Loss-of-function in testis-specific serine/threonine protein kinase triggers male infertility in an invasive moth.

Genetic biocontrol technologies present promising and eco-friendly strategies for the management of pest and insect-transmitted diseases. Although considerable advancements achieve in gene drive applications targeting mosquitoes, endeavors to combat agricultural pests have been somewhat restricted. Here, we identify that the testis-specific serine/threonine kinases (TSSKs) family is uniquely expressed in the testes of Cydia pomonella, a prominent global invasive species. We further generated male moths with disrupted the expression of TSSKs and those with TSSKs disrupted using RNA interference

Pervasive Endosymbiont Arsenophonus Plays a Key Role in the Transmission of Cotton Leaf Curl Virus Vectored by Asia II-1 Genetic Group of Bemisia tabaci.

Insects often coevolved with their mutualistic partners such as gut endosymbionts, which play a key in the physiology of host. Studies on such interactions between Bemisia tabaci and its primary and secondary endosymbionts have gained importance due to their indispensable roles in the biology of this insect. Present study reports the predominance of two secondary endosymbionts, Arsenophonus and Cardinium in the Asia II-1 genetic group of whitefly and elucidates their role in the transmission of its vectored Cotton leaf curl virus. Selective elimination of endosymbionts was optimized using seri

A rhamnose-binding lectin from Rhodnius prolixus and the impact of its silencing on gut bacterial microbiota and Trypanosoma cruzi.

Lectins are ubiquitous proteins involved in the immune defenses of different organisms and mainly responsible for non-self-recognition and agglutination reactions. This work describes molecular and biological characterization of a rhamnose-binding lectin (RBL) from Rhodnius prolixus, which possesses a 21 amino acid signal peptide and a mature protein of 34.6 kDa. The in-silico analysis of the primary and secondary structures of RpLec revealed a lectin domain fully conserved among previous insects studied. The three-dimensional homology model of RpLec was similar to other RBL-lectins. Docking p

Population genetics of New World screwworm from the Caribbean: insights from microsatellite data.

Diseases affecting livestock can have a significant impact on animal productivity and on trade of live animals, meat and other animal products, which, consequently, affects the overall process of economic development. The New World screwworm, Cochliomyia hominivorax (Coquerel) (Diptera: Calliphoridae), is an important parasitic insect pest in Neotropical regions. This species has been successfully eradicated from North and most of Central America by the sterile insect technique, but continues to affect the development of the livestock sector in most Caribbean economies. Here, we provide some i

Dirofilaria immitis encodes Di-nhr-7, a putative orthologue of the Drosophila ecdysone-regulated E78 gene.

Filarial parasites are responsible for several serious human diseases with symptoms such as lymphoedema, elephantiasis, and blindness. An understanding of how these parasites pass through developmental checkpoints may elucidate the general mechanisms of these illnesses and suggest potential targets for intervention. A useful model system for the study of human filariasis is the related nematode Dirofilaria immitis, the causative agent of dog heartworm disease. In D. immitis, molting from the third to the fourth larval stage can be induced in vitro by the insect hormone 20-OH ecdysone, suggesti

Prevalence, knowledge, attitudes, and practices regarding Chagas disease in Guanare, Venezuela: a cross-sectional study.

BACKGROUND: Chagas disease (CD) is endemic in 21 Latin American countries, placing approximately 75 million people at risk of infection. In Venezuela, CD has been recognized since 1919, with seroprevalence estimates reaching up to 45% between 1958 and 1968. However, current data on the epidemiology of CD in Venezuela are limited. METHODS: We conducted a cross-sectional study in September 2023 in Guanare municipality, located in northwestern Portuguesa state. Seroprevalence was determined by detecting anti-Trypanosoma cruzi IgG antibodies and assessing specific-IgG avidity enzyme-linked immuno

Effect of egg irradiation on development and sterility of wild-type and Wolbachia trans-infected Aedes aegypti mosquito vectors.

Sterile Insect Technique (SIT), Incompatible Insect Technique (IIT) or a combination of the two has become alternative promising vector control approaches. In order to apply these approaches, the targeted mosquitoes need to be sterilized and released. So far, the irradiation of mosquitoes has been conducted at the pupae or adult stages. In this study, we investigated the possibility of applying X-ray irradiation at the egg stage and also assessed the effect on the development and sterility of both wild-type and Wolbachia trans-infected Aedes aegypti mosquito vectors. The eggs of both wild-type

Spatial correlation network characteristics and influencing factors of urban ecological vulnerability in the Yellow River Basin, China.

Plant invasion poses a serious threat to biodiversity and ecosystem functions, while habitat heterogeneity significantly affects the invasion trend along latitudinal gradients. We reviewed the research progress on plant invasion and its biological interaction patterns along latitudes over the past decade. The phenotypic plasticity and ecolo-gical adaptability of many invasive plants increase with increasing latitude, thereby exacerbating the homogenization of native plant communities in high-latitude regions. The occurrence of natural enemy insects, insect feeding rates, soil pathogenic fungal

Detection of arsenic, mercury, cadmium and lead compounds in Calliphora vicina (Diptera: Calliphoridae) larvae and their potential impact on larval development.

Heavy metal exposure represents a substantial health risk to humans and other animals, in some cases leading to death. However, detecting these metals in decaying corpses can pose a considerable challenge. In such situations, the eventual presence of necrophagous insects feeding on the remains may serve as a valuable tool for detecting and measuring these toxic substances. Nonetheless, heavy metals might affect insect development and survival rates. This study explored the effects of increasing concentrations of arsenic, mercury, cadmium and lead compounds on the survival and development rate

Caste- and environment-associated differential expression of olfactory receptors in pest ants.

