HIF and MYC signaling in adrenal neoplasms of the neural crest: implications for pediatrics.

Pediatric neural crest-derived adrenal neoplasms include neuroblastoma and pheochromocytoma. Both entities are associated with a high degree of clinical heterogeneity, varying from spontaneous regression to malignant disease with poor outcome. Increased expression and stabilization of HIF2α appears to contribute to a more aggressive and undifferentiated phenotype in both adrenal neoplasms, whereas MYCN amplification is a valuable prognostic marker in neuroblastoma. The present review focuses on HIF- and MYC signaling in both neoplasms and discusses the interaction of associated pathways during

Central 5-HTR2C in the Control of Metabolic Homeostasis.

The 5-hydroxytryptamine 2C receptor (5-HTR2C) is a class G protein-coupled receptor (GPCR) enriched in the hypothalamus and the brain stem, where it has been shown to regulate energy homeostasis, including feeding and glucose metabolism. Accordingly, 5-HTR2C has been the target of several anti-obesity drugs, though the associated side effects greatly curbed their clinical applications. Dissecting the specific neural circuits of 5-HTR2C-expressing neurons and the detailed molecular pathways of 5-HTR2C signaling in metabolic regulation will help to develop better therapeutic strategies towards m

Strategies to Promote Long-Distance Optic Nerve Regeneration.

Mammalian retinal ganglion cells (RGCs) in the central nervous system (CNS) often die after optic nerve injury and surviving RGCs fail to regenerate their axons, eventually resulting in irreversible vision loss. Manipulation of a diverse group of genes can significantly boost optic nerve regeneration of mature RGCs by reactivating developmental-like growth programs or suppressing growth inhibitory pathways. By injury of the vision pathway near their brain targets, a few studies have shown that regenerated RGC axons could form functional synapses with targeted neurons but exhibited poor neural

Inactivation of the parietal reach region causes optic ataxia, impairing reaches but not saccades.

Lesions in human posterior parietal cortex can cause optic ataxia (OA), in which reaches but not saccades to visual objects are impaired, suggesting separate visuomotor pathways for the two effectors. In monkeys, one potentially crucial area for reach control is the parietal reach region (PRR), in which neurons respond preferentially during reach planning as compared to saccade planning. However, direct causal evidence linking the monkey PRR to the deficits observed in OA is missing. We thus inactivated part of the macaque PRR, in the medial wall of the intraparietal sulcus, and produced the h

Neuronal encoding of sound, gravity, and wind in the fruit fly.

The fruit fly Drosophila melanogaster responds behaviorally to sound, gravity, and wind. Exposure to male courtship songs results in reduced … sensed by these two neuronal groups travel in parallel from the fly ear to the brain, feeding into neural pathways reminiscent of the auditory and vestibular pathways in the human brain. Studies of the similarities between mammals and flies will lead to a better understanding of the principles of how sound and gravity information is encoded in the brain. Here, w …

Cornelia de Lange Syndrome: NIPBL haploinsufficiency downregulates canonical Wnt pathway in zebrafish embryos and patients fibroblasts.

Cornelia de Lange Syndrome is a severe genetic disorder characterized by malformations affecting multiple systems, with a common feature of severe mental retardation. Genetic variants within four genes (NIPBL (Nipped-B-like), SMC1A, SMC3, and HDAC8) are believed to be responsible for the majority of cases; all these genes encode proteins that are part of the 'cohesin complex'. Cohesins exhibit two temporally separated major roles in cells: one controlling the cell cycle and the other involved in regulating the gene expression. The present study focuses on the role of the zebrafish nipblb paral

Mitochondrial dysfunction in animal models of PTSD: Relationships between behavioral models, neural regions, and cellular maladaptation.

Post-traumatic stress disorder (PTSD) is a trauma-related condition that produces distressing fear memory intrusions, avoidance behaviors, hyperarousal, stress responses, insomnia and other symptoms. This review of rodent models of PTSD examines trauma effects on fear-related learning, cognition, and avoidance, emotional and arousal behaviors and on mitochondrial dysfunction in relevant neural pathways. The review focuses on research that includes four elements: consensus PTSD rodent models, behavioral phenotyping, mitochondrial dysfunction within key neural regions. This approach allows for t…

Clitoral sexual arousal: neuronal tracing study from the clitoris through the spinal tracts.

