Biosynthesis and function of gangliosides.

Gangliosides are unique acidic glycolipids that are selectively concentrated in the plasma membrane of cells. Surface labeling studies have demonstrated that at least a portion of the oligosaccharde chain of gangliosides extends beyond the hydrophe) is imbedded in the membrane bilayer. It is becoming increasingly apparent that gangliosides participate in the internalization of environmental signals elicited by cholera toxin and glycoprotein hormones such as thyrotropic hormone and chorionic gonadotropin as well as other substances such as interferon and possibly serotonin. The mechanism by whi

Strikingly Different Roles of SARS-CoV-2 Fusion Peptides Uncovered by Neutron Scattering.

Coronavirus disease-2019 (COVID-19), a potentially lethal respiratory illness caused by the coronavirus SARS-CoV-2, emerged in the end of 2019 and has since spread aggressively across the globe. A thorough understanding of the molecular mechanisms of cellular infection by coronaviruses is therefore of utmost importance. A critical stage in infection is the fusion between viral and host membranes. Here, we present a detailed investigation of the role of selected SARS-CoV-2 Spike fusion peptides, and the influence of calcium and cholesterol, in this fusion process. Structural information from sp

Mesenchymal Stem Cell-Derived Exosomes Ameliorate Delayed Neurocognitive Recovery in Aged Mice by Inhibiting Hippocampus Ferroptosis via Activating SIRT1/Nrf2/HO-1 Signaling Pathway.

Delayed neurocognitive recovery (dNCR) is a prevalent perioperative neurological complication in older patients and has common characteristics such as acute cognitive dysfunction, impaired memory, and inattention. Mesenchymal stem cell-derived exosomes (MSCs-Exo) are enclosed by a lipid bilayer contain proteins, DNA, miRNA, and other components, which are important mediators of intercellular communication. It has been reported that exosomes could play an important role in the treatment of neurodegenerative diseases, nerve injury, and other neurological diseases. In this study, we examined the

Photosensitive nanocapsules for use in imaging from poly(styrene-co-divinylbenzene) cross-linked with coumarin derivatives.

The study objective was to generate biocompatible probes and develop a stable macromolecule imaging system that are based on nanolipopolymersomes and can be used in living cells. We synthesized nanolipopolymersomes with a fluorescent polymer wall surrounded by an outer phospholipid shell that exhibits potential for the controlled delivery of diagnostic agents to cells. We describe a new type of probe suitable for dual detection methods (spectrophotometric and fluorescence). This aspect makes it unique among currently available probes because allows it to be detected with greater accuracy. We d

Effects of extracellular matrix protein functionalized fluid membrane on cell adhesion and matrix remodeling.

In order to study cellular responses and extracellular matrix protein remodeling mediated by biomaterials coating, we proposed a biomimetic construct containing protein-conjugated supported lipid bilayers (SLBs) as a cell culture platform. Single or multi-component proteins-bound SLBs were fabricated by conjugating type I collagen and/or fibronectin on the N-hydroxysulfosuccinimide-functionalized SLBs. The proposed protein-conjugated systems were quantitatively characterized by the quartz crystal microbalance with dissipation. NIH 3T3 fibroblasts were cultured on the model constructs and on ox…

Solution structure and interaction of cupiennin 1a, a spider venom peptide, with phospholipid bilayers.

The solution structure of cupiennin 1a, a 35 residue, basic antibacterial peptide isolated from the venom of the spider Cupiennius salei, has been determined by nuclear magnetic resonance (NMR) spectroscopy. The peptide was found to adopt a helix-hinge-helix structure in a membrane mimicking solvent. The hinge may play a role in allowing the amphipathic N-terminal helix and polar C-terminal helix to orient independently upon membrane binding, in order to achieve maximal antibacterial efficacy. Solid-state 31P and 2H NMR was used to further study the effects of cupiennin 1a on the dynamic prope

Interaction of zwitterionic penicillins with the OmpF channel facilitates their translocation.

