Na+-stimulated H+ movement and H+-stimulated Na+ uptake were studied in basolateral (blLPM) and canalicular (cLPM) rat liver membrane vesicles. H+ movement was monitored with the fluorescent amine acridine orange; 22Na uptake was assayed by a rapid Millipore filtration technique. In blLPM, inwardly directed Na+ gradients stimulated H+ efflux and outwardly directed Na+ gradients stimulated proton influx. Outwardly directed proton gradients (pH in 5.9/pH out 7.9) stimulated initial 22Na uptake rates 5- to 10-fold over pH-equilibrated conditions (pH in 7.9/pH out 7.9). Conversely, inwardly direct

Direct microscopy of keratinised specimens is a standard screening procedure that assists clinicians to differentiate true superficial mycoses from non-fungal disorders of the skin, nail and hair. Most clinical dermatologists use bright-field microscopy when searching for dermatophyte fungi in clinical samples while laboratory-based mycologists increasingly favour fluorescence microscopy in order to optimise visualisation of fungal elements. This study compared the validity and speediness of fluorescence microscopy vs. conventional light microscopy when screening for fungi in 206 dermatologica

The Bicaudal-C (Bic-C) gene was originally discovered in Drosophila melanogaster. The gene product Bic-C is thought to serve as an RNA-binding molecule targeting diverse proteins at the post-transcriptional level. Recent research has shown this gene to be conserved in many species, from Caenorhabditis elegans to humans. Disruption of this protein can disturb the normal migration direction of the anterior follicle cell of Drosophila oocytes, while mutation of a mouse Bicc1 (a mouse homologue of Bic-C) results in phenotypes mimicking human hereditary polycystic kidney disease (PKD). However, the

The photophysical and chiroptical properties of chiral emissive molecules can be influenced by both intrinsic molecular chirality and molecular organization, although their respective contributions remain insufficiently understood. In this study, we synthesized enantiomers of two Zn(II) complexes with different coordination environments to investigate how the ligand structure affects supramolecular organization and emission properties in axially chiral BINOL-derived Schiff-base Zn(II) complexes. The Schiff-base ligands were prepared by the condensation of a BINOL-derived aldehyde with amine co

Antarctic Mokoshia mucilaginosa and Mokoshia rubra are associated with improved salinity tolerance in Nicotiana tabacum through coordinated regulation of photosynthetic performance and antioxidant responses, highlighting the potential of polar bacteria as bioinoculants for sustainable agriculture in saline and cold-affected ecosystems. Soil salinity is an escalating global challenge that constrains crop productivity worldwide, with particularly severe impacts in marginal agroecosystems, including those in cold regions. Here, we provide evidence that two psychrotolerant Antarctic bacterial stra

BACKGROUND: Metabolic dysfunction-associated steatotic liver disease (MASLD) is a metabolic syndrome characterized by increased fat storage in hepatocytes. In the hepatocytes, autophagy protects against cytotoxic stress and harmful cellular conditions. In the hepatic stellate cells (HSCs), autophagy exerts pro-fibrotic properties and promotes the release of pro-inflammatory metabolites. We investigated the modulation of autophagy as a therapeutic approach for MASLD.
METHODS: Murine liver tissue and human hepatic cells were analyzed by reverse transcription quantitative polymerase chain reacti

Aging-associated changes in myeloid cells are incompletely understood. One of the culprits of aging is the downregulation of the Tert gene coding for the catalytic subunit of telomerase. Studies of mouse models with Tert knockout (KO) in specific cells have revealed the importance of the telomere-independent noncanonical function of TERT in supporting mitochondrial metabolism and protection from cell senescence. To investigate the role of TERT in myeloid cells (MCs), we analyzed mice with Tert KO in the LysM+ lineage, indelibly labeled with membrane green fluorescent protein (mG). Macrophage n

Traditional evaluations of the hair follicle cycle rely on invasive histological methods, limiting continuous longitudinal observation. It was hypothesized that non-invasive optical imaging, particularly two-photon excited fluorescence (TPEF) and second-harmonic generation (SHG), can accurately track and quantify in vivo dynamic changes in hair shafts, melanin, and collagen across multiple hair follicle cycles. The hair shafts of C57BL/6 mice were monitored in vivo during the anagen, catagen, and telogen phases using dermoscopy, photography, and electron microscopy. Specifically, TPEF under 80

Pursuing innovative modulation strategies and high asymmetry factors (glum) remains a cornerstone in developing circularly polarized luminescence (CPL) materials. Conventional approaches employ either bottom-up chiral small molecule self-assembly or top-down co-assembly of achiral luminophores with supramolecular chiral templates (e.g., cellulose nanocrystals (CNCs), liquid crystals). These templates act as pre-engineered "molds" to align achiral emitters within chiral matrices. However, co-assembling chiral small molecules with such templates remains underexplored, challenged by complex proto

