BACKGROUND: Viroporins are virally encoded ion channels involved in virus assembly and release. Human immunodeficiency virus type 1 (HIV-1) and influenza A virus encode for viroporins. The human coronavirus SARS-CoV-2 encodes for at least two viroporins, a small 75 amino acid transmembrane protein known as the envelope (E) protein and a larger 275 amino acid protein known as Orf3a. Here, we compared the replication of HIV-1 in the presence of four different β-coronavirus E proteins.
RESULTS: We observed that the SARS-CoV-2 and SARS-CoV E proteins reduced the release of infectious HIV-1 yields

OBJECTIVES: An understanding of the predictors of comorbidity among people living with HIV (PLWH) is critical for effective HIV care management. In this study, we identified predictors of comorbidity burden among PLWH based on machine learning models with electronic health record (EHR) data.
METHODS: The study population are individuals with a HIV diagnosis between January 2005 and December 2016 in South Carolina (SC). The change of comorbidity burden, represented by the Charlson Comorbidity Index (CCI) score, was measured by the score difference between pre- and post-HIV diagnosis, and dicho

Paediatric Acquired ImmunoDeficiency Syndrome (AIDS) is a life-threatening and infectious disease in which the Human Immunodeficiency Virus (HIV) is mainly transmitted through Mother-To- Child Transmission (MTCT) during pregnancy, labour and delivery, or breastfeeding. This review provides an overview of the distinct therapeutic alternatives to abolish the systemic viral replication in paediatric HIV-1 infection. Numerous classes of antiretroviral agents have emerged as therapeutic tools for downregulation of different steps in the HIV replication process. These classes encompass Non- Nucleosi

BACKGROUND: The C-C chemokine receptor Type 5 (CCR5) is a key receptor for human immunodeficiency virus type 1 (HIV-1) entry into T-cells and a variant allele, CCR5 delta-32, is associated with decreased viral replication and disease progression. Active HIV-1 replication is highly associated with accelerated rates of hepatic fibrosis. We postulated that CCR5 plays a role in the development of hepatic fibrosis and evaluated the longitudinal effect of natural or drug-induced CCR5 mutation and blockade on biomarkers of liver fibrosis in HIV-1 patients.
METHODS: To accomplish this goal, we examin

Regulatory T (Treg) cells play a key role in dampening excessive immune activation. However, antiretroviral therapy (ART) -naive HIV-1 infection maintains the immune system in a sustained state of activation that could alter both Treg cell surface markers and functions. As Treg cell surface markers are directly linked to their functions the overall objective of this study was to determine how ART-naive HIV infection affects the phenotypic properties of Treg cells. Our data showed that in ART-naive HIV-1 infection, Treg cells are dominated by effector (CD45RA+ CD27- CCR7- CD62L- ) and effecto

BACKGROUND: Women living with HIV/AIDS who drink alcohol are at increased risk for adverse health outcomes, but there is little evidence on best methods for reducing alcohol consumption in this population. We conducted a pilot study to determine the acceptability and feasibility of conducting a larger randomized clinical trial of naltrexone vs. placebo to reduce alcohol consumption in women living with HIV/AIDS.
METHODS: We designed the trial with input from community and scientific review. Women with HIV who reported current hazardous drinking (>7 drinks/week or ≥4 drinks per occasion) were

OBJECTIVE: HIV-1 subtype B infections are associated with MSM in the UK. Yet, around 13% of subtype B infections are found in those reporting heterosexual contact as transmission route. Using phylogenetics, we explored possible misclassification of sexual exposure among men diagnosed with HIV in the UK.
DESIGN: Viral gene sequences linked to patient-derived information were used to identify phylogenetic transmission chains.
METHODS: A total of 22,481 HIV-1 subtype B pol gene sequences sampled between 1996 and 2008 were analysed. Dated phylogenies were reconstructed and transmission clusters

BACKGROUND: South Africa recommends universal syphilis and HIV testing in pregnancy, with prompt antiretroviral therapy or penicillin treatment for women testing positive.
METHODS: We used a multistage, purposeful sampling strategy to retrospectively identify clinical records from a sample (7.3%) of 32,518 women delivering from January 2005 to June 2006 at 6 public clinics in the Northern Cape and Gauteng. Descriptive analyses and logistic regression were used to assess coverage and factors related to testing and treatment of HIV and syphilis.
RESULTS: Of 2379 women sampled, 93% accessed ant

Chlamydia trachomatis infection is one of the most prevalent bacterial STIs in the USA and worldwide, and women with C. trachomatis infection are at increased risk of acquiring HIV. Because immune activation at the genital mucosa facilitates HIV/SIV infection, C. trachomatis-mediated cytokine induction may contribute to increased HIV transmission in asymptomatic women. To begin to elucidate the mechanisms, we longitudinally analyzed profiles of innate immune factors and HIV infectivity in genital secretions from anatomically specific sites in asymptomatic women during C. trachomatis infection