To study translocation of beta-lactam antibiotics of different size and charge across the outer bacterial membrane, we combine an analysis of ion currents through single trimeric outer membrane protein F (OmpF) porins in planar lipid bilayers with molecular dynamics simulations. Because the size of penicillin molecules is close to the size of the narrowest part of the OmpF pore, penicillins occlude the pore during their translocation. Favorably interacting penicillins cause time-resolvable transient blockages of the small-ion current through the channel and thereby provide information about th…

Amyloid peptide channels: blockade by zinc and inhibition by Congo red (amyloid channel block).

Amyloid peptides are the major constituents of amyloid deposits in various amyloid diseases including Alzheimer's disease, type II diabetes mellitus, prion diseases and others. The hallmark of amyloid is the binding of the dye, Congo red, which creates characteristic staining due to the dye's ability to bind the beta sheet aggregates referred to as amyloid. Previous reports have demonstrated that several cytotoxic, amyloidogenic peptides can form ion channels in planar phospholipid bilayer membranes and have suggested that these channels may represent the pathogenic mechanism of cell and tissu

Interaction of internal Ba2+ with a cloned Ca(2+)-dependent K+ (hslo) channel from smooth muscle.

We have studied potassium currents through a cloned Ca(2+)-dependent K+ channel (hslo) from human myometrium. Currents were recorded in inside-out macropatches from membranes of Xenopus laevis oocytes. In particular, the inactivation-like process that these channels show at high positive potentials was assessed in order to explore its molecular nature. This current inhibition conferred a bell shape to the current-voltage curves. The kinetic and voltage dependence of this process suggested the possibility of a Ba2+ block. There were the following similarities between the inactivation process ob

Visualization of CD2 interaction with LFA-3 and determination of the two-dimensional dissociation constant for adhesion receptors in a contact area.

Many adhesion receptors have high three-dimensional dissociation constants (Kd) for counter-receptors compared to the KdS of receptors for soluble extracellular ligands such as cytokines and hormones. Interaction of the T lymphocyte adhesion receptor CD2 with its counter-receptor, LFA-3, has a high solution-phase Kd (16 microM at 37 degrees C), yet the CD2/LFA-3 interaction serves as an effective adhesion mechanism. We have studied the interaction of CD2 with LFA-3 in the contact area between Jurkat T lymphoblasts and planar phospholipid bilayers containing purified, fluorescently labeled LFA-…

Antivirals acting on viral envelopes via biophysical mechanisms of action.

Most antivirals target viral proteins and are specific for only one virus, or viral type. Whereas viral proteins are encoded in the plastic viral genome, virion lipids are not and their rearrangements during fusion are conserved among otherwise unrelated enveloped viruses. Antivirals that inhibit these lipid rearrangements could thus pose a high barrier to resistance and have broad-spectrum activity. Fusion occurs through a hemifusion stalk in which only the outer leaflets are fused and thus curved with a smaller radius for the polar heads than for the hydrophobic tails (negative curvature). O

Design, Synthesis, and Physicochemical Characterization of Boronic Acid-Linked Cationic Lipids: Aggregation, Diol Recognition, Membrane Perturbation, and DNA Complexation.

Boronic acids are attractive functional groups because they form reversible covalent bonds with cis-diols, enabling selective recognition of saccharides, glycoproteins, and related biomolecules. Although widely exploited in sensing and glycobiology, their incorporation into lipid-based systems remains relatively unexplored. Integrating boronic-acid-mediated molecular recognition into amphiphilic frameworks while preserving controlled self-assembly and biomolecular interactions continues to be a significant challenge. Here, we report the design, synthesis, and characterization of a new series o

Nutritional properties of extracellular vesicle-like particles from Sophora flavescens and Periplaneta americana with effects on streptozotocin-induced diabetic wound healing in rats.

INTRODUCTION: The medicinal plant Sophora flavescens (SF) and the insect Periplaneta americana (PA) have long been used in treating diabetic wounds. PA-derived extracellular vesicle-like particles (PA-EVLP) can accelerate diabetic wound healing. However, no evidence of the nutritional components and anti-diabetic wound effects of the combination of SF-EVLP with PA-EVLP (SF-PA-EVLPs) is available. METHODS: We separated SF-EVLP and PA-EVLP via ultracentrifugation and characterized them via nano flow cytometry, transmission electron microscopy, and atomic force microscopy. Ultra-high-performance

Feedback-controlled single-frequency microwave thermal cycling enables dynamic modulation of liposome membrane permeability.

