Fragmented rRNAs and lineage-specific RNA-binding proteins define myzozoan mitoribosomes.

Myzozoa, a clade of alveolate protists including Apicomplexa, Chromerida, Perkinsozoa, and dinoflagellates, possesses the most reduced mitochondrial genomes among eukaryotes. Apicomplexan parasites, such as Toxoplasma gondii and Plasmodium spp., retain mitochondrial genomes encoding only three proteins and highly fragmented rRNAs. Despite this reduction, recent structural studies show that T. gondii mitoribosome incorporates lineage-specific RNA-binding proteins as integral components to maintain a functional complex composed of over 50 rRNA fragments. The conservation of rRNA fragmentation an

A Bifunctional Antibody Conjugate for Marking the Location of DNA Binding Proteins on DNA Fibers.

The cellular response to DNA damage can be marked by the appearance of immunofluorescent foci of DNA Damage Response (DDR) proteins. These serve as surrogates for DNA damage and are frequently interpreted as denoting specific lesions. For example, double-strand breaks (DSBs) are potent inducers of the DDR, whose best-known factor is the phosphorylated histone variant H2AX (γ-H2AX). The association with DSBs is so well established that the reverse interpretation that γ-H2AX invariably implies DSBs is routine. However, this conclusion is inferential and has been challenged. The resolution of thi

Identification of RNA-binding protein RBMS3 as a potential biomarker for immunotherapy in bladder cancer.

RNA-binding protein (RBP) plays pivotal roles in the malignant progression of cancer by regulating gene expression. In this paper, we aimed to develop RBP-based prognostic signature and identify critical hub RBPs in bladder cancer (BLCA). Firstly, a risk model based on differentially expressed RBP gens (DERBPs) between normal and tumor tissues was successfully established, which can predict the tumor stromal score and drug sensitivity. Then two another RBP risk models based on miRNA-correlated RBPs or lncRNA-correlated RBPs were also established, and RBMS3 was identified as the overlapping gen

Fibrinogen-binding M-related proteins facilitate the recruitment of plasminogen by Streptococcus pyogenes.

Group A Streptococcus (GAS) M-related proteins (Mrp) are dimeric α-helical coiled-coil cell-wall-attached proteins. During infection, Mrp recruit human fibrinogen (Fg) to the bacterial surface, enhancing phagocytosis resistance and promoting growth in human blood. However, Mrp exhibit a high degree of sequence diversity, clustering into four evolutionarily distinct groups. It is currently unknown whether this diversity affects the host-pathogen interactions mediated by Mrp. In this study, nine Mrp sequences from the four major evolutionary groups were selected to examine the effect of sequence

Evaluation of Heparin-Binding Protein as a novel biomarker for the detection of periprosthetic joint infection.

BACKGROUND: Heparin-Binding Protein (HBP), released during neutrophil activation and degranulation, functions in antimicrobial defense, vascular integrity regulation, and immune signal amplification. As a key effector of the innate immune system, HBP is rapidly released in response to infectious stimuli and plays a pivotal role in the pathogenesis of infectious diseases. This study aimed to evaluate the diagnostic value of HBP in periprosthetic joint infection (PJI) and compare its performance with commonly used inflammatory biomarkers. METHODS: In this prospective study, 156 patients undergo

Proteomic Identification and Functional Analysis of Babesia microti Reveals Heparin-Binding Proteins.

Glycosaminoglycan (GAG) molecules on the surface of red blood cells play an important regulatory role in the invasion of merozoites of apicomplexan protozoa. Heparan sulfate, a type of GAG molecule, has been identified as an important receptor facilitating the invasion of red blood cells by these parasites. Proteins in the parasite that exhibit strong affinity for heparin may play a pivotal role in this invasion process. This study aims to use proteomics to identify Babesia microti proteins with high binding affinity to heparin. Bioinformatics was utilized to analyze the subcellular localizati

Calcium-Binding Proteins and Melatonin Receptors in the Central Auditory System of Aged Rats.

INTRODUCTION: Age-related hearing loss (ARHL) is a multifactorial disorder due to impairment in the peripheral or central auditory pathway (CAP). CAP impairment refers to difficulties in speech understanding, particularly in the presence of background noise, problems in discrimination, and integrating and separating binaural auditory information. Changes in CAP mechanisms, such as neurotransmitters or ion movement, may correlate with this impairment. The regulation of calcium (Ca2+) concentrations by Ca2+-binding proteins (CaBPs) is critical for synaptic function and plasticity, and changes ar

Binding to thrive: Decoding Atopobium spp. interactions with host proteins and immune cells.

OBJECTIVES: Gram-positive anaerobic cocci are opportunistic pathogens that exploit immune suppression or tissue injury to establish infections. Atopobium spp. are clinically relevant, being linked to bacterial vaginosis, abdominal wounds, pelvic abscesses, and dental infections. While clinical reports describe the involvement in detrimental host conditions, in-depth knowledge is missing to understand how these bacteria acquire essential nutrients to evade immune responses and directly interact with the human host. METHODS: Here we analyzed the growth of three blood-isolated Atopobium strains

Affinity of cefditoren for penicillin-binding proteins in bacteria and its relationship with antibiotic sensitivity.

