A novel bioencapsulation strategy for delivering plant growth promoting bacteria via cotton seeds to improve crop yield and shelf stability.

Cotton is a vital cash crop in India that is predominantly cultivated in rainfed regions and is increasingly affected by climate change, which leads to poor seed germination and seedling establishment. To address these challenges and support sustainable agriculture, the current investigation aimed to develop and optimize bioencapsulated cotton seeds using plant growth-promoting bacteria (PGPB) to enhance seed germination, bacterial viability, seed shelf life, and crop yield. The bioencapsulation process was optimized using Response Surface Methodology via inverse gelation with sodium alginate,

Development of nimesulide-loaded pH-triggered ocular in situ gels: in vitro characterization, MTT assay, and Drazie test.

OBJECTIVE: The present study created nimesulide-loaded in situ gels and assessed their potential application in ocular inflammation. METHODS: Because in situ gels have a low viscosity, sodium alginate, and hydroxypropyl methylcellulose combined to create a nimesulide CD complex that is easy to apply. However, due to the gelation creation with pH, washing and removing it from the ocular surface becomes challenging. Production was carried out using different ratios of sodium alginate, hydroxypropyl methylcellulose, and hyaluronic acid. RESULTS: Characterization studies led to the determination

Determining the Age of Every Cell Within Each Budding Yeast Microcolony Combining Single-Cell Microencapsulation With Confocal Microscopy.

Isogenic populations of Saccharomyces cerevisiae exhibit significant proliferative heterogeneity, with individual cells within a clonal culture displaying divergent growth rates and metabolic states. Investigating the origins of this variation requires a method to reconstruct the individual histories of cells within the population. This protocol describes a method for single-cell microencapsulation in alginate microspheres to create a physically stable, traceable, three-dimensional genealogical environment. By utilizing the alginate matrix to prevent daughter cell migration, the replicative hi

Comprehensive biocompatibility profiling of human pancreas-derived biomaterial.

INTRODUCTION: The importance of the extracellular matrix (ECM) to pancreatic islets has been clearly demonstrated, as isolated islets grown in culture or transplanted, quickly lose viability and function after their matrix associations have been stripped away during the isolation process. Therefore, recapitulating the islet niche is a critical objective to move the field of islet transplantation forward. METHODS: As a first step to recreating the islet microenvironment, we have recently developed a detergent-free decellularization method to obtain a decellularized solubilized ECM (dsECM) powd

Biomass-derived functional additive for highly efficient and stable lead halide perovskite solar cells with built-in lead immobilisation.

Despite notable progress in the power conversion efficiency (PCE) of lead halide perovskite solar cells (PSCs), their commercial viability remains limited by stability issues and the risk of lead contamination. Uncoordinated lead ions can introduce defects during perovskite crystallization, resulting in reduced stability and potential environmental contamination. Here, we synthesized a biomass-derived tetrabutylammonium alginate (TBA-Alg) polymer that forms a connected network at the perovskite surface and grain boundaries to effectively manage lead ions and passivate defects. The alginate gro

Active Edible Coatings to Mitigate Postharvest Diseases Causing Waste of Blueberries, Strawberries, and Cherry Tomatoes.

Packaging can help prolong the shelf life of perishable agrifoods. In the present investigation, edible coatings were tested to reduce food waste caused by filamentous fungi and increase the shelf-life of high-value products such as strawberries, tomatoes, and blueberries. Different combinations of sodium alginate and calcium chloride, and various immersion times were tested on tomato as a model. The ability to activate edible coatings with food-grade compounds/extracts, such as sodium bicarbonate or Moringa oleifera extract (MLE), was explored. The extract was also tested in vitro against som

Programmable and printable formaldehyde dehydrogenase as an excellent catalyst for biodegradation of formaldehyde.

As an environmental pollutant, formaldehyde can cause serious harm to the human body. Among many degradation methods, formaldehyde dehydrogenase from Pseudomonas putida (PFDH) exhibits broad potential because of its strong catalytic specificity and high degradation efficiency. However, the real application of PFDH in industry is limited by its instability and difficulties in recycling. In this work, the suitable printing conditions for immobilizing PFDH by three-dimensional (3D) printing technology were studied: the concentration of sodium alginate (SA) was 1.635 wt%, the concentration of CaCl

Shape-memory collagen scaffold combined with hyaluronic acid for repairing intervertebral disc.

