The involvement of transmembrane transport of Ca2+ in regulation of retinal and pineal serotonin N-acetyltransferase (NAT) activity was studied in vivo in hens. Intramuscular administration to hens of organic antagonists of voltage-sensitive calcium channels (VSCC), nimodipine, nifedipine and verapamil, resulted in a marked inhibition of the nocturnal increase of NAT activity in retina and pineal gland. In both tissues the inhibitory effect of nimodipine on the night-time rise of NAT activity was abolished by Bay K 8644, a VSCC agonist. Bay K 8644 administered to hens at the end of the light p…

Exogenous arachidonic acid does not stimulate insulin release in Ca++-containing medium, but a potent effect was unmasked by extracellular Ca++ depletion. This secretion met several criteria of exocytotic release. It did not require the oxygenation of arachidonate or its esterification into islet membranes, but was potentiated by the presence of 16.7 mM glucose such that 33 microM arachidonate could reverse the inhibitory effects of extracellular Ca++ removal on glucose-induced insulin secretion. Arachidonic acid alone stimulated a rise in intracellular Ca++ concentrations in dispersed islet c

Calcium pyrophosphate deposition (CPPD) disease is a common crystalline arthritis that has historically been under-studied compared to other rheumatic diseases including gout. For decades, the presence of CPPD in joints has been consider of uncertain clinical importance, with conflicting reports on whether CPPD leads to osteoarthritis progression or other adverse outcomes. A growing body of literature in the past five years has challenged the prevailing view of CPPD disease as "just chondrocalcinosis." These findings have arrived together with recent developments in classifying CPPD disease fo…

Alginate hydrogels are widely used for encapsulation; however, their inherent porosity can limit protection of probiotic cells under harsh gastrointestinal conditions. Here, glucomannan-rich salep was incorporated into Ca-alginate beads to modulate network interactions and evaluate their influence on probiotic performance. Beads containing 2% (w/v) total polymer (alginate; alginate/salep 1.5/0.5 and 1.0/1.0, w/v) were produced by extrusion into CaCl2. Lactobacillus acidophilus LA-5 was encapsulated and the beads were characterized in terms of diameter, mechanical resistance, molecular interact

Although calcium homeostasis is disrupted in metabolic diseases, its metabolic regulation remains unclear. Here, we identify a mechanism by which fumarate suppresses sarco/endoplasmic reticulum (ER) calcium ion-adenosine triphosphatase (SERCA) activity via succination of a conserved cysteine residue, impairing ER calcium uptake and promoting metabolic dysfunction in Drosophila. In mammalian cells, high glucose or fumarate inhibits SERCA activity and increases ER calcium release and cytosolic and mitochondrial calcium levels. Mechanistically, we show that fumarate covalently modifies SERCA2b at…

RBFOX1 is an RNA-binding protein that regulates alternative splicing and RNA processing in the neurons, skeletal muscle, and heart. We intended to define the role of RBFOX1 in regulating calcium homeostasis to maintain normal cardiac function. We generated cardiomyocyte-specific Rbfox1 gene-deletion mice (cKO). The cardiomyocyte-specific deletion of RBFOX1 was confirmed by Western blotting and immunohistochemistry. The cKO mice showed mild hypertrophy and depressed cardiac function under homeostatic conditions, which did not deteriorate with age. Pressure overload by trans-aortic constriction …

Pyroptosis is a promising strategy for tumor immunotherapy, but nanomaterial-based pyroptosis inducers remain underexplored. Here, we developed a pH-responsive calcium nanomodulator by loading curcumin onto ultrathin Ca/Al layered double hydroxide nanosheets (LDH-CUR). LDH-CUR induces pyroptosis via mitochondrial calcium overload. Upon internalization by tumor cells, it degrades in acidic environments, releasing calcium ions while curcumin blocks calcium efflux, leading to calcium surges. This triggers reactive oxygen species production, cytochrome C release, and caspase-3 activation. Caspase-…

Electrochemical pH-swing systems have demonstrated significant potential across diverse applications, including chemical production, carbon capture, and water treatment. However, conventional systems predominantly depend on costly ion exchange membranes, which are often plagued by fouling and scaling challenges. Here, we introduce an ingenious electrochemically assisted calcium silicate (EACS) system capable of achieving a rapid pH swing from 8.5 to 10 within 1 h through the in situ utilization of H+ and OH- ions, eliminating the need for membranes. The EACS system incorporates a novel 3D-prin…

Extracellular calcium is crucial for the normal function of muscle spindle sensory afferents. They express multiple calcium buffering proteins. Extracellular calcium is essential for recycling of synaptic-like vesicles (SLVs) in the terminals and for the stretch-evoked inward calcium current of the receptor potential. Conversely, removal of calcium from the extracellular medium abolishes stretch-evoked action potentials (APs). However, the calcium channel(s) involved and mechanism(s) of action are unknown. This study begins identifying the channels involved and their actions. Specific calcium …

Live-cell Ca2+ imaging is an important tool to detect activation of receptors by a putative ligand/drug and complements studies on transport processes, as intracellular Ca2+ changes provide direct evidence for substrate fluxes. Organoid-based systems offer numerous advantages over other in vitro systems such as cell lines, primary cells, or tissue explants, and in particular, intestinal organoid culture has revolutionized research on functional gastrointestinal processes. Calcium imaging using the fluorescent Ca2+ indicator Fura-2-AM can be applied to 3D intestinal organoids, which show an exc…

Calcium (Ca2+) is an essential regulator of cardiac function. Particularly, Ca2+ is the primary link between the electrical signals regulating contractility of the myocytes and thus allowing the heart chambers to relax and refill completely with blood. Additionally, Ca2+ controls numerous other activities including gene transcription, cell growth, and survival. The abnormal Ca2+ regulation and cycling in the cardiomyocyte following various stressor's, insults and during cardiac disease development has been found to be a primary culprit leading to cellular dysfunction and potentially to cell de…

Glaucoma is one of the most common causes of treatable visual impairment in the developed world, affecting approximately 64 million people worldwide, some of whom will be bilaterally blind from irreversible optic nerve damage. The optic nerve head is a key site of damage in glaucoma where there is fibrosis of the connective tissue in the lamina cribrosa (LC) extracellular matrix. As a ubiquitous second messenger, calcium (Ca2+) can interact with various cellular proteins to regulate multiple physiological processes and contribute to a wide range of diseases, including cancer, fibrosis, and gla…

Select Aspergillus species can produce oxalate as a fermentation byproduct, which may react with calcium ions to produce insoluble calcium oxalate crystals in tissues. These crystals are frequently associated with pulmonary Aspergillus infections, yet are rarely described in primary cutaneous aspergillosis. Herein, we report the presence of calcium oxalate crystals detected on cutaneous specimens from primary cutaneous Aspergillus niger and Aspergillus fumigatus infections in an immunocompromised, premature infant. No metabolic sources of oxalosis were found.

Calcium (Ca2+) is involved as a signalling mediator in a broad variety of physiological processes. Some of the fastest responses in human body like neuronal action potential firing, to the slowest gene transcriptional regulation processes are controlled by pathways involving calcium signalling. Under pathological conditions these mechanisms are also involved in tumoral cells reprogramming, resulting in the altered expression of genes associated with cell proliferation, metastatisation and homing to the secondary metastatic site. On the other hand, calcium exerts a central function in nocicepti…