PURPOSE: Although genital tactile stimulation is regarded as a precursor to sexual arousal and a recognized initiator of central nervous system arousal, specific afferent neural pathways transmit sensory stimuli of arousal, beginning at the epithelial level on the clitoris and following the course of arousal stimuli through the central nervous system. Limited knowledge exists of the pathway from the cutaneous receptors of nerves originating in the epithelial tissue of the clitoris and continuing to spinal cord afferents. Such information may contribute to an understanding of sexual arousal, pa…

The Nerve-Induced Adipose Stem Cells Promote Nerve Repair in Stress Urinary Incontinence by Regulating Schwann Cell Repair Phenotype Conversion Through Activation of the Notch Pathway.

Stress urinary incontinence (SUI) currently lacks effective treatment options, and the restoration of neurological function remains a major challenge, with unmet clinical needs. Research has indicated that adipose-derived stem cells (ADSCs) can be induced to differentiate into neural-induced adipose-derived stem cells (NI-ADSCs) under specific inductive conditions, exhibiting excellent neuroregenerative capabilities. ADSCs were obtained from female SD rats and induced into NI-ADSCs. In vitro, NI-ADSCs were co-cultured with Schwann cells (SCs) to investigate their effects on SC proliferation an

The Brain-tumor axis: Tumor pathogenesis and therapeutic strategies from a cancer neuroscience perspective.

Cancer research has undergone a fundamental paradigm shift, moving beyond a tumor-cell-centric framework toward a systems-level understanding in which the nervous system occupies a central regulatory role. The brain and its peripheral extensions engage in sophisticated, bidirectional crosstalk with tumors across multiple biological scales. This complex relationship, now termed the brain-tumor axis, operates through mechanisms spanning systemic neuroendocrine regulation, local neural-tumor interactions, and the distinctive biology of the central nervous system as both a primary and metastatic t

Exosomes originating from neural stem cells undergoing necroptosis participate in cellular communication by inducing TSC2 upregulation of recipient cells following spinal cord injury.

JOURNAL/nrgr/04.03/01300535-202511000-00030/figure1/v/2024-12-20T164640Z/r/image-tiff We previously demonstrated that inhibiting neural stem cells necroptosis enhances functional recovery after spinal cord injury. While exosomes are recognized as playing a pivotal role in neural stem cells exocrine function, their precise function in spinal cord injury remains unclear. To investigate the role of exosomes generated following neural stem cells necroptosis after spinal cord injury, we conducted single-cell RNA sequencing and validated that neural stem cells originate from ependymal cells and unde

Targeting the nervous system: Mechanistic insights into neuro-tumor communication and therapeutic opportunities.

The nervous system has emerged as an active regulator of the tumor ecosystem. Neural inputs from the central, peripheral, and enteric nervous systems influence tumor initiation, progression, metastasis, and therapeutic response through innervation and signal transmission. In addition to regulating tumor cell states, neural signaling reshapes stromal and immune compartments, promoting stemness, invasion, angiogenesis, metabolic adaptation, and immune evasion. Tumors, in turn, actively remodel their neural niche through neurogenesis, axonal remodeling, and tumor-nerve feedforward circuits. The b

Neural crest and cardiovascular development: a 20-year perspective.

BACKGROUND: Twenty years ago this year was the first publication describing a region of neural crest cells necessary for normal cardiovascular development. Ablation of this region in chick resulted in persistent truncus arteriosus, mispatterning of the great vessels, outflow malalignments, and hypoplasia or aplasia of the pharyngeal glands. METHODS: We begin with a historical perspective and then review the progress that has been made in the ensuing 20 years in determining the direct and indirect contributions of the neural crest cells, now termed cardiac neural crest cells, in cardiovascular

A working feedback-based framework for goal-directed spatial memory deficits in Alzheimer's disease: Feedforward and feedback interactions along the RSC-MEC-CA1-RSC axis.

