Certain mutations in the unc-105 II gene of the nematode Caenorhabditis elegans have dominant effects on morphology and behavior: animals become small, severely hypercontracted and paralyzed. These unc-105 mutants revert both spontaneously and with mutagens at high frequencies to a wild-type phenotype. Most of the reversion events are intragenic, apparently because the null (loss-of-function) phenotype of unc-105 is wild type. One revertant defined an extragenic suppressor locus, sup-20 X. Such suppressor alleles of sup-20 are rare, and the apparent null phenotype of sup-20 is embryonic lethal

Klippel-Trenaunay Syndrome with hemicranial hypertrophy and macrodactyly is reported. The patient was a 3-year-old Japanese woman with an extensive nevus flammeus which extended over the right side of the neck, axilla, upper arm, and leg. Her right extremities were longer and thicker than left, and showed apparent macrodactyly in the right hand. Her skin of right side body was irregular, rough surface and was pigmented prominently. Hyperthermia was present in the hypertrophic limbs, but hyperhidrosis was none. She had moderate mental retardation, delayed speech. Her hemicranial hypertrophy (sa

Cytogenetic effects in preimplantation 4-8-cell mouse embryos have been investigated after treating paternal postmeiotic germ cells with triethylenemelamine (TEM). Dose-levels of TEM which do not affect fertilization but yield high incidence of dominant-lethal mutations in sperm and spermatids were shown to produce relatively high frequencies of (a) premature chromosome condensation (PCC), (b) structural chromosome anomalies (breakage-reunion phenomena), and (c) micronuclei in these embryos. The results indicate that genetic death of embryos is mainly due to imbalance (i.e. loss) of genetic ma

UNLABELLED: Zika virus (ZIKV) has multiple lineages and strains that cause a range of disease severity, underscoring the need to elucidate differential neuropathogenesis mechanisms. Here, we performed systematic, side-by-side comparisons of African, Asian, and American ZIKV lineage infections using cerebral organoids derived from human embryonic stem cells, a relevant human model experimental system. African lineage ZIKV strains, as well as the ancestral Asian Malaysia strain, persistently infected neural progenitor cells, causing apoptosis and severe disruption of ventricular cytoarchitecture

The ParaHox homeobox genes Gsx, Xlox, and Cdx are evolutionarily related to Hox genes and form part of the ANTP-class homeobox gene repertoire. Comparative genomic data indicate that ParaHox genes were already present before the cnidarian-bilaterian split. Across metazoans, ParaHox genes often show conserved associations with the anteroposterior body axis and have been implicated in both gut patterning and neural development, although gene complements and genomic organization vary substantially among lineages. To investigate ParaHox gene deployment in Gnathifera, we identified orthologs of Gsx

OBJECTIVES: Postoperative arrhythmias are frequent after tetralogy of Fallot (ToF) repair, yet anatomic substrate and preventive strategies remain poorly defined. Using hierarchical phase-contrast tomography (HiP-CT), the atrioventricular conduction system in pediatric ToF specimens was investigated nondestructively and in 3-dimensions (3D).
METHODS: Eighteen whole-heart specimens (11 ToF, 7 controls) were imaged at the European Synchrotron Radiation Facility. Segmentation and 3D renderings demonstrated gross morphology. Morphology, size, depth, and course of the nonbranching bundle and right

INTRODUCTION AND IMPORTANCE: Diprosopus, or craniofacial duplication, is an extremely rare congenital malformation, representing only 0.4% of conjoined twins with an estimated prevalence of 1 in 15 million births. Fewer than 40 cases have been reported worldwide. We describe a case of partial craniofacial duplication in a child from rural Ethiopia, highlighting the challenges of delayed diagnosis and management in a resource-limited setting.
CASE PRESENTATION: A 7-year-old girl presented with a congenital mandibular mass that had remained untreated since birth due to a lack of antenatal care

Maternal immune activation (MIA) contributes to neurodevelopmental disorders with male-biased prevalence. To disentangle the contributions of sex chromosomes (XX vs. XY) and gonads (ovaries vs. testes) in shaping fetal responses to MIA, we used the four-core genotypes (FCG) mouse model. Offspring representing all combinations of sex chromosome and gonadal status were generated and exposed to poly(I:C) or saline at embryonic day (E)12.5. We assessed maternal serum, fetal placentas, and fetal brain cytokines at E13.5; at E17.5, fetal development was evaluated via microultrasound and CT imaging.

Stem cell therapy presents a new solution to cure degenerative diseases of the retina, which normally have limited ability to regenerate after injury. Stem cells are cells capable of self-renewal and differentiation into more specialized cells, and can be derived from both embryonic and adult sources. This definition encompasses a heterogeneous group of cells possessing varied potency and characteristics. Current attempts in applying stem cell technology to the retina have primarily focused on regenerating the retinal pigment epithelium (RPE) and light-sensing photoreceptor cells. They have no

Fetus-in-fetu (FIF) is an uncommon congenital anomaly arising from aberrant monozygotic twinning, in which a malformed parasitic twin becomes incorporated within the body of the host. Despite its rarity, FIF remains clinically relevant because it is frequently misdiagnosed as a teratoma, particularly when detected outside the neonatal period or at atypical anatomical sites. This review present a Preferred Reporting Items for Systematic Reviews and Meta-Analyses (PRISMA)-informed narrative synthesis of the human literature on FIF, integrating embryological concepts, diagnostic imaging features,

