Using whole-cell patch-clamp techniques, we demonstrate, for the first time, that rat submandibular acinar cells contain a tetraethylammonium (TEA)-insensitive, Ca(2+)-activated K+ conductance which is not attributable to large conductance, voltage-sensitive, Ca(2+)-dependent K+ channels (maxi-K+ channels). Taken together with our recent K+ efflux and fluid secretion studies in intact rat submandibular gland, we postulate that the K+ conductance reported here may be involved in the basolateral K+ efflux pathway activated by cytosolic Ca2+ concentration during secretion by this gland.

The properties of K+ channels in the isolated oxyntic cells of the bullfrog (Rana catesbeiana) were investigated using the patch-clamp method. The kinetic characteristics of a purified insect dihydrofolate reductase (DHFR) have been described. The Km values for the substrate dihydrofolate and the cofactor NADPH have been estimated by primary and secondary Hanes plots to be 0.3 and 5.2 microM, respectively. Membrane potentials of medullary chromaffin cells of the adrenal gland of the mouse were measured in situ. Resting potential (-54.3 +/- 8.8 mV) depended on extracellular [K+] as predicted by the constant-field equation with a permeability ratio, PNa/PK, of 0.09. By means of the system of ion-selective electrodes the coupling transport of K+, H+ and Cl- ions in liposome suspension was studied. Nigericin and tributyl tin (TBT) were used as inductors of cation and anion exchange. Effect of pH gradient created on the membrane on ionic transport was studied. The latter was shown to control the antiport exchange of both, cations and anions. At the joint effect of nigericin and TBT the yield of K+ and Cl- ions from liposomes independent of the pH gradient was observed.
