Regulatory protein BBD18 of the lyme disease spirochete: essential role during tick acquisition?

The Lyme disease spirochete Borrelia burgdorferi senses and responds to environmental cues as it transits between the tick vector and vertebrate host. Failure to properly adapt can block transmission of the spirochete and persistence in either vector or host. We previously identified BBD18, a novel plasmid-encoded protein of B. burgdorferi, as a putative repressor of the host-essential factor OspC. In this study, we investigate the in vivo role of BBD18 as a regulatory protein, using an experimental mouse-tick model system that closely resembles the natural infectious cycle of B. burgdorferi.

Integrated diagnostic strategy for Theileria equi in camels: combining real-time PCR and cELISA for molecular and serological characterization in Egypt.

Theileria equi is a globally significant tick-borne pathogen causing equine piroplasmosis, yet its occurrence in dromedary camels (Camelus dromedarius) is an understudied phenomenon. This study provides an integrated molecular and serological characterization of T. equi in dromedary camels in Egypt. A total of 210 blood samples were collected from apparently healthy slaughtered camels in Giza, Sharqia, and Cairo governorates and subjected to an integrated diagnostic approach comprising microscopic examination, conventional PCR (cPCR), species-specific cPCR, real-time PCR (rtPCR), and competiti

Hepatitis C virus infection and its associated factors among prisoners in a Nigerian prison.

BACKGROUND: The prison population is considered at high risk of acquiring infectious diseases due to confined conditions, behavioral factors, injection drug use, unprotected sexual activity, non-professional tattooing and scarification, and needle sharing. Hepatitis C virus (HCV) is a blood borne pathogen mostly transmitted via percutaneous exposure that results in inflammation of the liver. It is one of the public health problem worldwide and is the principal cause of parenterally transmitted non-A, non-B hepatitis. The study was aimed at evaluating the prevalence of HCV among prison inmates

Host associations of mosquitoes at eastern equine encephalitis virus foci in Connecticut, USA.

BACKGROUND: Eastern equine encephalitis virus (EEEV) is a highly pathogenic mosquito-borne arbovirus, with active transmission foci in freshwater hardwood swamps in eastern North America, where enzootic transmission is maintained between the ornithophilic mosquito, Culiseta melanura, and wild passerine birds. The role of other locally abundant mosquito species in virus transmission and their associations with vertebrate hosts as sources of blood meals within these foci are largely unknown but are of importance in clarifying the dynamics of enzootic and epidemic/epizootic transmission. METHODS

Original Research: Exploring the Use of Passive vs. Active Insulin Safety Pen Needle Devices in a Pediatric Population: A Feasibility Study.

BACKGROUND: Insulin pens are the mainstay of insulin delivery in the pediatric population, especially among patients unable to use an insulin pump. Safety pen needle (SPN) devices have been embraced by both nurses and patients because they limit the risks of needlestick injury and exposure to blood-borne pathogens. With the commonly used traditional passive SPN device, however, it can be difficult to observe that the dose has been accurately or fully administered. PURPOSE: The purpose of this study was to determine nurses' perceptions about the feasibility of using an active SPN device (speci…

Enhance fashion classification of mosquito vector species via self-supervised vision transformer.

Vector-borne diseases pose a major worldwide health concern, impacting more than 1 billion people globally. Among various blood-feeding arthropods, mosquitoes stand out as the primary carriers of diseases significant in both medical and veterinary fields. Hence, comprehending their distinct role fulfilled by different mosquito types is crucial for efficiently addressing and enhancing control measures against mosquito-transmitted diseases. The conventional method for identifying mosquito species is laborious and requires significant effort to learn. Classification is subsequently carried out by

Comparative Characterization of the Sindbis Virus Proteome from Mammalian and Invertebrate Hosts Identifies nsP2 as a Component of the Virion and Sorting Nexin 5 as a Significant Host Factor for Alphavirus Replication.

Recent advances in mass spectrometry methods and instrumentation now allow for more accurate identification of proteins in low abundance. This technology was applied to Sindbis virus, the prototypical alphavirus, to investigate the viral proteome. To determine if host proteins are specifically packaged into alphavirus virions, Sindbis virus (SINV) was grown in multiple host cells representing vertebrate and mosquito hosts, and total protein content of purified virions was determined. This analysis identified host factors not previously associated with alphavirus entry, replication, or egress.

