Metabolic reprogramming of tomato roots during rhizobacteria-mediated defense against Erwinia persicina: modulation by gold nanoparticle conjugation.

Rhizobacteria-induced systemic resistance (ISR) is an established strategy for enhancing plant tolerance to biotic stress, yet its metabolic consequences under nanoparticle-assisted delivery remain poorly understood. Here, we investigated metabolic reprogramming in tomato roots (Solanum lycopersicum L.) challenged with the pathogen Erwinia persicina following treatment with PGPR strain Stenotrophomonas rhizophila (Sr) applied either alone or conjugated to phycosynthesized gold nanoparticles using Caulerpa sertularioides. Bionanogold synthesis was confirmed by UV-Visible surface plasmon resonan…

Encapsulation strategies to enhance the stability and bioaccessibility of bioactive components from tomato by-product extracts for nutraceutical applications.

Tomato processing generates large volumes of by-products that remain underexploited despite their high content of bioactive compounds with potential health benefits. In this study, the extraction of tomato by-products to produce an extract with neuroprotective-related bioactivity is targeted. Moreover, three encapsulation approaches aimed at increasing the stability of their bioactive compounds were evaluated and compared, namely, spray-drying (SDE), non-modified lecithin liposomes (NLE), and inulin-modified lecithin liposomes (ILE). Encapsulation efficiency, stability under accelerated condit

AaMYB17 Regulates the Biosynthesis of Anthocyanins in the Fruit of Actinidia arguta.

To reveal the relationship of anthocyanin biosynthesis in Actinidia arguta, we cloned a typical R2R3-MYB transcription factor-AaMYB17 gene from the cultivar "Dahongpao." Expression analysis showed that AaMYB17 was significantly negatively correlated with anthocyanin accumulation and the expression of most of the structural genes in the anthocyanin biosynthetic pathway. Functional verification using transient overexpression, virus-induced gene silencing (VIGS), and heterologous expression in Arabidopsis thaliana and tomato demonstrated that AaMYB17 acts as a broad-spectrum repressor that signif

Integrative multi-omics analysis uncovers the critical role of plant-derived ferulic acid influencing the occurrence of tomato Fusarium foot rot disease.

Fusarium foot rot, caused by Fusarium solani (Mart.) Sacc., is a major threat to tomato production resulting in a tremendous loss of yield. The interaction between tomato and F. solani has not been well studied. Here we apply an integrative approach with physiological, transcriptomic, and metabolomic analysis to investigate the molecular basis of tomato defense responses to F. solani infection. Pathogen-infected plants have reduced leaf chlorophyll fluorescence, plant elongation, stem thickness and root growth compared to non-infected control. Transcriptome analysis reveals 5654 differentially

Effects of a slowly fermentable fiber mixture against the background of a high-protein diet on insulin sensitivity and metabolic health in individuals with overweight: a randomized, placebo-controlled trial.

The gut microbiota ferments dietary fibers, producing short-chain fatty acids (SCFA). Enhanced SCFA production in the distal colon has been linked to improved cardiometabolic health. However, most fibers are fermented proximally, resulting in increased protein fermentation distally, producing metabolites putatively harmful to metabolic health. This 12-week randomized, placebo-controlled trial aimed to improve metabolic health through increasing distal SCFA production while inhibiting proteolytic fermentation using a fiber supplement that increased distal SCFA production in vitro. We assessed t

Pleiotropic Phenotypes of the Tomato diageotropica Mutant Enable Resistance to Ralstonia solanacearum.

Quantitative disease resistance (QDR) is the most common form of disease resistance in crops, but it is challenging to understand at the cellular level due to the involvement of many genes and biological processes. Ralstonia solanacearum, the causal agent of bacterial wilt disease, is a destructive plant pathogen of Solanaceous species that is best controlled by quantitatively resistant varieties, but few QDR genes are known. We previously found that a tomato auxin pathway mutant known as diageotropica (dgt) has enhanced resistance to R. solanacearum. Here, we show that, as with wild-type quan

SlDCD and SlLCD increased the salt tolerance in tomato seedlings by enhancing antioxidant and photosynthesis capacity.