Penicillin-binding proteins (PBPs) are the targets of β-lactam antibiotics; however, changes in the affinity of PBPs for beta-lactam antibiotics often affect the susceptibility of bacteria to antibiotics. The purpose of this study was to elucidate the mechanism by which cefditoren, an oral third-generation cephalosporin, binds PBPs. The minimal inhibitory concentration (MIC), bactericidal curves, and inhibition zone comparisons were assessed to evaluate the antibacterial activity of cefditoren. PBP1A and PBP2X proteins from Streptococcus pneumoniae were purified, and their ability to bind to c

Investigation of choline-binding protein of CbpD in the pathogenesis of Streptococcus suis type 2.

Streptococcus suis serotype 2 (S. suis type 2, SS2) is one of the zoonotic pathogens known to induce meningitis, septicemia, and arthritis in both pigs and humans, resulting in public health concerns. CbpD, also termed CrfP, is one of the choline-binding proteins (CBPs) that was found as a murein hydrolase in SS2 and plays crucial roles in natural genetic transformation under the control of ComRS-ComX regulatory system by a previous study. Nonetheless, the possible functions of CbpD in virulence and pathogenesis in SS2 remain unclear. In this study, a cbpD gene mutant (ΔcbpD) with its compleme

Differential Role of the RAC1-Binding Proteins FAM49b (CYRI-B) and CYFIP1 in Platelets.

Platelet function at vascular injury sites is tightly regulated through the actin cytoskeleton. The Wiskott-Aldrich syndrome protein-family verprolin-homologous protein (WAVE)-regulatory complex (WRC) activates lamellipodia formation via ARP2/3, initiated by GTP-bound RAC1 interacting with the WRC subunit CYFIP1. The protein FAM49b (Family of Unknown Function 49b), also known as CYRI-B (CYFIP-Related RAC Interactor B), has been found to interact with activated RAC1, leading to the negative regulation of the WRC in mammalian cells. To investigate the role of FAM49b in platelet function, we stud

Role of WAVE3 as an actin binding protein in the pathology of triple negative breast cancer.

Breast cancer, a prevalent global health concern, has sparked extensive research efforts, particularly focusing on triple negative breast cancer (TNBC), a subtype lacking estrogen receptor (ER), progesterone receptor, and epidermal growth factor receptor. TNBC's aggressive nature and resistance to hormone-based therapies heightens the risk of tumor progression and recurrence. Actin-binding proteins, specifically WAVE3 from the Wiskott-Aldrich syndrome protein (WASP) family, have emerged as major drivers in understanding TNBC biology. This review delves into the intricate molecular makeup of TN

Vaccine-induced neutralizing antibodies bind to the H protein of a historical measles virus.

Measles is a highly contagious airborne viral disease. It can lead to serious complications and death and is preventable by vaccination. The live-attenuated measles vaccine (LAMV) derived from a measles virus (MV) isolated in 1954 has been in use globally for six decades and protects effectively by providing a durable humoral and cell-mediated immunity. Our study addresses the temporal stability of epitopes on the viral surface glycoprotein hemagglutinin (H) which is the major target of MV-neutralizing antibodies. We investigated the binding of seven vaccine-induced MV-H-specific monoclonal an

The RNA-binding protein Adad1 is necessary for germ cell maintenance and meiosis in zebrafish.

The double stranded RNA binding protein Adad1 (adenosine deaminase domain containing 1) is a member of the adenosine deaminase acting on RNAs (Adar) protein family with germ cell-specific expression. In mice, Adad1 is necessary for sperm differentiation, however its function outside of mammals has not been investigated. Here, through an N-ethyl-N-nitrosourea (ENU) based forward genetic screen, we identified an adad1 mutant zebrafish line that develops as sterile males. Further histological examination revealed complete lack of germ cells in adult mutant fish, however germ cells populated the g

Redox processes in Cu-binding proteins: the "in-between" states in intrinsically disordered peptides.

We report on a concept that some of us first described a decade ago for pure electron transfer [V. Balland, C. Hureau and J.-M. Savéant, Proc. Natl. Acad. Sci. U. S. A., 2010, 107, 17113]. In the present viewpoint, based on more recent results, we refine and extend this "in-between state" concept to explain the formation of reactive oxygen species by copper ions bound to the amyloid-β (Aβ) peptide involved in Alzheimer's disease. In such intrinsically disordered peptides, the Cu coordination is versatile due to the lack of stable folding and the presence of multiple possible binding anchors. H

The Pneumococcal Protein SufC Binds to Host Plasminogen and Promotes Its Conversion into Plasmin.

Streptococcus pneumoniae causes otitis media, sinusitis, and serious diseases such as pneumonia and bacteremia. However, the in vivo dynamics of S. pneumoniae infections and disease severity are not fully understood. In this study, we investigated pneumococcal proteins detected in the bronchoalveolar lavage fluid of an S. pneumoniae-infected mouse, which were assumed to be expressed during infection. Analysis of three proteins with unknown infection-related functions revealed that recombinant Fe-S cluster assembly ATP-binding protein (SufC) binds to the host plasminogen and promotes its conver

Zinc finger proteins and ATP-binding cassette transporter-dependent multidrug resistance.

BACKGROUND: Multidrug resistance (MDR) remains a significant challenge in cancer treatment, leading to poor clinical outcomes. Dysregulation of ATP-binding cassette (ABC) transporters has been identified as a key contributor to MDR. Zinc finger proteins (ZNPs) are key regulators of transcription and have emerged as potential contributors to cancer drug resistance. Bridging the knowledge gap between ZNPs and MDR is essential to understand a source of heterogeneity in cancer treatment. This review sought to elucidate how different ZNPs modulate the transcriptional regulation of ABC genes, contri