BACKGROUND AND OBJECTIVES: In October 2005, individual donation nucleic acid amplification testing (ID-NAT) for HIV, HBV and HCV was introduced in the Western Cape Province of South Africa. After 5 years, the impact on HIV, HBV and HCV transmission risk was assessed.
MATERIALS AND METHODS: A total of 649745 donations were tested by ID-NAT using the Ultrio assay on the Tigris instrument (Novartis Diagnostics) and for anti-HIV, HBsAg and anti-HCV (Abbott Prism). Initial reactive samples were repeated in duplicate. Discrepant repeat reactive samples were subjected to confirmatory assays. ID-NAT

OBJECTIVE: Testing the hypothesis of an association between knowledge and sexual risk behaviour (SRB) amongst community-clinic workers in Chile, explained by the confounding effect of self-perceived vulnerability to HIV.
METHODS: A cross-sectional survey was analyzed; it was nested within a quasi-experimental study of 720 community-clinic workers in Santiago. The SRB score combined the number of sexual partners and condom use (coded as "high"/"low" SRB). Knowledge of HIV (a 25-item index) was coded as "inadequate"/"adequate" knowledge. Self-perceived vulnerability to HIV was categorised as be

OBJECTIVE: This exploratory study quantified potential barriers to repeat HIV testing, as well as perceptions supporting further testing among women previously tested for HIV. The study also determined associations between summative measures of barriers (and supportive factors) and women's intent to be tested for HIV.
DESIGN AND SETTING: One hundred forty-three women (95.4% African-American) who attended an urgent care center, and reported a history of HIV testing, completed a face-to-face interview.
RESULTS: Frequently perceived barriers included perceptions that repeat testing was unnecess

To evaluate care provided to women living with HIV in São Paulo, Brazil, based on their own experiences, patients from HIV/AIDS reference clinics answered a questionnaire on circumstances of HIV testing, health personnel's attitudes at diagnosis, adherence to follow-up, services provided by care centers and access to laboratory monitoring. From September 1999 to February 2000, 1,068 women were interviewed. Most had over 8 years of education and reported having regular sexual partners, being mothers and family caregivers. They were diagnosed as HIV-infected for 4.36 +/- 3.15 years. The majority

Women represent an increasing proportion of AIDS cases and anecdotal reports suggest some face substantial risks when others learn they are HIV-positive. The purpose of this paper is to describe women's fears and experiences regarding disclosure of their HIV status. Fifty HIV-positive women, ages 16-45 from urban teaching hospital outpatient clinics, were interviewed using an in-depth, qualitative interview. Eighty-six percent of the women were African American and 56% were current or former IVDU. At the time of the interview, 88% of the women had known their HIV status for a year or more. All

OBJECTIVE: To report a case of an HIV elite controller (EC) who developed disease progression following SARS-CoV-2 infection, characterized by the breakdown of prior immune control.
CASE PRESENTATION: We report a case of a Chinese male EC with HIV-1 CRF01_AE and protective HLA-B*57:01, who had sustained undetectable viral load and stable CD4+ T-cell counts for over 10 years. Following SARS-CoV-2 infection, he developed persistent HIV viral rebound (peaking at 4430 copies/ml) for eight months, accompanied by a profound CD4+ T-cell decline to 174 cells/µl, and subsequently initiated antiretrovi

Men who have sex with men (MSM) account for nearly 70% of new HIV infections annually. Although awareness of pre-exposure prophylaxis (PrEP) is growing, uptake remains low among young MSM. Peer navigators are well positioned to guide patients through PrEP care, provide psychosocial support, build trust, and demystify PrEP. Given strong evidence to support mobile health (mHealth) apps for enhancing HIV prevention knowledge, our team created MyPEEPS Plus, a HIV prevention combination intervention of mHealth and an electronic peer (e-peer) navigation program. In this article, we describe developm

Background: Previous studies have demonstrated the efficacy of membrane-anchored C-peptides against HIV and have shown that the surface expression level of these peptides is critical for effective protective activity. Methods: Bicistronic genetic constructs were designed for GPI-anchored expression of protective C-peptides on the cell membrane. The constructs were delivered using lentiviral vectors and tested in primary CD4+ lymphocytes and hematopoietic stem cells. Results: The bicistronic configuration significantly enhanced surface expression levels compared to monocistronic constructs and

The transformation of HIV into a manageable chronic condition has unveiled new clinical challenges associated with aging-related pathologies, including bone disease. This review explores the intricate relationship between HIV, antiretroviral therapy (ART), and bone disease, highlighting the necessity of early diagnosis and preventative strategies to mitigate the increased risk of osteopenia, osteoporosis, and fractures in people living with HIV (PLWHIV). It synthesizes the current literature to elucidate the multifactorial etiology of bone pathology in this population, that includes direct vir
