Effect of Linker Sequence on the Interdomain Dynamics of Self-Sufficient CYP116B5-SOX Chimeras.

In this work, the design of CYP116B5-SOX-(PPII) is presented. This is an artificial self-sufficient enzyme in which the P450 116B5 peroxygenase and the H2O2-generating sarcosine oxidase (SOX) are linked together in a fusion complex. A polyproline type II helix (PPII) linker was used to connect the two enzymes, obtaining a rigid fusion protein. CYP116B5-SOX-(PPII) displayed 10-fold increased activity in terms of p-nitrophenol oxidation (k cat: 32.1 ± 0.5 min-1; K M: 150 ± 15 μM) compared to the isolated heme domain CYP116B5-hd, and more than 2-fold higher activity compared to CYP116B5-SOXa fus

Development of novel enzyme immobilization methods employing formaldehyde or triethoxysilylbutyraldehyde to fabricate immobilized enzyme microreactors for peptide mapping.

The digestion of proteins with proteolytic enzymes has expedited the analysis of peptide mapping. Here, we compared the digestion efficiency of soluble chymotrypsin (CT) with two immobilized CT preparations using bovine serum albumin (BSA) as the substrate. An efficient method of immobilizing chymotrypsin using formaldehyde (FA) was optimized and the conditions were applied to assess a novel immobilization reagent, triethoxysilylbutaraldehyde (TESB). Efforts to determine the best enzyme-to-substrate (E : S) ratios during digestion of denatured BSA with single-use FA-CT enzyme particles were pe

Mercury and metallothioneins in blood fractions and tissues of captive Morelet's crocodiles in Quintana Roo, Mexico.

Even though studies of heavy metals and their relation with metallothioneins (MTs) in reptile tissues have been conducted, research on heavy metals and MTs in organs and blood fractions of crocodylians is limited. To date there are no studies on the distribution of MTs in organs and their relation with mercury (Hg), or the concentration of MTs in plasma and erythrocytes of crocodylians. In order to understand the role of MTs in crocodiles, our aim was to assess the detoxification mechanisms for exposure to metals in Morelet's crocodile (Crocodylus moreletii) by using two biomarkers (Hg and MTs

Novel antibodies reveal inclusions containing non-native SOD1 in sporadic ALS patients.

Mutations in CuZn-superoxide dismutase (SOD1) cause amyotrophic lateral sclerosis (ALS) and are found in 6% of ALS patients. Non-native and aggregation-prone forms of mutant SOD1s are thought to trigger the disease. Two sets of novel antibodies, raised in rabbits and chicken, against peptides spaced along the human SOD1 sequence, were by enzyme-linked immunosorbent assay and an immunocapture method shown to be specific for denatured SOD1. These were used to examine SOD1 in spinal cords of ALS patients lacking mutations in the enzyme. Small granular SOD1-immunoreactive inclusions were found in

A novel founder BBS1 mutation explains a unique high prevalence of Bardet-Biedl syndrome in the Faroe Islands.

BACKGROUND/AIM: Bardet-Biedl syndrome is a multiorgan disease presenting with retinitis pigmentosa leading to blindness. The aim of the study was to investigate the genetic background of Bardet-Biedl syndrome in the Faroe Island. It was hypothesised that a common genetic background for the syndrome would be found. METHODS: Patients were identified from the files of the Retinitis Pigmentosa Register at the National Eye Clinic, Denmark. The diagnosis of Bardet-Biedl syndrome was verified from medical files. Mutational screening of BBS1, BBS2, BBS4, MKKS and BBS10 was done by denaturing high-per

Prevalence of BRCA1 and BRCA2 mutations in Pakistani breast and ovarian cancer patients.