Constructing perovskite heterostructures with restricted interface charge transfer is crucial for improving stability and optoelectronic performance, as well as expanding multifunctional applications. Herein, a one-step solvent-free thermal assisted epitaxial growth strategy is proposed to construct BaMoO4/CsPbX3 (X = Cl, Br, I) heterostructures. Derived from the high lattice matching of 90.3 %-100 %, CsPbX3 QDs were epitaxially grown in situ on the surface of the BaMoO4 substrate. Compared with CsPbBr3 QDs, the BaMoO4/CsPbBr3 heterostructure exhibited significantly improved photoluminescence

Vaccination of bovine calves is one of the main policies for bovine brucellosis control in endemic areas. However, the effect of animal age on vaccine immunogenicity is still unknown and could help to determine an ideal age for vaccination, in order to maximize immune response. Thus, the objective of this study was to compare the in vitro expression of IFN-γ by stimulated PBMC after vaccination with B. abortus S19 and RB51 strains in calves vaccinated at different ages, between 3 and 8 months. Cell-mediated immune response was assessed through culture of peripheral blood mononuclear cells (PBM

Statins are one of the top prescribed medications and are used for preventing or treating cardiovascular diseases. Myalgia, muscle fatigue, weakness, and inflammation are the most common side effects of these drugs collectively named statin-associated muscle symptoms (SAMS). The mechanisms underlying SAMS remain unclear. Given that statins inhibit 3-hydroxy-3-methylglutaryl coenzyme A reductase, the rate-limiting enzyme of mevalonate pathway, responsible for synthesis of cholesterol and other vital molecules, SAMS may be mediated by multiple reasons. Herein, using unbiased whole transcriptome

Multitarget drug discovery, including host-directed therapy, is particularly promising for tuberculosis (TB) due to the resilience of Mycobacterium tuberculosis (Mtb) as well as the complexity of the host's immune response. In this proof-of-concept study, we used high-content imaging to test a novel panel of dual glycogen synthase kinase 3 beta (GSK-3β) and histone deacetylase (HDAC) 1 and 6 inhibitor candidates for their efficacy in reducing the growth of green fluorescent protein (GFP)-expressing mycobacteria in human primary macrophages. We demonstrate that all ten test compounds, also incl

Amidst the health crisis caused by the rise of multi-resistant pathogenic microorganisms, Antimicrobial Peptides (AMPs) have emerged as a potential alternative to traditional antibiotics. In this sense, Cm-p5 is an AMP with fungistatic activity against the yeast Candida albicans. Its antimicrobial activity and selectivity have been well characterized; however, the mechanism of action is still unknown. This study used biophysical approaches to gain insight into how this peptide exerts its activity. Stability and fluidity of lipid membrane were explored by liposome leakage and Laurdan generalize

Bolaamphiphiles or bolaforms have drawn particular interest in drug and gene delivery, and studies of bolaforms have been growing continuously. Bolaforms, due to their unique structure, exhibit specific self-assembly behavior in water. The present work aims to develop biodegradable cationic bolaforms with a better gene transfection ability. In this work, a novel cationic bolaform (Bola-1) with head groups bearing hydroxyl (OH) functionality was designed and synthesized to investigate self-assembly and gene transfection efficiency. The self-assembly behavior of Bola-1 in water was compared with

This paper presents the photophysical and biological properties of eight 3-imino-1,8-naphthalimides. The optical properties of the compounds were investigated in the solvents that differed in their polarity (dichloromethane, acetonitrile, and methanol), including three methods of sample preparation using different pre-dissolving solvents such as dimethyl sulfoxide or chloroform. In the course of the research, it was found that there are strong interactions between the tested compounds and DMSO, which was visible as a change in the maximum emission band (λem) of the neat 3-imino-1,8-naphthalimi

Our goal was to identify the factors with the strongest influence on the minimum lamina cribrosa (LC) oxygen concentration as potentially indicative of conditions increasing hypoxia risk. Because direct measurement of LC hemodynamics and oxygenation is not yet possible, we developed 3D eye-specific LC vasculature models. The vasculature of a normal monkey eye was perfusion-labeled post-mortem. Serial cryosections through the optic nerve head were imaged using fluorescence and polarized light microscopy to visualize the vasculature and collagen, respectively. The vasculature within a 450 μm-thi

The construction of main group heteroatom-stereogenic compounds is of great importance due to their intriguing chemical, physical, biological, and stereoelectronic properties. Despite that organoboron compounds are widely used in organic chemistry, the creation of a tetrahedral boron-stereogenic center in one enantiomeric form remains highly challenging. Given the labile nature of ligands attached to the tetracoordinate boron atom, only a handful of enantioenriched boron-stereogenic compounds have been reported via resolution or a chiral substrate-induced diastereoselective approach. To date c

Alzheimer's Disease (AD) is the most common neurodegenerative disease, and efficient therapeutic and early diagnostic agents for AD are still lacking. Herein, we report the development of a novel amphiphilic compound, LS-4, generated by linking a hydrophobic amyloid-binding distyrylbenzene fragment with a hydrophilic triazamacrocycle, which dramatically increases the binding affinity toward various amyloid β (Aβ) peptide aggregates, especially for soluble Aβ oligomers. Moreover, upon the administration of LS-4 to 5xFAD mice, fluorescence imaging of LS-4-treated brain sections reveals that LS-4