Liposomes are widely used as delivery platforms for therapeutic drugs, nucleic acids, and diagnostic agents. Because cargo release is governed by the permeability of the lipid bilayer, controlling membrane permeability is essential for regulating molecular release from liposomes. Various stimuli, including pH, light, ultrasound, magnetic fields, and temperature, have been explored to control liposomal membrane permeability and cargo release. Among these approaches, microwave (MW) irradiation has also been investigated as a means of modulating liposome permeability, with early studies demonstra

Aging impact on canine extracellular vesicles production, size, and miRNA content.

Increasing life expectancy has been accompanied by a higher prevalence of age-associated conditions, underscoring the need to better understand biological mechanisms of aging and to identify accessible biomarkers. Extracellular vesicles (EVs) are lipid-bilayer vesicles that mediate intercellular communication by transporting proteins, lipids, and regulatory RNAs, including microRNAs (miRNAs). Here, we investigated whether the abundance, morphology, and molecular cargo of plasma-derived EVs differ between young and senior dogs. Blood samples were obtained from 24 clinically healthy dogs in Gama

Engineering Plant-Derived Exosome-like Nanoparticles as Bioinspired Nanocarriers: From Physicochemical Properties to Tumor Delivery Performance.

Plant-derived exosome-like nanoparticles (PELNs) are lipid bilayer nanostructures containing endogenous lipids, proteins, nucleic acids, and phytochemicals, which have attracted increasing interest as bioinspired carriers for cancer therapy. This review evaluates how plant source, isolation, purification procedures, vesicle composition, cargo-loading strategy, and administration route shape the quality and tumor-delivery performance of PELNs. The available evidence indicates that plant source and processing are major determinants of particle size, purity, surface charge, cargo profile, and bio

Stem cell-derived extracellular vesicles -mediated bone regeneration: mechanisms, targeted delivery, and clinical perspectives in promoting angiogenesis.

Angiogenesis is indispensable for bone regeneration and tightly coupled to osteogenesis, yet inadequate vascularization remains a common cause of repair failure in critical-sized defects, compromised fractures, and osteochondral lesions. Conventional growth-factor delivery and cell transplantation rarely reproduce the spatiotemporal vascular program required for functional healing, motivating the search for controllable and translatable alternatives. Mesenchymal stem/stromal cell (MSC)-derived extracellular vesicles (EVs) have emerged as a modular, cell-free modality that packages regenerative

Bacterial Extracellular Vesicles as Regulators of the Gut Microbiome-Circadian Axis in Inflammatory and Metabolic Diseases.

Bacteria-derived extracellular vesicles (BEVs) have emerged as key mediators of intercellular and interkingdom communication. These nanoscales, lipid bilayer-enclosed particles are secreted by both Gram-positive and Gram-negative bacteria and carry diverse bioactive cargoes, including proteins, lipids, nucleic acids, and metabolites. Through the delivery of these molecules, BEVs regulate host physiology by modulating immune responses, epithelial barrier integrity, and gut microbial community composition. Accumulating evidence indicates that BEVs derived from probiotics or commensal bacteria pr

Plasma-derived DNA containing-extracellular vesicles induce STING-mediated proinflammatory responses in dermatomyositis.

Objectives: Extracellular vesicles (EVs) are lipid bilayer membrane vesicles that are present in various bodily fluids and have been implicated in autoimmune disease pathogenesis. Type I interferons (IFN), specifically IFN-β, are uniquely elevated in dermatomyositis (DM). The stimulator of interferon genes (STING) works as a critical nucleic acid sensor and adaptor in type I IFN signaling with possible implications in autoimmune diseases such as DM. In the current study, we investigated whether circulating EVs contribute to proinflammatory effects in DM, whether these proinflammatory responses
