Altered structural connectivity in white matter lesion-induced mild cognitive impairment: a probabilistic fiber tracking and graph theory analysis.

White matter lesions (WMLs) are a common cause of vascular cognitive impairment, yet the nature of whole-brain structural connectivity alterations in WML-related mild cognitive impairment (MCI) remains incompletely understood. In this study, an integrated approach combining whole-brain probabilistic fiber tracking with graph theory analysis was employed to investigate structural network changes in 33 patients with WML-related MCI and 37 healthy controls. The results revealed that, at the MCI stage, the brain's structural network exhibited not only reduced connectivity in specific neural circui

Disrupted multi-frequency topological properties of brain functional networks in attention networks after repetitive subconcussion.

Attention impairment constitutes a primary cognitive deficit following subconcussive head impacts. However, the underlying neural mechanisms remain unclear. This study investigated neural alterations in attention networks associated with repetitive subconcussive exposure by integrating source-level electroencephalography (EEG) functional connectivity analysis with the Attention Network Test (ANT). Source-level functional connectivity matrices were constructed using the phase-locking value (PLV) to characterize connectivity patterns across multiple frequency bands (delta, theta, alpha, and beta

Temporal-orbitofrontal pathway regulates choices across physical reward and visual novelty.

Expectation of perceptual novelty shapes our daily decisions and has an impact on how we evaluate primary rewards. To identify the underlying circuitry, we trained subjects to choose between offers associated with varying probabilities of receiving large juice rewards and experiencing novel objects. Anticipated novelty modulated valuation by increasing both choice preference for and motivational value of large rewards. This novelty-reward interaction was reflected in a circuit comprising anterior ventral medial temporal cortex (AVMTC), implicated in detecting and predicting novelty, and orbito

Three parsimonious spatiotemporal patterns in cerebellum reveal individual traits in function and behavior.

The cerebellum plays a critical role in large-scale brain functional connectivity structure and synchronizing global waves propagation. Despite extensive studies on cortical spatiotemporal architecture, particularly in zero-lag and time-lag synchrony, leaving the parsimonious cerebellar representational structure largely overlooked. Using complex principal component analysis (CPCA) on resting-state fMRI data from the Human Connectome Project, we delineate three dominant low-dimensional spatiotemporal patterns within the cerebellum, manifesting as unimodal-to-transmodal progression, functional

Top-down control of sustained attention by medial prefrontal cortex-locus coeruleus (mPFC-LC) projection neurons during the rodent continuous performance test (rCPT).

The medial prefrontal cortex (mPFC) plays a pivotal role in attention by exerting top-down control to allocate cognitive resources toward behaviorally relevant stimuli based on learned context and expectations. mPFC neurons project to multiple cortical and subcortical regions, including the locus coeruleus (LC)-the brain's primary source of norepinephrine (NE). The mPFC also receives inputs from the LC, which release NE to modulate mPFC neuronal activity and downstream cellular signaling. While enhanced functional connectivity between the mPFC and LC in mice during sustained attention tasks su

Double dissociation between conduction aphasia and conduction agraphia supports a ventro-dorsal partition of the left arcuate fasciculus.

We identified in two awake surgery cases a postoperative double dissociation between phonological and graphemic output buffer deficits. Using lesion-symptom mapping from ischaemic mini-strokes and preoperative tractography, we demonstrated that the phonological (resp. graphemic) disorder fitted with ventral (resp. dorsal) damage to the AF. Further studies are needed to confirm our hypothesis of a ventro-dorsal functional organization within the AF for phonological versus graphemic processing.

Dimorphic substrates, monomorphic outcomes: Social behavior neural circuit tuning in social monogamy.

Social behaviors are dramatically influenced by variations in mating systems that shape sex differences in the neural substrates underlying such displays. In polygamous species, distinct evolutionary pressures on males and females produce pronounced behavioral and anatomical sexual dimorphism, supported by strongly sexually differentiated social behavior and social decision-making networks at levels of brain structure, connectivity, physiology, and gene expression. By contrast, socially monogamous species display more similar or monomorphic social behaviors-such as shared parenting, convergent

Whole-brain input organization of Kremen1-positive striatal spiny projection neurons revealed by monosynaptic rabies tracing.

Determining the organization of inputs to subtypes of striatal spiny projection neurons (SPNs) is essential to understand how the striatum integrates information to regulate motor control and decision making. Here, we employed monosynaptic rabies tracing in Kremen1 2A-Cre knock-in mice to map inputs to Kremen1-positive (Kremen1 +) SPNs, a subtype enriched in patch (striosome) compartments of the dorsal striatum. Starter cells were broadly distributed along the rostrocaudal axis of the dorsal striatum. Whole-brain analysis revealed that Kremen1 + SPNs receive prominent inputs from motor, somato

Single-neuron connectome of nucleus accumbens.

The nucleus accumbens (NAc) orchestrates a range of critical physiological functions, including reward and motivation, yet single-neuron-level connectivity of its inputs and outputs remains less comprehensively characterized. Here, we examined the organizing principle of input and projection of medium spiny neurons (MSNs) across NAc in adult male mice at single-cell resolution. Our results revealed topographic distribution principles of these neurons and their terminals in the substantia innominata (SI) subdivision. Rabies virus tracing uncovered different monosynaptic inputs to Tac2+ (D1 subt

Anatomical characterization of premotor-motor connectivity in humans by paired intraoperative direct cortical stimulation.

