mTOR inhibitor RAD001 (Everolimus) enhances the effects of imatinib in chronic myeloid leukemia by raising the nuclear expression of c-ABL protein.

Constitutive tyrosine kinase (TK) activity of p210 BCR-ABL fusion protein of chronic myeloid leukemia (CML) usurps physiological functions of normal p145 c-ABL protein. Accordingly, its inhibition by imatinib mesylate (IM) lets p145 c-ABL translocate into the nuclear compartment, which drives cell growth arrest and apoptotic death. Here we show that IM and the mammalian target of rapamycin (mTOR) inhibitor RAD001 (Everolimus) have additive effects on BCR-ABL-expressing cells. Those effects are at least partly conditional upon the enhanced nuclear accumulation of p145 c-ABL through events encom

Prion protein glycosylation.

The transmissible spongiform encephalopathies (TSE), or prion diseases are a group of transmissible neurodegenerative disorders of humans and animals. Although the infectious agent (the 'prion') has not yet been formally defined at the molecular level, much evidence exists to suggest that the major or sole component is an abnormal isoform of the host encoded prion protein (PrP). Different strains or isolates of the infectious agent exist, which exhibit characteristic disease phenotypes when transmitted to susceptible animals. In the absence of a nucleic acid genome it has been hard to accommod

Dual Roles of p53-Related Protein Kinase in Cytoskeletal Remodeling and Translational Control.

P53-related protein kinase (PRPK; TP53RK) is an evolutionarily conserved atypical kinase whose biology reveals an unexpected integration of translational control, cytoskeletal regulation, and cell-type-specific stress responses. As a core component of the TCTC complex (Threonyl-Carbamoyl Transferase Complex), PRPK participates in the universally conserved synthesis of the tRNA modification t6A, an essential process for translational fidelity, proteostasis, and cellular viability. Structural and biochemical studies from yeast to humans highlight a deeply conserved ATPase-driven mechanism in whi

A generalizable framework for genetic code expansion in bacterial systems beyond E. coli.

Genetic code expansion (GCE) enables the site-specific incorporation of noncanonical amino acids into proteins, providing a powerful platform to investigate and engineer biological functions with molecular precision. While GCE has facilitated transformative studies in Escherichia coli, its utility in bacteria other than E. coli has been hindered by the lack of universal, high-efficiency methods for its implementation and optimization. To address this limitation, we introduce a modular, extensible platform for the generalizable implementation and optimization of GCE in phylogenetically diverse

Targeting geranylgeranylation reduces adrenal gland tumor burden in a murine model of prostate cancer metastasis.

The isoprenoid biosynthetic pathway (IBP) is critical for providing substrates for the post-translational modification of proteins key in regulating malignant cell properties, including proliferation, invasion, and migration. Inhibitors of the IBP, including statins and nitrogenous bisphosphonates, are used clinically for the treatment of hypercholesterolemia and bone disease respectively. The statins work predominantly in the liver, while the nitrogenous bisphosphonates are highly sequestered to bone. Inhibition of the entire IBP is limited by organ specificity and side effects resulting from

Methods for study of protein dynamics and protein-protein interaction in protein-ubiquitination by electron paramagnetic resonance spectroscopy.

Ubiquitination is a post-translation modification whereby the C-terminal end of ubiquitin (Ub) is covalently attached to the amino group of a lysine in a target protein. Additional ubiquitin groups are added using Ub-Ub linkages to form a polyubiquitin chain. A 26S protease complex specifically binds polyubiquitinated proteins and degrades them in an ATP-dependent manner. The target lysine in the substrate protein resides in a domain that is recognized by the ubiquitination machinery in a temporally and spatially controlled manner. The accessibility and the molecular dynamics of the target dom

Comprehensive genome-wide proteomic analysis of human placental tissue for the Chromosome-Centric Human Proteome Project.