Medical aspects of bio-terrorism.

INTRODUCTION: Bioterrorism is a terrorist action involving the intentional release or dissemination of a biological warfare agent (BWA), which includes some bacteria, viruses, rickettsiae, fungi or biological toxins. BWA is a naturally occurring or human-modified form that may kill or incapacitate humans, animals or plants as an act of war or terrorism. BWA is a weapon of choice for mass destruction and terrorism, because of the incubation period, less effective amount than chemical warfare agents, easily distribution, odorless, colorless, difficult to detect, no need of specialized equipment

Efficacy and immunological correlates of an mRNA-LNP vaccine for protection against an emerging rickettsial pathogen.

A rickettsial pathogen, Ehrlichia chaffeensis, causes an emerging tick-borne disease called human monocytic ehrlichiosis (HME). E. chaffeensis in ticks expresses a porin OMP-1B, the immunodominant major outer membrane protein, which is required for nutrient uptake across the outer membrane of bacteria. We developed an OMP-1B-encoding mRNA-lipid nanoparticle (hereafter OMP-1B mRNA-LNP) vaccine. Immunization of naïve mice with OMP-1B mRNA-LNP showed significant protection against E. chaffeensis infection following a challenge with E. chaffeensis-infected tick cells. Following vaccination, increa

Seroprevalence of Borrelia burgdorferi sensu lato in horses in Spain.

Borrelia burgdorferi sensu lato is a tick-borne zoonotic pathogen of increasing importance in Europe, responsible for Lyme borreliosis and affecting both humans and domestic animals. In horses, infection is often subclinical and difficult to diagnose due to variable clinical presentation and limitations in diagnostic methods. Epidemiological data remain limited in several countries, including Spain. This study aimed to estimate the first nationwide seroprevalence in horses in Spain and to identify associated risk factors. A total of 716 serum samples from clinically healthy horses across 17 au

Clinically Irrelevant Terminal 16q21 Deletion Detected by NIPT Is Attributable to Inherited Fragility at FRA16B.

Genome-wide non-invasive prenatal testing (NIPT) is a powerful tool for prenatal detection of the common aneuploidies causing Down-, Edwards-, and Patau syndrome. Its genome-wide reach also enables the detection of unbalanced structural chromosomal abnormalities. We report a case where NIPT indicated a ~25 Mb terminal deletion of the long arm of chromosome 16, later confirmed to be mosaic in maternal blood and buccal cells (in 35%-45% of cells). Remarkably, the same mosaic deletion was seen in the child, who was born healthy at term after an uneventful pregnancy. Further analysis using bromode

Plasma Metagenomic Sequencing Expedites Diagnosis of Disseminated BCG in an Infant With IKBKB Mutation.

BACKGROUND: Infants with inborn errors of immunity (IEI), born in countries where Bacillus-Calmette-Guerin (BCG) vaccination is recommended at birth, are at risk of developing infectious complications following vaccination. A prompt diagnosis of disseminated BCG infection in these infants is essential, as many will require stem cell transplantation (SCT) for the immunologic cure. In patients with IEI, the mortality risk from disseminated mycobacterial infection is high, both before and following SCT. METHODS: A 7-month-old Qatari infant with an IEI, homozygous IKBKB gene mutation, was evaluat

Detection of new Crimean-Congo haemorrhagic fever virus genotypes in ticks feeding on deer and wild boar, Spain.

Crimean-Congo haemorrhagic fever virus (CCHFV) is the causative agent of the severe tick-borne, often fatal, zoonotic Crimean-Congo haemorrhagic fever (CCHF), which is widely distributed worldwide. The CCHFV transmission to humans occurs through tick bite, crushing of engorged ticks or contact with infected host blood. Previously, CCHFV genotype Africa III was reported in Spain. Given the emergence of CCHF and the role of ticks in pathogen maintenance and transmission, we investigated the presence and genotype identity of the virus in tick species parasitizing abundant wild host species in sou