Among Asian countries, Pakistan has the highest rates of breast and ovarian cancer. To assess the contribution of the BRCA1 and BRCA2 germ line mutations to these high rates, we conducted the first study of 176 Pakistani breast and ovarian cancer patients, selected on family history and on age of diagnosis. Comprehensive BRCA mutation screening was performed using a range of techniques, including denaturing high-pressure liquid chromatography, single strand conformational polymorphism analysis and protein truncation test, followed by DNA sequencing. Thirty deleterious germ-line mutations were

A molecular and clinical study of Larsen syndrome caused by mutations in FLNB.

BACKGROUND: Larsen syndrome is an autosomal dominant osteochondrodysplasia characterised by large-joint dislocations and craniofacial anomalies. Recently, Larsen syndrome was shown to be caused by missense mutations or small inframe deletions in FLNB, encoding the cytoskeletal protein filamin B. To further delineate the molecular causes of Larsen syndrome, 20 probands with Larsen syndrome together with their affected relatives were evaluated for mutations in FLNB and their phenotypes studied. METHODS: Probands were screened for mutations in FLNB using a combination of denaturing high-performa

BRCA1 and BRCA2 mutation analysis in breast-ovarian cancer families from northeastern Poland.

Sixty high-risk breast and/or ovarian cancer families from North-Eastern Poland were screened for germline mutations in BRCA1 (MIM# 113705) and BRCA2 (MIM# 600185), using a combination of protein truncation test, denaturing high-performance liquid chromatography and direct sequencing. Sixteen (27%) of the families were found to carry nine different BRCA mutations, including 14 families with BRCA1 mutation and two families with BRCA2 mutation. The results suggest the presence of two strong BRCA1 founder mutations in the Polish population - 5382insC (6 families) and 300T>G (Cys61Gly; 3 families)

[Time-resolved immunofluorometric assay for DNA and DNA level in human serum].

It is widely accepted that normal levels of extracellular DNA in the blood are quite low and can be elevated in systemic lupus erythematosus, some cancers, and after irradiation. The goal of this work was the development of a quantitative assay for DNA measurement in serum of healthy donors as well as in patients. Several approaches to IgG antibody induction against native mammalian DNA were tested. Antiserum containing such antibodies was obtained following immunization of rabbits with denatured DNA complexed to methylated BSA (in one of six animals). A time-resolved immunofluorimetric sandwi

Protective effect of administration of skim milk on exogenous and endogenous infection in mice.

In order to minimize the denaturation of proteins in milk, normal cow's milk was pasteurized at 61 C for 20 min. The protective effects of the thus prepared skim milk (low-heat skim milk) on exogenous and endogenous infection were examined as compared with conventional skim milk which was pasteurized at 121 C for 2 sec. The antibody titers to Listeria monocytogenes and Escherichia coli of low-heat skim milk were almost equal to that of raw milk, while no antibody was detected in the conventional skim milk. When mice were given low-heat skim milk or conventional skim milk, the incidence of the

Absence of p53 mutation in Japanese patients with malignant thyroid lymphoma.

Abnormalities of the gene encoding the sequence-specific DNA-binding protein, nuclear phosphorprotein p53, are among the most common genetic alterations observed in human cancers. A mutation of this tumor suppressor gene has been reported with a low prevalence in differentiated thyroid carcinomas, while the prevalence was high in undifferentiated thyroid carcinomas. We used denaturing gradient gel electrophoresis (DGGE) to probe for mutations of p53 gene in order to determine its role in the genesis of malignant thyroid lymphoma. Involvement of 27 samples had been proven by histopathologic exa

Secretion-defective mutations in the signal sequence for Saccharomyces cerevisiae invertase.

Nine mutations in the signal sequence region of the gene specifying the secreted Saccharomyces cerevisiae enzyme invertase were constructed in vitro. The consequences of these mutations were studied after returning the mutated genes to yeast cells. Short deletions and two extensive substitution mutations allowed normal expression and secretion of invertase. Other substitution mutations and longer deletions blocked the formation of extracellular invertase. Yeast cells carrying this second class of mutant gene expressed novel active internal forms of invertase that exhibited the following proper

The structural basis of cell-mediated immunological reactions of collagen. Characteristics of cutaneous delayed hypersensitivity reactions in specifically sensitized guinea-pigs.