As a starting point of the Chromosome-Centric Human Proteome Project (C-HPP), we established strategies of genome-wide proteomic analysis, including protein identification, quantitation of disease-specific proteins, and assessment of post-translational modifications, using paired human placental tissues from healthy and preeclampsia patients. This analysis resulted in identification of 4239 unique proteins with high confidence (two or more unique peptides with a false discovery rate less than 1%), covering 21% of approximately 20, 059 (Ensembl v69, Oct 2012) human proteins, among which 28 prot

In silico characterization of the OSBPL6 gene and its potential role in ascites syndrome in broiler chickens.

Ascites syndrome is a multifactorial metabolic disorder in broiler chickens, characterized by pulmonary hypertension, fluid accumulation, and high mortality, leading to significant economic losses. The oxysterol-binding protein-like 6 gene (OSBPL6) has been associated with ascites susceptibility, but its molecular role remains unclear. Therefore, this study was aimed to perform a comprehensive in-silico analysis of chicken OSBPL6, including analyses of physicochemical properties, structural features, post-translational modifications, protein-protein interactions, promoter motifs, and tissue-sp

Post-translational modification of LipL32 during Leptospira interrogans infection.

BACKGROUND: Leptospirosis, a re-emerging disease of global importance caused by pathogenic Leptospira spp., is considered the world's most widespread zoonotic disease. Rats serve as asymptomatic carriers of pathogenic Leptospira and are critical for disease spread. In such reservoir hosts, leptospires colonize the kidney, are shed in the urine, persist in fresh water and gain access to a new mammalian host through breaches in the skin. METHODOLOGY/PRINCIPAL FINDINGS: Previous studies have provided evidence for post-translational modification (PTM) of leptospiral proteins. In the current study

The testis-specific E3 ubiquitin ligase RNF133 is required for fecundity in mice.

BACKGROUND: Ubiquitination is a post-translational modification required for a number of physiological functions regulating protein homeostasis, such as protein degradation. The endoplasmic reticulum (ER) quality control system recognizes and degrades proteins no longer needed in the ER through the ubiquitin-proteasome pathway. E2 and E3 enzymes containing a transmembrane domain have been shown to function in ER quality control. The ER transmembrane protein UBE2J1 is a E2 ubiquitin-conjugating enzyme reported to be essential for spermiogenesis at the elongating spermatid stage. Spermatids from

Porphyromonas gingivalis infection induces lysine lactylation reprogramming in human umbilical vein endothelial cells.

INTRODUCTION: Porphyromonas gingivalis (Pg), a keystone periodontal pathogen, is a known risk factor for atherosclerosis and cardiovascular disease. Lysine lactylation (Kla) is an emerging post-translational modification (PTM) that bridges cellular metabolism and epigenetic regulation. However, the involvement of Kla in bacteria-induced endothelial dysfunction remains unexplored. This study aims to characterize the global lactylation landscape in human umbilical vein endothelial cells (HUVECs) following Pg infection. METHODS: HUVECs were infected with Pg, and their lactylome was analyzed usin

RNA-centered regulation in SFTSV infection: mechanisms, host interactions, and translational implications.

Severe fever with thrombocytopenia syndrome virus (SFTSV) is an emerging tick-borne pathogen that causes substantial morbidity and high case fatality in severe cases, representing a major public health concern in East Asia. Although protein-mediated mechanisms of SFTSV replication and immune evasion have been extensively studied, accumulating evidence indicates that viral infection is also influenced by RNA-centered regulatory processes. These processes include viral RNA transcription and replication, epitranscriptomic modification, host RNA-binding proteins, RNA interference-related pathways,

2-D reference map of Bacillus anthracis vaccine strain A16R proteins.

Bacillus anthracis has always been an important pathogen because it can cause lethal inhalational anthrax, and may be used as a bioweapon or by bioterrorists. In this study, a 2-DE reference map and database of B. anthracis A16R was constructed. In total, 534 spots were processed, and 406 spots representing 299 proteins were identified. Gel-estimated pIs and molecular masses mostly matched well with their theoretical predictions, but some discrepancies also existed. Spot and protein corresponding analysis revealed that post-translational modifications might be common in B. anthracis. Through t

Prognostic biomarkers and regulatory mechanisms associated with lysine β-hydroxybutyrylation modification in prostate cancer.