Actinobacteria of mangrove habitats are the potential source of natural products. The work has been undertaken to optimize cultural conditions and elucidate the bioactive compounds produced by a mangrove derived potent rare actinobacterium. Pseudonocardia parietis VLCH-6 was proved as the potent actinobacterium strain, which was derived from the Machilipatnam mangrove habitats of Andhra Pradesh. This identification was based on polyphasic taxonomy, which included morphological, cultural, physiological, biochemical, and molecular approaches. During optimization process, ISP-2 (YMD) broth was re

With the rise in immunocompromised individuals and patients with immune-related comorbidities such as COVID-19, the rate of fungal infections is growing. This increase, along with the current plateau in antifungal drug development, has made understanding the pathogenesis and dissemination of these organisms more pertinent than ever. The mouse model of fungal infection, while informative on a basic scientific level, has severe limitations in terms of translation to the human disease. Here we present data supporting the implementation of the human cerebral organoid model, which is generated from

This study compared the follicular growth, superovulatory response, and in vivo embryo production after administering two doses of porcine follicle-stimulating hormone (pFSH) in Santa Inês ewes. The estrous cycle of 36 multiparous ewes was synchronized with the Day 0 protocol and superovulated with 133 mg (G133, n=18) or 200 mg (G200, n=18) of pFSH. Ultrasonographic evaluations of the ovaries were performed, ewes were mated and submitted to non-surgical embryo recovery. Viable blastocysts were stained with Nile Red and Hoechst. The G200 had a greater number of medium and large follicles, as we

Sonic hedgehog (Shh) is a morphogen with important roles in embryonic development and in the development of a number of cancers. Its activity is modulated by interactions with binding partners and co-receptors including heparin and heparin sulfate proteoglycans (HSPG). To identify antagonists of Shh/heparin binding, a diverse collection of 34,560 chemicals was screened in single point 384-well format. We identified and confirmed twenty six novel small molecule antagonists with diverse structures including four scaffolds that gave rise to multiple hits. Nineteen of the confirmed hits blocked bi

Mediastinal cysts (MCs) are rare lesions that can arise from embryonic remnants of various mediastinal structures. MCs usually are incidental findings in dogs and cats; however, they can reach a mass-like appearance. The description of MCs on CT in dogs is limited. This retrospective, single-center, descriptive, prevalence study aimed to determine the prevalence of presumed mediastinal cysts (PMCs) in dogs and assess their CT characteristics. Dogs that underwent a thoracic CT scan from January 2019 to August 2021 were included. CT images were evaluated for the presence of PMCs by two diagnosti

Human early development sets the stage for embryonic and adult life but remains difficult to investigate. A solution came from the ability of stem cells to organize into structures resembling preimplantation embryos-blastocysts-that we termed blastoids. This embryo model is available in unlimited numbers and could thus support scientific and medical advances. However, its predictive power depends on how faithfully it recapitulates the blastocyst. Here, we describe how we formed human blastoids that (1) efficiently achieve the morphology of the blastocyst and (2) form lineages according to the

BACKGROUND: Rubinstein-Taybi syndrome (RSTS) is a multi-system neurodevelopmental condition caused by deficiency of CREBBP (16p13.3) or EP300 (22q13.2). Müllerian agenesis, or Mayer-Rokitansky-Küster-Hauser (MRKH) syndrome, is defined as congenital agenesis of the uterus, cervix, and upper vagina without a definite genetic cause.
INDEX CASE AND CASE SERIES: We present a 14-year-old female with RSTS type 1 (CREBBP, c.4395-2A>C) and MRKH, the first documented in the literature. Following presentation to Gynecology for anticipatory guidance regarding future menstrual suppression and follow-up of

Glaucomatous optic neuropathy (GON) is the major cause of irreversible visual loss worldwide and can result from a range of disease etiologies. The defining features of GON are retinal ganglion cell (RGC) degeneration and characteristic cupping of the optic nerve head (ONH) due to tissue remodeling, while intraocular pressure remains the only modifiable GON risk factor currently targeted by approved clinical treatment strategies. Efforts to understand the mechanisms that allow species such as the zebrafish to regenerate their retinal cells have greatly increased our understanding of regenerati

Skeletal stem/progenitor cells (SSPCs), characterized by self-renewal and multipotency, are essential for skeletal development, bone remodeling, and bone repair. These cells have traditionally been known to reside within the bone marrow, but recent studies have identified the presence of distinct SSPC populations in other skeletal compartments such as the growth plate, periosteum, and calvarial sutures. Differences in the cellular and matrix environment of distinct SSPC populations are believed to regulate their stemness and to direct their roles at different stages of development, homeostasis

Imidacloprid is a systemic insecticide that belongs to the neonicotinoid class of chemicals that act on the central nervous system of insects. Imidacloprid is used to control sucking insects, chewing insects such as termites, soil insects, and fleas on pets, as well as to treat structures, crops, soil, and seeds. As a result of these factors, this pesticide may end up in the aquatic environment via municipal discharges and runoff. Although the presence of imidacloprid in aquatic environments has been underreported as widespread, the toxic effects of this pesticide may have serious implications