Spatial navigation deficits are among the earliest and most clinically significant cognitive impairments in Alzheimer's disease (AD), particularly when navigation depends on goal-directed spatial memory. This review defines goal-directed spatial memory as a task-oriented construct and proposes a network-level framework for interpreting early AD navigation deficits. We synthesize anatomical, physiological, behavioral, and disease-related evidence concerning the retrosplenial cortex (RSC), medial entorhinal cortex (MEC), hippocampal CA1, and CA1-RSC feedback interactions. We propose the RSC-MEC-

Disruption of the claustrum-ACC pathway contributes to human mind blanking.

The claustrum is a highly connected structure hypothesized to orchestrate conscious experience, yet its role in humans remains enigmatic. To address this question, we prospectively investigated patients with drug-resistant epilepsy who underwent stereoelectroencephalography (SEEG) implantation driven by clinical indications, with electrode trajectories optimized to target the claustrum. Across the eight participants, the stimulated claustrum was left-sided in six and right-sided in two. Focal stimulation of the left claustrum reproducibly induced a transient arrest of ongoing thought and a red

A touch-guided neural circuit regulates motivated gnawing to maintain dental alignment.

How hindbrain circuits integrate peripheral and central signals to regulate complex oral behaviors is poorly understood. In rodents, gnawing is essential for localized tooth wear to offset lifelong incisor growth. Whether this process relies on specific sensory input to guide localized tooth wear and is actively regulated by neural mechanisms remains unresolved. Here, we identify somatostatin-expressing neurons in the spinal trigeminal nucleus oralis as a central relay distributing tactile input to motor execution and motivational circuits. These neurons receive input from a genetically distin

Distinct Patterns of Directed Brain Connectivity in Focused Attention, Open Monitoring, and Loving Kindness Meditation: An Electroencephalographic Granger Causality Study with Long-term Meditators.

The present study applied spectral Granger causality (GC) analysis to EEG recordings obtained during focused attention meditation (FAM), open monitoring meditation (OMM), and loving kindness meditation (LKM) in highly experienced meditators. The aim of the investigation was to characterize distinct connectivity signatures associated with each meditation style by examining the strength, frequency, and direction of interregional interactions. These differences were expected to specify the neural connectivity profiles of the cognitive and affective state of each meditative practice. Multivariate

Differential Functional Connectivity Between Hippocampus and Prefrontal Cortex Is Associated With Heterogeneity in Open Field Exploration and Is Disrupted Following Maternal Immune Activation.

A common paradigm in behavioral neuroscience involves recording neural activity from freely moving rodents as they forage in an open field. This procedure is often used in studies investigating spatial navigation, where recording is conducted in regions such as the hippocampus and entorhinal cortex. It is usually assumed that there is no systematic variation in behavior in this paradigm, thereby allowing spatial representations to be examined without the confounding effects of behavior change. Here we show that the behavior of rats in this paradigm can be algorithmically divided into at least

Correction of connectivity induced by autocorrelation and filtering in resting state functional near-infrared spectroscopy data.

BACKGROUND: Seed-based correlation is commonly used to assess resting state functional connectivity in functional near-infrared spectroscopy (fNIRS). The sample independence assumption underlying correlation is violated in both unfiltered and filtered fNIRS data, due to the non-white frequency spectra of acquired fNIRS signals. This violation inflates the false positive rate, induces spurious correlations, and consequently alters connectivity results. NEW METHOD: We extend an existing correction framework originally developed for fMRI, to accommodate the coloured spectral properties of fNIRS

A Cortico-Basal Ganglia-Thalamic Network Model Linking Intermittent Postural Control to Sway-Related Beta-Band Oscillations.

Electroencephalographic (EEG) studies of human quiet stance demonstrate beta-band event-related desynchronization (beta-ERD) during the micro-fall phase of postural sway, followed by event-related synchronization (beta-ERS; post-movement beta rebound) during the micro-recovery phase. These modulations correlate with intermittent ankle muscle inactivation that exploits the stable manifolds of an unstable upright equilibrium; however, how such sway-related beta dynamics arise within closed-loop brain-body interactions remains unclear. Here, we investigated a possible circuit-level account of the