Cell-mediated immunological properties of collagen were studied in guinea-pigs employing cutaneous delayed hypersensitivity reactions. The animals were sensitized by a single injection of highly purified native collagen in Freund's complete adjuvant (FCA). Reactivity could be induced with 150 μg of calf collagen. Maximal reactivity was obtained 20 days after sensitization and persisted for more than 3 months. Histologically, the reactions displayed the typical features of delayed reactions with infiltration of predominantly mononuclear cells. No reactivity was induced in animals injected with

Tamm Plasmon Resonance Responsiveness to SARS-CoV-2 Virus-Like Particles.

Bioresponsive optical materials that convert nanoscale biointerface events into measurable spectral signals are of growing interest for sensing and antiviral applications. Here, we show that a Tamm plasmon (TP) device, consisting of a SiO2/TiO2 distributed Bragg reflector capped with nanostructured silver, responds to SARS-CoV-2 virus-like particles (VLPs) in a concentration-dependent manner without selective functionalization. Upon VLP exposure, the conventional wavelength shift (Δλ) of the TP resonance is small (∼4 nm), whereas the resonance depth decreases systematically, by about 60% at 10

Nanosecond-pulsed plasma protects against bacterial fruit blotch and promotes melon seedling growth.

BACKGROUND: Bacterial fruit blotch (BFB), known as the 'cancer' of cucurbits, is a seed-borne disease of melons caused by Acidovorax citrulli. Traditional chemical treatments for BFB are ineffective and adversely affect the environment. Using dielectric barrier discharge (DBD) nanosecond-pulsed plasma technology, melon seeds were treated to promote germination and growth and to control BFB. RESULTS: Based on the evaluation parameters of seed germination, seedling growth, leaf yellowing and bacterial infection after seed plasma treatments, 9 min at 20 kV was selected as the optimal plasma disc

Hypothermia Is a Potential New Therapy for a Subset of Tumors with Mutant p53.

The tumor suppressor p53 gene is mutated in approximately 50% of all human tumors. Many tumor-associated mutant p53 proteins misfold into a common, denatured conformation and accumulate to high levels in human tumors. In such tumors, these mutant forms of p53 provide a "gain of function" to promote tumor progression. Therefore, targeting mutant p53 has become an attractive approach for cancer therapy. In this issue, the study by Lu and colleagues supports the premise that certain forms of mutant p53 are temperature sensitive in conformation; these forms of p53 are mutant in conformation at phy

Kinetics of cyanide binding as a probe of local stability/flexibility of cytochrome c.

Effect of anions of the Hofmeister series (thiocyanate, perchlorate, iodide, bromide, nitrate, chloride, sulfate, and phosphate) on local and global stability and flexibility of horse heart ferricytochrome c (cyt c) has been studied. Global stability of cyt c was determined by iso/thermal denaturations monitored by change in ellipticity in the far-UV region and its local stability was determined from absorbance changes in the Soret region. Particularly, relative stability/flexibility of the Met80-heme iron bond has been assessed by analysis of binding of cyanide into the heme iron. Both global

Formulating erythropoietin-loaded sustained-release PLGA microspheres without protein aggregation.

We report a simple method to microencapsulate erythropoietin (EPO, a protein easily denatured and antigenized by contact with water-organic solvent interfaces) into poly(lactic-co-glycolic acid) (PLGA) microspheres with minimal aggregation. This formulation process involved an aqueous-aqueous emulsion formed at reduced temperature. EPO was first dissolved in water together with dextran (MW=70,000) and polyethylene glycol (MW=8000), followed by a freezing process during which dextran separated out as the dispersed phase with EPO partitioned in preferentially. The frozen sample was then lyophili