BACKGROUND: Intervertebral disc degeneration (IVDD) is a common cause of chronic low back pain (LBP) and a socioeconomic burden worldwide. Conservative therapies and surgical treatments provide only symptomatic pain relief without promoting intervertebral disc (IVD) regeneration. Therefore, the clinical demand for disc regenerative therapies for disc repair is high. METHODS: In this study, we used a rat tail nucleotomy model to develop mechanically stable collagen-cryogel and fibrillated collagen with shape-memory for use in minimally invasive surgery for effective treatment of IVDD. The coll

Extraction of Fucoidan from Turbinaria decurrens and the Synthesis of Fucoidan-Coated AgNPs for Anticoagulant Application.

Brown seaweeds usually contain alginate as a major polymer. The second major sulfated polymer in brown seaweeds is fucoidan, which has huge potential in medicinal applications. In this study, the photosynthetic pigments from Turbinaria decurrens were first extracted using chloroform/methanol in the ratio of 1:1 (v/v), followed by fucoidan extraction with yields of 5.58% (crude) and 1.28% (purified fucoidan) from the dry weight of seaweed, whereas alginate was extracted with a yield of 14.7% DW of seaweed. The isolated fucoidan possessing anticoagulation property was identified and characterize

Evidence of In Vivo Existence of Borrelia Biofilm in Borrelial Lymphocytomas.

Lyme borreliosis, caused by the spirochete Borrelia burgdorferi sensu lato, has grown into a major public health problem. We recently identified a novel morphological form of B. burgdorferi, called biofilm, a structure that is well known to be highly resistant to antibiotics. However, there is no evidence of the existence of Borrelia biofilm in vivo; therefore, the main goal of this study was to determine the presence of Borrelia biofilm in infected human skin tissues. Archived skin biopsy tissues from borrelial lymphocytomas (BL) were reexamined for the presence of B. burgdorferi sensu lato u

Peptide-modified Substrate for Modulating Gland Tissue Growth and Morphology In Vitro.

In vitro fabricated biological tissue would be a valuable tool to screen newly synthesized drugs or understand the tissue development process. Several studies have attempted to fabricate biological tissue in vitro. However, controlling the growth and morphology of the fabricated tissue remains a challenge. Therefore, new techniques are required to modulate tissue growth. RGD (arginine-glycine-aspartic acid), which is an integrin-binding domain of fibronectin, has been found to enhance cell adhesion and survival; it has been used to modify substrates for in vitro cell culture studies or used as

Unsaturated guluronate oligosaccharide enhances the antibacterial activities of macrophages.

Alginate from marine seaweeds is receiving continuous attention owing to its wide physiological activities. Herein, we sought to elucidate possible effects of alginate-derived polyguluronate (PG) and unsaturated guluronate oligosaccharide (GOS) on antibacterial activities of macrophages. Our results showed that, in contrast to PG, GOS markedly increased the phagocytosis of IgG-opsonized Escherichia coli and Staphylococcus aureus and further inhibited the survival of intracellular bacteria in macrophages. In line with this, GOS treatment resulted in the enhanced expression of Fcγ receptors on m

The effect of human PBMCs immobilization on their Аβ42 aggregates-dependent proinflammatory state on a cellular model of Alzheimer's disease.

The leading pathological mechanisms of Alzheimer's disease are amyloidosis and inflammation. The presented work was aimed to study the effect of human peripheral blood mononuclear cells (hPBMcs) cells-matrix adhesion on their pro-inflammatory state in vitro. Although direct interaction of Аβ42 to PBMC is not a cellular model of Alzheimer's disease, PBMCs may serve as test cells to detect Аβ42-dependent molecular effects in monitoring disease progression. Peripheral blood mononuclear cells (PBMCs) are used to assess changes in cytokines released in response to diseases or Alzheimer's disease-sp

Preparation and drug release performance of different gelation type polysaccharide/β-lactoglobulin fiber composite gels.

Self-assembled protein fibers have attracted much attention in the fields of medicine and food because of their high aspect ratio, polymorphic structure and strong surface hydrophobicity. In this study, three different gelation types of polysaccharides/β-lactoglobulin fiber (Fblg) composite gels, including ionic alginate-Fblg gels, synergistic xanthan-Fblg gels, and double network agar-Fblg gels, were first prepared. The interactions between the polysaccharides and the Fblgs, the microstructure and mechanical properties of the composite gels were investigated using the light scattering, scanni