Using gene silence and heterologously overexpression, hydrogen sulfide synthesis-related genes l-cysteine desulfhydrase and d-cysteine desulfhydrase have been shown to enhance salt tolerance in tomato seedlings. Hydrogen sulfide (H2S) plays an important role in alleviating abiotic stress. L-Cysteine desulfhydrase (LCD) and D-cysteine desulfhydrase (DCD) are two important H2S synthesis enzymes. Until now, whether and how SlDCD and SlLCD increase salt tolerance in plant are still unknown. Here, we explored the effects of SlDCD and SlLCD on salt tolerance in tomato seedlings by silencing SlDCD an

Genome-wide characterization of nitric oxide-induced NBS-LRR genes from Arabidopsis thaliana and their association in monocots and dicots.

BACKGROUND: Nitric oxide (NO) is pivotal in regulating the activity of NBS-LRR specific R genes, crucial components of the plant's immune system. It is noteworthy that previous research has not included a genome-wide analysis of NO-responsive NBS-LRR genes in plants. RESULTS: The current study examined 29 NO-induced NBS-LRR genes from Arabidopsis thaliana, along with two monocots (rice and maize) and two dicots (soybean and tomato) using genome-wide analysis tools. These NBS-LRR genes were subjected to comprehensive characterization, including analysis of their physio-chemical properties, phy

Expression of a Functional Recombinant Human Glycogen Debranching Enzyme (hGDE) in N. benthamiana Plants and in Hairy Root Cultures.

BACKGROUND: Glycogen storage disease type III (GSDIII, Cori/Forbes disease) is a metabolic disorder due to the deficiency of the Glycogen Debranching Enzyme (GDE), a large monomeric protein (about 176 kDa) with two distinct enzymatic activities: 4-α-glucantransferase and amylo-α-1,6-glucosidase. Several mutations along the amylo-alpha-1,6-glucosidase,4-alphaglucanotransferase (Agl) gene are associated with loss of enzymatic activity. The unique treatment for GSDIII, at the moment, is based on diet. The potential of plants to manufacture exogenous engineered compounds for pharmaceutical purpose

Anti-hypertrophic and anti-apoptotic effects of short peptides of potato protein hydrolysate against hyperglycemic condition in cardiomyoblast cells.

Cardiomyocyte hypertrophy is a critical pathological phenomenon observed in diabetic cardiomyopathy. Various molecular events including the Calcineurin/nuclear factor of activated T-cell (NFAT) mediated signaling contributes to the pathogenesis of cardiac hypertrophy. While different new therapeutic interventions are investigated in order to overcome pathological hypertrophic effects, recent studies on peptide hydrolysates from common foods have gained interest. In this study the cytoprotective efficiency of two short peptides DIKTNKPVIF (DF) and a dipeptide IF from a potato protein hydrolysat

Modulation of Enhanced Antioxidant Activity by Hydrogen Sulfide Antagonization of Ethylene in Tomato Fruit Ripening.

Ethylene (C2H4) and hydrogen sulfide (H2S) play important physiological roles in regulating fruit ripening and senescence. The mechanism of H2S in ethylene-induced tomato fruit ripening and senescence is still unknown. Here, we show that exogenous H2S reduced the production of superoxide anion (·O2-), malondialdehyde (MDA), and H2O2 in tomato fruit. Further, additional H2S was found to induce the activities of guaiacol peroxidase, catalase, ascorbate peroxidase, and superoxide dismutase compared with C2H4 treatment alone, whereas the activities of lipoxygenase, polyphenol oxidase, and phenylal

Susceptibility of Salmonella enterica Isolates from Tomato Farm Environments to Fatty Acids Naturally Found on Tomato Fruit.

Salmonella enterica subsp. enterica can colonize tomato fruit as it interacts with fruit surface compounds. The exometabolome of tomato fruit contains a mixture of compounds, including fatty acids, which could affect Salmonella fitness. Fatty acids detected in fruit exudates were investigated for Salmonella inhibition. Pelargonic, lauric, myristic, palmitic, margaric, stearic, and oleic acids were suspended in water dissolved in dimethyl sulfoxide (DMSO) or emulsified in water and quillaja saponin to assess how bioavailability impacted Salmonella growth. The minimum inhibitory concentrations o
