Fatal familial insomnia, a prion disease with a mutation at codon 178 of the prion protein gene.

BACKGROUND: We previously described two members of a family affected by an apparently genetically determined fatal disease characterized clinically by progressive insomnia, dysautonomia, and motor signs and characterized pathologically by severe atrophy of the anterior ventral and mediodorsal thalamic nuclei. Five other family members who died of this disease, which we termed "fatal familial insomnia," had broader neuropathologic changes suggesting that fatal familial insomnia could be a prion disease. METHODS: We used antibodies to prion protein (PrP) to perform dot and Western blot analyses…

Early development of the retinal line of decussation in normal and albino ferrets.

This work describes the retinal origin of the crossed and uncrossed projections in newborn, 9-day-old and adult normally pigmented and albino ferrets. Horseradish peroxidase (HRP) was injected unilaterally into the thalamic and midbrain visual centers of ferrets to label retinal ganglion cells retrogradely. In normally pigmented adults, the retinal line of decussation was sharp and passed through the area centralis. Ganglion cells with uncrossed axons occupied the entire temporal retina. In albino adults, ganglion cells with uncrossed axons were distributed in the periphery of the temporal ret

Microglial cGAS-STING-TBK1 activation in paraventricular thalamus mediates sleep and emotional disturbances in lupus mice.

Sleep and emotional disturbances are prevalent in systemic lupus erythematosus (SLE) patients, yet the neuroinflammatory mechanisms remain unclear. Here, we investigated this issue using pristane-induced lupus (PIL) mice. We firstly confirmed that PIL mice exhibited progressive fragmentation of NREM sleep and decreased cumulative sleep time, correlating with blood-brain barrier (BBB) leakage and IgG deposition in the paraventricular thalamus (PVT). Concurrently, PVT neurons showed aberrant excitatory activity, including a high level of cFos expression, decreased amplitude and increased rate of

"Hyperglutamatergic cortico-striato-thalamo-cortical circuit" breaker drugs alleviate tics in a transgenic circuit model of Tourette׳s syndrome.

The brain circuits underlying tics in Tourette׳s syndrome (TS) are unknown but thought to involve cortico/amygdalo-striato-thalamo-cortical (CSTC) loop hyperactivity. We previously engineered a transgenic mouse "circuit model" of TS by expressing an artificial neuropotentiating transgene (encoding the cAMP-elevating, intracellular A1 subunit of cholera toxin) within a small population of dopamine D1 receptor-expressing somatosensory cortical and limbic neurons that hyperactivate cortico/amygdalostriatal glutamatergic output circuits thought to be hyperactive in TS and comorbid obsessive-compul

Comparative anatomy of marmoset and mouse cortex from genomic expression.

Advances in mouse neural circuit genetics, brain atlases, and behavioral assays provide a powerful system for modeling the genetic basis of cognition and psychiatric disease. However, a critical limitation of this approach is how to achieve concordance of mouse neurobiology with the ultimate goal of understanding the human brain. Previously, the common marmoset has shown promise as a genetic model system toward the linking of mouse and human studies. However, the advent of marmoset transgenic approaches will require an understanding of developmental principles in marmoset compared to mouse. In

Deep brain stimulation in the subthalamic area is more effective than nucleus ventralis intermedius stimulation for bilateral intention tremor.

BACKGROUND: The ventro-lateral thalamus is the stereotactic target of choice for severe intention tremor. Nevertheless, the optimal target area has remained controversial, and targeting of the subthalamic area has been suggested to be superior. PATIENTS AND METHODS: Eleven patients with disabling intention tremor of different etiology (essential tremor (n = 8), multiple sclerosis (n = 2) and one with, spinocerebellar ataxia) were implanted bilaterally with DBS electrodes targeted to the ventro-lateral thalamus using micro-recording and micro-stimulation. Among five tracks explored in parallel

Membrane bistability in thalamic reticular neurons during spindle oscillations.