The human motor cortex represents the major source of corticospinal output and receives input from a constellation of sensorimotor regions in the surrounding cerebral cortex and subcortical regions. Here we tested cortico-cortical connections by means of direct cortical stimulation in intraoperative settings. We used short trains of 3 pulses (0.2 ms duration, 2 ms ISI) as test stimuli to the motor cortex and as conditioning stimuli to the premotor cortex in 14 human patients undergoing brain surgery for tumour resection under general anaesthesia with intraoperative neurophysiological monitorin

Synergistic Reinforcement Learning by Cooperation of the Cerebellum and Basal Ganglia.

The cerebral cortex, cerebellum, and basal ganglia are essential for flexible learning in mammals. Although traditionally thought to operate under different learning rules, recent evidence suggests that both the basal ganglia and the cerebellum may employ reinforcement learning mechanisms. This raises the question of how these structures coordinate when a common reward prediction error mechanism is active. To address this issue, we first examined output signals from the basal ganglia and cerebellum following the activity of the cerebral cortex. We recorded single-neuron activity from the outpu

Convergence and Segregation of Excitatory and Inhibitory Afferents in the Paraventricular Thalamic Nucleus.

The paraventricular thalamic nucleus (PVT) integrates subcortical signals related to arousal, stress, addiction, and anxiety with top-down cortical influences. Increases or decreases in PVT activity exert profound, long-lasting effects on behavior related to motivation, addiction, and homeostasis. Yet the sources of its subcortical excitatory and inhibitory afferents, their distribution within the PVT, and their integration with layer-specific cortical inputs remain unclear. Using transgenic male and female mice selective for GABAergic and glutamatergic neurons, or for different cortical layer

Investigation of symptom-specific functional connectivity patterns in Parkinson's disease.

Parkinson's disease (PD) is a complex neurodegenerative disease, characterized by pronounced heterogeneity in symptoms. This study investigates the functional connectivity (FC) patterns associated with distinct symptom clusters, aiming to elucidate the heterogeneity in PD and uncover the neural mechanisms underlying its motor and cognitive symptoms. Resting-state functional MRI (rs-fMRI) data from 55 non-demented PD patients and 24 healthy controls (HC) were used to perform seed-to-seed FC analyses. A clustering algorithm was applied to the cognitive and motor scores of all PD patients to gene

Disconnection between parietal and temporal areas without simultanagnosia: a case study of prosopagnosia.

This study presents a neuropsychological evaluation of a unique case of prosopagnosia (patient EP) with atypical lesion patterns, characterized by intact face-selective nodes but significant damage to the Vertical Occipital Fasciculus (VOF). Given the presumed interruption of ventral-parietal connectivity, we focused on assessing the potential presence of simultanagnosia and its potential relationship to his face recognition deficits. Our neuropsychological battery included tests of global and local processing, scene perception, and face recognition. Results revealed intact global processing a

Topological and geometric signatures of brain network dynamics in Alzheimer's disease.

INTRODUCTION: This study explores magnetic resonance imaging (MRI) as a promising non-invasive approach to monitor Alzheimer's disease (AD) and related dementias. We investigate whether dynamic functional connectivity (dFC), which captures time-varying neural interactions, can reveal sex-specific brain network disruptions in AD that conventional static connectivity analyses may miss. METHODS: We analyzed dFC in the Open Access Series of Imaging Studies (OASIS-3) dataset across three diagnostic groups (normal cognition, mild cognitive impairment, dementia), stratified by sex, and regressed out

State-dependent functional network connectivity alterations in post-stroke dementia with subcortical lesions.

BACKGROUND: Recent studies have revealed disrupted dynamic functional network connectivity (dFNC) in stroke or dementia brains. However, it remains unclear how the dFNC was altered in post-stroke dementia (PSD). OBJECTIVE: This study aimed to explore PSD-specific alterations in the dFNC and their association with clinical assessments. METHODS: We included 19 normal controls (NC), 16 PSD, and 20 post-stroke non-dementia (PSND) patients who underwent resting-state functional magnetic resonance imaging scan. Cerebello-Thalamo-Cortical Dysconnectivity in Schizophrenia Spectrum Disorders: A Resting-State Functional Magnetic Resonance Imaging Meta-Analysis.

BACKGROUND: Cerebello-thalamo-cortical (CTC) network dysfunctions are well documented in schizophrenia spectrum disorders (SSDs) and preclinical states. However, small samples and methodological heterogeneity often limit individual neuroimaging studies. To overcome these challenges, we conducted a coordinate-based meta-analysis to characterize CTC alterations across illness stages and examine associations with symptom dimensions. METHODS: Our meta-analysis was preregistered and followed the Preferred Reporting Items for Systematic Reviews and Meta-Analyses (PRISMA) guidelines and the recommen

Neural basis underlying the effects of trait and state anxiety on premature ejaculation revealed by resting-state interhemispheric functional connectivity.

INTRODUCTION: Anxiety is considered to play a key role in the development and maintenance of premature ejaculation (PE). In addition, PE patients often co-occur with anxiety, however, the central mechanisms underlying this comorbidity have remained elusive. This study aimed to explore whether trait and state anxiety-related PE shared common or distinct mechanisms in the brain. METHODS: A total of 90 PE patients and 45 healthy controls (HCs) were enrolled. Patients were divided into subgroups including 45 patients with low trait anxiety (LTA) and 45 patients with high trait anxiety (HTA) by st
