Integrated genomic analysis defines molecular subgroups in dilated cardiomyopathy and identifies novel biomarkers based on machine learning methods.

Aim: As the most common cardiomyopathy, dilated cardiomyopathy (DCM) often leads to progressive heart failure and sudden cardiac death. This study was designed to investigate the molecular subgroups of DCM. Methods: Three datasets of DCM were downloaded from GEO database (GSE17800, GSE79962 and GSE3585). After log2-transformation and background correction with "limma" package in R software, the three datasets were merged into a metadata cohort. The consensus clustering was conducted by the "Consensus Cluster Plus" package to uncover the molecular subgroups of DCM. Moreover, clinical characteri

Identification of key biomarkers and therapeutic targets in sepsis through coagulation-related gene expression and immune pathway analysis.

Sepsis, characterized by a widespread and dysregulated immune response to infection leading to organ dysfunction, presents significant challenges in diagnosis and treatment. In this study, we investigated 203 coagulation-related genes in sepsis patients to explore their roles in the disease. Through differential gene expression analysis, we identified 20 genes with altered expression patterns. Subsequent correlation analysis, visualized through circos plots and heatmaps, revealed significant relationships among these genes. Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG)

An Integrative analysis of single-cell RNA-seq, transcriptome and Mendelian randomization for the Identification and validation of NAD+ Metabolism-Related biomarkers in ulcerative colitis.

Ulcerative colitis (UC) is a chronic and refractory inflammatory disease of the colon and rectum. This study utilized bioinformatics methods to explore the potential of Nicotinamide adenine dinucleotide (NAD+) metabolism-related genes (NMRGs) as key genes in UC. Using the GSE87466 dataset, differentially expressed NMRGs were identified through differential expression analysis, weighted gene co-expression network analysis (WGCNA), and NMRG scoring. These NMRGs were used as exposure factors, with UC as the outcome, to identify causal candidate genes through Mendelian randomization (MR) analysis.

Automatic Sleep-Stage Scoring in Healthy and Sleep Disorder Patients Using Optimal Wavelet Filter Bank Technique with EEG Signals.

Sleep stage classification plays a pivotal role in effective diagnosis and treatment of sleep related disorders. Traditionally, sleep scoring is done manually by trained sleep scorers. The analysis of electroencephalogram (EEG) signals recorded during sleep by clinicians is tedious, time-consuming and prone to human errors. Therefore, it is clinically important to score sleep stages using machine learning techniques to get accurate diagnosis. Several studies have been proposed for automated detection of sleep stages. However, these studies have employed only healthy normal subjects (good sleep

Predictive Accuracy of the Sport Concussion Assessment Tool 3 and Vestibular/Ocular-Motor Screening, Individually and In Combination: A National Collegiate Athletic Association-Department of Defense Concussion Assessment, Research and Education Consortium Analysis.

BACKGROUND: Vestibular and ocular symptoms in sport-related concussions are common. The Vestibular/Ocular-Motor Screening (VOMS) tool is a rapid, free, pen-and-paper tool that directly assesses these symptoms and shows consistent utility in concussion identification, prognosis, and management. However, a VOMS validation study in the acute concussion period of a large sample is lacking. PURPOSE: To examine VOMS validity among collegiate student-athletes, concussed and nonconcussed, from the multisite National Collegiate Athletic Association-Department of Defense Concussion Assessment, Research

Decoding functional changes in the brain following ischemic stroke: a multimodal feature approach integrating fNIRS with machine learning and deep learning.

BACKGROUND: Ischemic stroke (IS) induces significant structural and functional remodelling of brain networks. While functional near-infrared spectroscopy (fNIRS) offers high temporal resolution for monitoring cerebral hemodynamics, its potential for capturing complex dysfunction patterns remains underutilized. This study investigated task-evoked and resting-state fNIRS responses to decode neurovascular deficits in IS patients compared to normal controls (NCs). METHODS: A total of 160 participants (115 IS patients and 45 NCs) were enrolled and underwent fNIRS recording across three paradigms:

Deciphering the molecular landscape of Sjögren's disease, mucosa-associated lymphoid tissue lymphoma, and thyroid cancer: unraveling the complexities of disease mechanisms and diagnostic biomarkers.

BACKGROUND: Sjögren's disease (SjD), mucosa-associated lymphoid tissue lymphoma (MALT lymphoma), and thyroid cancer (THCA) are clinically distinct yet immunologically intertwined diseases characterized by chronic inflammation and aberrant immune activation. However, the shared molecular basis linking autoimmunity and oncogenesis remains poorly defined. METHODS: We performed an integrative transcriptomic analysis across three datasets (TCGA-THCA, GSE40611/GSE127952 for SjD, and GSE25638 for MALT lymphoma), totaling 749 samples. Analyses included differential expression, Gene Ontology (GO)/Kyot

Pharmacogenomic characterization of a uric acid metabolism-related signature associated with prognosis and drug sensitivity in gastric cancer.

Gastric cancer (GC) remains a major cause of cancer-related mortality worldwide. Increasing evidence suggests that metabolic reprogramming contributes to GC progression, yet the prognostic significance of uric acid metabolism-related genes in GC remains unclear. This study aimed to characterize uric acid metabolism-related genes with prognostic and pharmacological relevance in gastric cancer and to explore their potential value for risk stratification and drug-response prediction. Differentially expressed genes between tumor and normal tissues were obtained from public databases and intersecte

Integrative bioinformatic analysis prioritizes TIMP1 and FN1 as angiogenesis-related candidate genes in diabetic foot ulcers.