As human society continues to grow and evolve, so does the need for effective pest management strategies. Olfactory-mediated control methods, such as attractant and repellent compounds, are a proposed strategy for mitigating the damaging effects of some insect pests, most notably ants, that rely on olfaction for communication. To develop such compounds, it is first important to comprehensively understand the target species' olfactory transcriptome in order to guide future targeted functional characterization of relevant olfactory proteins. Here, we perform bulk RNA-seq analysis of antennae fro

Sec61s and Sec62/Sec63 Genes Are Essential for Survival by Regulating the Gut and Cuticle Development in Locusta migratoria.

In eukaryotes, a lot of proteins are transported across the endoplasmic reticulum by the heterotrimeric Sec61 channel. And post-translational transport needs another Sec62/Sec63 complex. However, functions of these genes are poorly explored in insects. In this study, we first identified five Sec genes, named Sec61α, Sec61β, Sec61γ, Sec62 and Sec63, in Locusta migratoria. Quantitative reverse-transcription polymerase chain reaction (RT-qPCR) analysis showed that these five genes were expressed in muti-tissues, including wing pad, leg, foregut, midgut, gastric cecum, hindgut, and highly expresse

Diversity of mosquito natural enemies and their feeding efficacy on Aedes vectors.

BACKGROUND OBJECTIVES: Mosquito-bome diseases are a major health issue among communities in Asia, and dengue has become a serious public health concern in Sri Lanka with varying magnitude since 1960. With the realization of the failures and limitations of current vector control strategies, the authorities are moving towards simple, low- cost, and eco-friendly vector management methods. Thus, the present study was devised to evaluate the potential densities of mosquito natural enemies in natural water habitats in Galle district, Sri Lanka, and to investigate the feeding efficacy of prominent mo

Knockout of juvenile hormone receptor, Methoprene-tolerant, induces black larval phenotype in the yellow fever mosquito, Aedes aegypti.

The yellow fever mosquito, Aedes aegypti, vectors human pathogens. Juvenile hormones (JH) control almost every aspect of an insect's life, and JH analogs are currently used to control mosquito larvae. Since RNA interference does not work efficiently during the larval stages of this insect, JH regulation of larval development and mode of action of JH analogs are not well studied. To overcome this limitation, we used a multiple single guide RNA-based CRISPR/Cas9 genome-editing method to knockout the methoprene-tolerant (Met) gene coding for a JH receptor. The Met knockout larvae exhibited a blac

Optimization of the sterilizing doses and overflooding ratios for the South American fruit fly.

The Sterile Insect Technique (SIT) is an autocidal control method that relies on inundative releases of sterilized mass-reared insects. This technology has been used in several area-wide programmes for the suppression/eradication of fruit fly populations. Choosing the optimum sterilizing dose and the sterile release density is an essential step of the SIT. Considering unsolved issues related to the application of this technique against Anastrepha fraterculus (Wiedemann), this study aimed to define accurately the central target dose for both sexes of this species and to verify the induction of

Tyrosine Detoxification Is an Essential Trait in the Life History of Blood-Feeding Arthropods.

Blood-feeding arthropods are vectors of infectious diseases such as dengue, Zika, Chagas disease, and malaria [1], and vector control is essential to limiting disease spread. Because these arthropods ingest very large amounts of blood, a protein-rich meal, huge amounts of amino acids are produced during digestion. Previous work on Rhodnius prolixus, a vector of Chagas disease, showed that, among all amino acids, only tyrosine degradation enzymes were overexpressed in the midgut compared to other tissues [2]. Here we demonstrate that tyrosine detoxification is an essential trait in the life his

Effects of Transgenic cry1Ca Rice on the Development of Xenopus laevis.

In fields of genetically modified, insect-resistant rice expressing Bacillus thuringiensis (Bt) proteins, frogs are exposed to Bt Cry proteins by consuming both target and non-target insects, and through their highly permeable skin. In the present study, we assessed the potential risk posed by transgenic cry1Ca rice (T1C-19) on the development of a frog species by adding purified Cry1Ca protein or T1C-19 rice straw into the rearing water of Xenopus laevis tadpoles, and by feeding X. laevis froglets diets containing rice grains of T1C-19 or its non-transformed counterpart MH63. Virulent Hessian fly larvae manipulate the free amino acid content of host wheat plants.

Gall-forming insects induce host plants to form specialized structures (galls) that provide immature life stages of the insect access to host plant nutrients and protection from natural enemies. Feeding by larvae of the Hessian fly (Mayetiola destructor Say) causes susceptible host wheat plants to produce a gall-like nutritive tissue that supports larval growth and development. To determine if changes in host plant free amino acid levels are associated with virulent Biotype L Hessian fly larval feeding, we quantified free amino acid levels in crown tissues of susceptible Newton wheat plants 1,

Use of chemical communication in the management of freshwater aquatic species that are vectors of human diseases or are invasive.

Chemical communication occurs when both originator (signaller) and one or more receiver(s) possess specializations for chemical exchange of information. Chemical information can be used by a wide variety of species to locate food and mates, avoid predators and engage in social interactions. In this review, we focus on chemical signalling between mates or cues from nest sites or hosts by selected aquatic pest species and indicate how chemical information can be used to manage pests. The pests are vectors of disease (blood-sucking insects) or invasive species (crayfishes and fishes) that have ex
