and Apodemus species in Europe and Asia. Thus, these rodents are well-established reservoirs of TBFVs. Wild-caught Peromyscus species have demonstrated seropositivity against Powassan virus, the only TBFV known to circulate in North America, suggesting that they may play an important role in the biology of the virus in this geographic region. However, vi

High-resolution melting PCR assay, applicable for diagnostics and screening studies, allowing detection and differentiation of several Babesia spp. infecting humans and animals.

The goal of the study was to design a single tube PCR test for detection and differentiation of Babesia species in DNA samples obtained from diverse biological materials. A multiplex, single tube PCR test was designed for amplification of approximately 400 bp region of the Babesia 18S rRNA gene. Universal primers were designed to match DNA of multiple Babesia spp. and to have low levels of similarity to DNA sequences of other intracellular protozoa and Babesia hosts. The PCR products amplified from Babesia DNA isolated from human, dog, rodent, deer, and tick samples were subjected to high-reso

Concerns about the widespread use of rodent models for human risk assessments of endocrine disruptors.

Fetal testis is a major target of endocrine disruptors (EDs). During the last 20 years, we have developed an organotypic culture system that maintains the function of the different fetal testis cell types and have used this approach as a toxicological test to evaluate the effects of various compounds on gametogenesis and steroidogenesis in rat, mouse and human testes. We named this test rat, mouse and human fetal testis assay. With this approach, we compared the effects of six potential EDs ((mono-(2-ethylhexyl) phthalate (MEHP), cadmium, depleted uranium, diethylstilboestrol (DES), bisphenol

Patterns of covariation in the masticatory apparatus of hystricognathous rodents: implications for evolution and diversification.

The mammalian masticatory apparatus is a highly plastic region of the skull. In this study, a quantification of shape variation, the separation of phylogeny from ecology in the genesis of shape brings new insights on the relationships between morphological changes in the cranium, mandible, and muscle architecture. Our study focuses on the Ctenohystrica, a clade that is remarkably diverse and exemplifies a rich evolutionary history in the Old and New World. Current and past rodent diversity brings out the limitations of the qualitative descriptive approach and highlights the need for using inte

Seasonal abundance and epidemiological indices of potential plague vectors Dinopsyllus lypusus (Siphonaptera: Hystrichopsyllidae) and Ctenophthalmus calceatus (Siphonaptera: Ctenophthalmidae) on rodents captured from three habitat types of Hatcliffe and Dzivarasekwa suburbs of Harare, Zimbabwe.

The seasonal abundance of Dinopsyllus lypusus Jordan and Rothschild and Ctenophthalmus calceatus Waterson (potential vectors of plague in southern Africa) were studied on rodent hosts captured in selected habitat types of two periurban suburbs of Harare, Zimbabwe. Removal trapping was used to capture the rodents, from which fleas were collected and identified. Prevalence (proportion of animals infested) and specific flea index (SFI = number of fleas per animal) were calculated for each species of rodent host. Cohabitation of the two flea species on the host and its implications were also asses