BACKGROUND: Diabetic foot ulcers (DFUs) represent a severe chronic complication of diabetes and are characterized by persistent impairment of wound healing, accompanied by defective angiogenesis, chronic inflammation, and dysregulated extracellular matrix remodeling. Although impaired angiogenesis is widely recognized as a key pathological feature of DFUs, its associated molecular alterations have not been systematically characterized at the transcriptomic and cellular levels. METHODS: In this study, bulk transcriptomic data were analyzed in combination with machine learning-based gene priori

A generalizable OPM-MEG framework for time-resolved language decoding during natural speech production.

Non-invasive decoding of rapidly evolving cognitive and motor states is limited by trade-offs between temporal precision, spatial fidelity and tolerance to natural movement. We present a modality-matched benchmarking framework that evaluates optically pumped magnetometer magnetoencephalography (OPM-MEG) against electroencephalography (EEG) during visually cued overt speech production. Ten native Mandarin speakers produced six isolated vowel rhymes (100 repetitions per class) while OPM-MEG and EEG were recorded under the same task. We compared six feature representations and five classifiers us

An interpretable, clinically grounded framework for digital speech biomarker development in neurodegenerative diseases.

INTRODUCTION: Communication ability-a key determinant of quality of life-is frequently affected and progressively declines in neurodegenerative diseases. Effective management of progressive communication disorders requires a personalized approach to deliver timely interventions tailored to the evolving profiles of communicative impairment, thereby supporting functional communication throughout the disease course. To this end, reliable tools capable of detecting and quantifying both disease-specific patterns of communicative impairment and within-disease phenotypic variability are urgently need

A unified multimodal learning framework for sentiment analysis and mental health indicators from YouTube videos.

This study presents a multimodal deep learning framework designed to analyze sentiment patterns in YouTube videos and explore their association with early indicators of mental well-being. The approach integrates textual transcripts, vocal characteristics, and facial expressions into a unified representation to capture the emotional depth that individual modalities often miss. The model was trained and evaluated on a curated dataset of diverse YouTube content, and the results consistently showed that the fused architecture performed better than unimodal baselines. Compared with text-only or aud

Machine learning analysis of s-EASIX for predicting 30-day mortality in sepsis patients from MIMIC-IV.

Endothelial dysfunction is an important risk factor for the progression of sepsis. The simplified endothelial activation and stress index (s-EASIX) serves as an indirect measure of endothelial activation, whose dynamic changes have an unclear association with prognosis in sepsis. Therefore, we conducted this study to investigate the association between clinical subphenotypes indicated by s-EASIX trajectories and 30-d mortality in sepsis. Based on MIMIC-IV v3.1, the association of s-EASIX dynamic trajectories with 30-d mortality in sepsis was investigated in this retrospective cohort analysis.

Resection-inspired histopathological diagnosis of cerebral cavernous malformations using quantitative multiphoton microscopy.

Rationale: Cerebral cavernous malformation (CCM) is prone to recurring microhemorrhage, which can lead to drug-resistant epilepsy. Surgical resection is the first choice to control seizures for CCM-associated epilepsy. At present, removal of resection-related tissue only depends on cautious visual identification of CCM lesions and perilesional yellowish hemosiderin rim by the neurosurgeon. Inspired by the resection requirements, we proposed quantitative multiphoton microscopy (qMPM) for a histopathology-level diagnostic paradigm to assist clinicians in precisely complete resection. Methods: A

HLA-DPA1 as a diagnostic biomarker differentiating early- and late-onset preeclampsia.

The occurrence and development of a wide range of preeclampsia (PE), especially early-onset preeclampsia (EOPE), is closely associated with the immune system. The objective of this research is to utilize machine learning techniques to discover key immune biomarkers and evaluate their predictive potential. We sourced mRNA expression profiles from the GSE60438 + GSE75010 dataset in the Gene Expression Omnibus (GEO) and retrieved immune-related genes from the ImmPort database. Subsequently, we selected immune genes associated with EOPE and late-onset preeclampsia (LOPE) for differential expressio

Development of a diagnostic model for ovarian cancer based on machine learning algorithms and functional analysis of key biomarker SOX17.

BACKGROUND: Ovarian cancer (OC) demonstrates the poorest prognosis among gynecological malignancies, with five-year survival rates below 45%, primarily due to late-stage diagnosis. To address this challenge, we systematically identified OC-specific differentially expressed genes (DEGs) to develop a robust diagnostic model based on eleven machine learning algorithms. Furthermore, we explored the potential mechanism of key DEG in OC. METHODS: We acquired RNA sequencing data of 426 tissues (352 °C tumor and 74 adjacent non-tumor) from the Gene Expression Omnibus (GEO) repository. Following rigor

Neural Markers of Interocular Grouping During Binocular Rivalry With MEG.

Binocular rivalry (BR) occurs when incompatible images are simultaneously presented, one to each eye, and elicit perceptual alternations between image dominance and suppression. Remarkably, BR accommodates interocular grouping (IOG) such that portions of two globally coherent images shown separately to each eye, results in frequent global pattern perception. We investigated the neural correlates of BR and IOG with magnetoencephalography (MEG). Stimuli consisted of red or green orthogonal gratings flickering at 5 or 6.67 Hz, allowing for analysis of tagged fundamental, intermodulation frequenci

Divergent eye-brain-heart fatigue signatures during sustained simulated agricultural operations.

Decoding the dynamic physiological responses of the human central and autonomic nervous systems to sustained cognitive loads is essential for occupational health and neuroergonomics. This study investigates the multidimensional neurophysiological mechanisms of fatigue during complex human-machine interactions, utilizing a simulated agricultural operation paradigm. We developed a multimodal "eye-brain-heart" physio-logging framework, synchronously recording 21-channel EEG, ECG, and pupillometry in 12 experienced subjects over a continuous 5-hour protocol.
