The accumulation of the two branched-chain fatty acids phytanic acid and pristanic acid is known to play an important role in several diseases with peroxisomal impairment, like Refsum disease, Zellweger syndrome and α-methylacyl-CoA racemase deficiency. Recent studies elucidated that the toxic activity of phytanic acid and pristanic acid is mediated by multiple mitochondrial dysfunctions, generation of reactive oxygen species and Ca2+ deregulation via the InsP3-Ca2+ signaling pathway in glial cells. However, the exact signaling mechanism through which both fatty acids mediate toxicity is still

The liver is a metabolically active organ vital for carbohydrate, protein, and lipid metabolism as well as detoxification of harmful substances. Here, we integrated multiple reaction monitoring (MRM) profiling, untargeted lipidomics, and desorption electrospray ionization-mass spectrometry imaging (DESI-MSI) to identify age-associated changes in lipid profiles in the liver of adult mice across three age groups: adult (3-4 months), midaged (10 months), and old (19-21 months). Comparative lipidomic analysis revealed age-dependent remodeling of membrane phospholipids and sphingolipids. Aging was

This review outlines the health risks associated with excessive dietary intake of n-6 polyunsaturated fatty acids (PUFAs), particularly linoleic acid (C18:2n-6, LA), which is highly prevalent in the Western diet. It proposes a targeted nutritional strategy to reduce n-6 PUFA overconsumption and increase n-3 PUFA intake, aiming to restore a healthier fatty acid balance and counteract imbalance-induced pathogenetic consequences. The conceptual framework builds on the foundational insights of William E. M. Lands regarding PUFA-driven eicosanoid imbalance. It extends these principles by integratin

Inducing immunogenic cell death (ICD) in cancer cells provides a promising approach in immunotherapy, however, oxidative stress relief and metabolic plasticity enhancement limit the immune-stimulating effect of traditional ICD inducer in sentinel lymph nodes (SLN), owing to the metabolism of fatty acids. In this article, a biocompatible green liposome CL-Lip was designed to not only selectively induce ICD in primary and metastatic 4T1 cells, but also relieved the immunosuppression in invasive SLN. CL-Lip is composed of engineered liposomes modified with linoleic acid and catalase, which synerg

Delayed cerebral ischemia (DCI) following aneurysmal subarachnoid haemorrhage (aSAH) is a complex and acute condition with limited options for early detection and effective treatments. The plasma levels of individual polyunsaturated fatty acids (PUFA) and their bioactive metabolites (oxylipins) of aSAH patients both at admission and over time remain largely unexplored, particularly concerning the development of DCI. In this study, plasma samples of aSAH patients were collected at admission and on days 4, 10, and 21 post-admission. ASAH patients who did not develop DCI were age- and sex matched

Florfenicol, a widely used veterinary antibiotic, has now been frequently detected in various water environments and human urines, with high concentrations. Accordingly, the ecological risks and health hazards of florfenicol are attracting increasing attention. In recent years, antibiotic exposure has been implicated in the disruption of animal glucose metabolism. However, the specific effects of florfenicol on the glucose metabolism system and the underlying mechanisms are largely unknown. Herein, zebrafish as an animal model were exposed to environmentally relevant concentrations of florfeni

Cost is the crucial impediment in commercializing microalgal biodiesel. Therefore, cultivating microalgae in cost-effective nutrients reduces the upstream process cost remarkably. Thus, in this study, sugar cane bagasse hydrolysate (SBH) as a lucrative carbon supplement for Chlorococcum sp. and subsequent lipid extraction via an optimized solvent system for biodiesel production was investigated. Characterization of SBH revealed the presence of various monosaccharides and other sugar derivatives such as glucose, fructose, xylose, arabinose, etc. The maximum dry cell weight of 1.7 g/L was estima

Metabolic syndrome (MS) is a common metabolic disorder characterized by obesity, insulin resistance, cardiovascular disease and gut microbiota dysbiosis. Pu-erh tea and its ingredient theabrownin have known functions on the reduction of body weight gain and fat accumulation. However, few studies systematicly analyze the different contributions and mechanisms of their anti-metabolic syndrome functions through multi-omics combination analysis. Here, we used metagenomics, transcriptomics and metabolomics technology to investigate the anti-metabolic syndrome mechanism of Pu-erh tea and theabrownin

Coronary heart disease (CHD) is a complex human disease associated with inflammation and oxidative stress. The underlying mechanisms and diagnostic biomarkers for the different types of CHD remain poorly defined. Metabolomics has been increasingly recognized as an enabling technique with the potential to identify key metabolomic features in an attempt to understand the pathophysiology and differentiate different stages of CHD. We performed comprehensive metabolomic analysis in human plasma from 28 human subjects with stable angina (SA), myocardial infarction (MI), and healthy control (HC). Sub

Obesity-related changes in adipose tissue alter local secretory profiles and influence interactions with other surrounding cells, including cancer cells. The continued delivery of nutrients shifts the metabolism of fat cells, making them act as a lipid reservoir. Typically, research on obesity mechanisms relies on animal models, which, however, do not allow analysis of metabolic processes at the cellular level. Hence, there is a need to develop a reliable in vitro model of human obesity that can help explore changes in the function of obese adipocytes compared to their lean counterparts. This