BACKGROUND: Prostate cancer (PCa) is a highly prevalent malignant tumor in males. Lysine β-hydroxybutyrylation (Kbhb), an emerging post-translational modification, plays a critical role in tumorigenic processes. However, its functional mechanism in PCa remains elusive. This study aims to identify Kbhb-related prognostic genes in PCa and provide novel insights for its diagnosis, prognosis and treatment. METHODS: Candidate genes were obtained through differential expression analysis and intersection analysis. Univariate Cox proportional hazards model and least absolute shrinkage and selection o

O-GlcNAcylation regulates the DNA damage repair function of MND1 in breast cancer.

INTRODUCTION: As a meiosis-specific gene, Meiotic Nuclear Divisions 1 (MND1) plays a crucial role in homologous recombination and is generally silenced in somatic cells. The expression pattern and functional significance of MND1 in breast cancer (BRCA) have not been elucidated. This study systematically investigated the expression, subcellular localization, and pathological function of MND1 in BRCA, with particular focus on the post-translational regulation of MND1 through O-GlcNAcylation (O-GlcNAc) and its underlying molecular mechanisms. METHODS: Immunohistochemical analysis and subcellular

Triose-phosphate isomerase deficiency is associated with a dysregulation of synaptic vesicle recycling in Drosophila melanogaster.

INTRODUCTION: Numerous neurodegenerative diseases are associated with neuronal dysfunction caused by increased redox stress, often linked to aberrant production of redox-active molecules such as nitric oxide (NO) or oxygen free radicals. One such protein affected by redox-mediated changes is the glycolytic enzyme triose-phosphate isomerase (TPI), which has been shown to undergo 3-nitrotyrosination (a NO-mediated post-translational modification) rendering it inactive. The resulting neuronal changes caused by this modification are not well understood. However, associated glycation-induced cytoto

Characterization of Novel PHEX Variants in X-linked Hypophosphatemic Rickets and Genotype-PHEX Activity Correlation.

BACKGROUND: X-linked hypophosphatemia (XLHR) is the most common genetic form of hypophosphatemic rickets (HR), which is caused by phosphate regulating endopeptidase homolog X-linked (PHEX) gene mutation. At present, the genotype-phenotype relationship of XLHR and the pathogenic role of PHEX are not fully understood. METHODS: In this study, we summarized clinical features in a new cohort of 49 HR patients and detected 16 novel PHEX and 5 novel non-PHEX variants. Subsequently, we studied the pathogenesis of new variants by protein expression, glycosylation analysis, subcellular localization, an

Protein O-GlcNAcylation alleviates small intestinal injury induced by ischemia-reperfusion and oxygen-glucose deprivation.

Protein O-GlcNAcylation is a dynamic post-translational protein modification that regulates fundamental cellular functions in both normal physiology and diseases. The levels of protein O-GlcNAcylation are determined by flux of the hexosamine biosynthetic pathway (HBP), which is a branch of glycolysis, and are directly controlled by a pair of enzymes: O-GlcNAc transferase (OGT) and O-GlcNAcase (OGA). An increase in protein O-GlcNAcylation has been shown to have protective effects on ischemia-related insults in the heart and brain. To determine whether O-GlcNAcylation plays a beneficial role in

Revelation of Proteomic Indicators for Colorectal Cancer in Initial Stages of Development.

Background: Colorectal cancer (CRC) at a current clinical level is still hardly diagnosed, especially with regard to nascent tumors, which are typically asymptotic. Searching for reliable biomarkers of early diagnosis is an extremely essential task. Identification of specific post-translational modifications (PTM) may also significantly improve net benefits and tailor the process of CRC recognition. We examined depleted plasma samples obtained from 41 healthy volunteers and 28 patients with CRC at different stages to conduct comparative proteome-scaled analysis. The main goal of the study was