The thalamic reticular (RE) nucleus is a major source of inhibition in the thalamus. It plays a crucial role in regulating the excitability of thalamocortical networks and in generating some sleep rhythms. Current-clamp intracellular recordings of RE neurons in cats under barbiturate anesthesia revealed the presence of membrane bistability in approximately 20% of neurons. Bistability consisted of two alternate membrane potentials, separated by approximately 17-20 mV. While non-bistable (common) RE neurons fired rhythmic spike-bursts during spindles, bistable RE neurons fired tonically, with bu

Cortical stimulation for central neuropathic pain: 3-D surface MRI for easy determination of the motor cortex.

Motor cortex electric stimulation has been reported to be effective for the treatment of central post-stroke pain and trigeminal neuropathic pain. Five patients with pain due to injury of the trigeminal nerve and with abnormalities of facial sensibility, as well as two patients suffering of a post-stroke thalamic pain, were subjected to stimulation applied epidurally on the motor cortex. Quadripolar electrodes were implanted under local anaesthesia and the precise location of the motor cortex was determined on three-dimensional surface MRI the day prior to surgery. In our experience, correct t

Imaging of vestibular function and disorders and its clinical relevance.

PURPOSE OF REVIEW: To discuss recent advances in imaging of the structural organization and functional connectivity of central vestibular disorders with various MRI techniques. Vestibular paroxysmia, in particular the characteristics of neurovascular cross-compression of the eighth nerve, serves as an example of the peripheral vestibular disorders. MAIN FINDINGS: The bilateral vestibular system is intricately connected with other sensory systems and is hierarchically organized within sensory, motor, cognitive, emotional, and memory networks. This has been demonstrated by fMRI using galvanic v

Brain Damage and Gene Expression Across Hereditary Spastic Paraplegia Subtypes.

BACKGROUND: Spinal cord has been considered the main target of damage in hereditary spastic paraplegias (HSPs), but mounting evidence indicates that the brain is also affected. Despite this, little is known about the brain signature of HSPs, in particular regarding stratification for specific genetic subtypes. OBJECTIVE: We aimed to characterize cerebral and cerebellar damage in five HSP subtypes (9 SPG3A, 27 SPG4, 10 SPG7, 9 SPG8, and 29 SPG11) and to uncover the clinical and gene expression correlates. METHODS: We obtained high-resolution brain T1 and diffusion tensor image (DTI) datasets

Interhemispheric dominance switching in a neural network model for birdsong.

Male zebra finches produce a sequence-invariant set of syllables, separated by short inspiratory gaps. These songs are learned from an adult tutor and maintained throughout life, making them a tractable model system for learned, sequentially ordered behaviors, particularly speech production. Moreover, much is known about the cortical, thalamic, and brain stem areas involved in producing this behavior, with the premotor cortical nucleus HVC (proper name) being of primary importance. In a previous study, our group developed a behavioral neural network model for birdsong constrained by the struct

Deep brain stimulation for Tourette syndrome: a single-center series.

OBJECTIVE Tourette syndrome (TS) is a complex neuropsychiatric disorder characterized by multiple motor and phonic tics. While pharmacological and behavioral therapy can be effective in most patients, a subset of patients remains refractory to treatment. Increasing clinical evidence from multiple centers suggests that deep brain stimulation (DBS) of the medial thalamus can be effective in many cases of refractory TS. METHODS The authors retrospectively reviewed outcomes in 13 patients with refractory TS who underwent medial thalamic DBS performed by their team over a 7-year period. Patients we

Altered dynamics in the circadian oscillation of clock genes in dermal fibroblasts of patients suffering from idiopathic hypersomnia.

From single cell organisms to the most complex life forms, the 24-hour circadian rhythm is important for numerous aspects of physiology and behavior such as daily periodic fluctuations in body temperature and sleep-wake cycles. Influenced by environmental cues - mainly by light input -, the central pacemaker in the thalamic suprachiasmatic nuclei (SCN) controls and regulates the internal clock mechanisms which are present in peripheral tissues. In order to correlate modifications in the molecular mechanisms of circadian rhythm with the pathophysiology of idiopathic hypersomnia, this study aime
