β-Catenin Upregulates the Constitutive and Virus-Induced Transcriptional Capacity of the Interferon Beta Promoter through T-Cell Factor Binding Sites.

Rapid upregulation of interferon beta (IFN-β) expression following virus infection is essential to set up an efficient innate antiviral resp … asizing the role of β-catenin in the innate antiviral response, we show here that highly pathogenic Rift Valley fever virus (RVFV) targets the Wnt/β-catenin pathway and the formation of active TCF/β-catenin complexes at the transcriptional and protein level in RVFV-infected cells and mice.

Spatial and temporal distribution of West Nile virus in horses in Israel (1997-2013)--from endemic to epidemics.

With the rapid global spread of West Nile virus (WNV) and the endemic state it has acquired in new geographical areas, we hereby bring a thorough serological investigation of WNV in horses in a longstanding endemic region, such as Israel. This study evaluates the environmental and demographic risk factors for WNV infection in horses and suggests possible factors associated with the transition from endemic to epidemic state. West Nile virus seroprevalence in horses in Israel was determined throughout a period of more than a decade, before (1997) and after (2002 and 2013) the massive West Nile f

The RNA helicase eIF4A as a novel target in insect cells to combat arboviral infections.

Arthropod-borne viruses cause major global health burdens, yet few vaccines or antivirals exist. Targeting host factors required for viral replication offers a promising approach. The DEAD-box RNA helicase eIF4A, a core component of the translation initiation complex eIF4F, unwinds structured 5' UTRs and is therefore critical also for the translation of many viral RNAs. The compound classes rocaglates and pateamines are potent eIF4A inhibitors in mammalian cells. Here we show that the natural rocaglate silvestrol strongly inhibits Rift Valley fever virus replication (RVFV) in human cells witho…

Oropouche virus NSs protein suppresses host transcription by targeting the RNA polymerase II RPB1 protein.

UNLABELLED: Oropouche fever is a debilitating disease caused by Oropouche virus (OROV), an arthropod-borne member of the Peribunyaviridae fa … of its C-terminal domain (CTD) at serine 2 and serine 5 residues. When expressed from a recombinant Rift Valley fever virus MP-12 strain, OROV NSs colocalized with NPM1 and contributed to its nucleolar redistribution. Furthermore, expression of OROV NSs induced a marked reduction in the hyperphosphorylated RNAP IIo form, which was largely restored upon treatment wi …

RVFV virulence factor NSs triggers the mitochondrial MCL-1-BAK axis to activate pathogenic NLRP3 pyroptosis.

Infection of Rift Valley fever virus (RVFV), a highly pathogenic mosquito-borne zoonotic virus, triggers severe inflammatory pathogenesis but the underlying mechanism of inflammation activation is currently unclear. Here, we report that the non-structural protein NSs of RVFV triggers mitochondrial damage to activate the NLRP3 inflammasome leading to viral pathogenesis in vivo. It is found that the host transcription inhibition effect of NSs causes rapid down-regulation of myeloid cell leukemia-1(MCL-1), a pro-survival member of the Bcl-2 (B-cell lymphoma protein 2) protein family. MCL-1 down-r…

Extended protection against phlebovirus infection conferred by recombinant adenovirus expressing consensus interferon (DEF201).

Punta Toro virus (PTV; Bunyaviridae, Phlebovirus) is related to Rift Valley fever virus (RVFV), a pathogenic agent which causes severe disease in humans and livestock primarily in the sub-Saharan region of Africa. The recent range expansion of RVFV and the potential for its intentional release into naïve populations pose a significant threat to public health and agriculture. Studies modeling disease in rodents and nonhuman primates have shown that PTV and RVFV are highly sensitive to the antiviral effects of alpha interferon (IFN-α), an important component of the innate antiviral host response…

Suitability of transiently expressed antibodies for clinical studies: product quality consistency at different production scales.

Transgenic human monoclonal antibodies derived from humanized mice against different epitopes of the Middle East respiratory syndrome coronavirus (MERS-CoV), and chimeric llama-human bispecific heavy chain-only antibodies targeting the Rift Valley fever virus (RVFV), were produced using a CHO-based transient expression system. Two lead candidates were assessed for each model virus before selecting and progressing one lead molecule. MERS-7.7G6 was used as the model antibody to demonstrate batch-to-batch process consistency and, together with RVFV-107-104, were scaled up to 200 L. Consistent exp…

Host genetic variation in susceptibility to Punta Toro virus.

Infection of small laboratory animals by Punta Toro virus (PTV), family Bunyaviridae, genus Phlebovirus, is a model for the study of the human pathogen Rift Valley fever virus (RVFV). We have identified inbred mouse strains with significant differences in host response to the Adames strain of PTV. Nine inbred strains of mice representing major branches in the Mus musculus phylogeny were inoculated subcutaneously with a high dose of PTV in survival experiments. Two inbred strains of mice, NZW/LacJ and 129S1/SvImJ, died ~4 days after PTV infection, whereas 7 other strains survived the challenge …

Characterization of the non-specific humoral and cellular antiviral immunity stimulated by the chloroform-methanol residue (CMR) fraction of Coxiella burnetii.

Modulation of the immune response by the chloroform-methanol residue (CMR) of phase I Coxiella burnetii whole cell was studied in Rift Valley fever virus-infected, or in naive endotoxin-non-responder C3H/HeJ mice. A single dose of CMR completely protected the mice from viral infection. Treating virus-infected mice with antibodies directed against interferons alpha/beta (IFN-alpha beta) and gamma (IFN-gamma) eliminated the CMR-induced protection. CMR stimulated the production of high levels of IFN-alpha/beta and 2'-5'-oligoadenylate synthetase activities in sera of the CMR-treated mice. IFN-gam…
