Indole-3-acetic acid production is rare among gut bacteria and reflects OFOR-driven amino acid oxidation in acetogens.

Indole-3-acetic acid (IAA) is a tryptophan-derived gut microbial metabolite with reported anti-inflammatory activities, but the organisms and anaerobic pathways that support robust production remain unclear. Screening 206 human gut bacterial isolates by LC-MS revealed that IAA production is rare: only five strains exceeded the limit of quantitation, and high-capacity production was confined to the acetogens Blautia hydrogenotrophica and Intestinibacter bartlettii. Across growth conditions, IAA was a minor product that rose alongside carbohydrate-sensitive, OFOR-linked catabolism of multiple am…

Acetic acid can catalyze succinimide formation from aspartic acid residues by a concerted bond reorganization mechanism: a computational study.

Succinimide formation from aspartic acid (Asp) residues is a concern in the formulation of protein drugs. Based on density functional theory calculations using Ace-Asp-Nme (Ace = acetyl, Nme = NHMe) as a model compound, we propose the possibility that acetic acid (AA), which is often used in protein drug formulation for mildly acidic buffer solutions, catalyzes the succinimide formation from Asp residues by acting as a proton-transfer mediator. The proposed mechanism comprises two steps: cyclization (intramolecular addition) to form a gem-diol tetrahedral intermediate and dehydration of the in…

Acetic acid upregulates the expression of genes for fatty acid oxidation enzymes in liver to suppress body fat accumulation.

We investigated the effect of acetic acid (AcOH) on the prevention of obesity in high-fat-fed mice. The mice were intragastrically administrated with water or 0.3 or 1.5% AcOH for 6 weeks. AcOH administration inhibited the accumulation of body fat and hepatic lipids without changing food consumption or skeletal muscle weight. Significant increases were observed in the expressions of genes for peroxisome-proliferator-activated receptor alpha (PPARalpha) and for fatty-acid-oxidation- and thermogenesis-related proteins: acetyl-CoA oxidase (ACO), carnitine palmitoyl transferase-1 (CPT-1), and unco…

Determination of valproic acid (di-N-propyl acetic acid) in plasma by gas-liquid chromatography with pre-column butylation.

A gas-liquid chromatographic procedure for the determination of the anti-convulsant drug valproic acid in plasma or serum is described. Valproic acid is extracted from acidified plasma (or serum) into toluene containing octanoic acid as the internal standard. The toluene layer is back extracted with a methanolic solution of tetramethylammonium hydroxide; an aliquot of the methanolic layer is mixed after phase separation with N,N-dimethylacetamide and 1-iodobutane. After centrifugation the clear supernatant (which is stable for at least 24 h) is analysed by gas-liquid chromatography, on a 3% OV

Biophysical and Computational Investigation of the Interaction between Indole-3-Acetic Acid and DNA.

In this work, the interaction of indole-3-actic acid (IAA) -the plant growth hormone auxin- with salmon sperm DNA was investigated by different method such as UV-Vis absorption and fluorescence emission spectroscopy, viscosity measurement and voltammetric methods. Also, the computational strategy including molecular dynamics simulation and molecular docking study was used to consider the details of IAA-DNA interaction. The experimental results suggested that the interaction of IAA with DNA is predominantly intercalative. The thermodynamic parameters (ΔG, ΔH, and ΔS) suggested that hydrogen bon

Quantitative Metaproteomic Characterization of Acetic Acid Bacteria Reveals Functional Dynamics During Verdejo Wine Acetification.

Background: Acetification is a complex process driven by acetic acid bacteria (AAB), in which high ethanol and acidity levels require strong microbial metabolic adaptation. Although the microbiota involved in vinegar production has been described, the functional mechanisms that enable these bacteria to maintain metabolic activity remain poorly understood. In this study, the functional dynamics of AAB during Verdejo vinegar acetification were analyzed using a quantitative metaproteomic approach. Methods: Acetification was performed in submerged culture under semi-continuous conditions, and samp…

Evaluation of Acetic Acid-Induced Chronic Gastric Ulcer Healing by Propionyl-L-Carnitine Administration.

The aim of our study was to investigate the healing effect of propionyl-L-carnitine (PLC) on chronic gastric ulcers and its underlying mechanisms. This study included rats with gastric ulcers induced by applying serosal glacial acetic acid. These rats were then given either saline (vehicle) or PLC at doses of 60 and 120 mg/kg, administered orally 3 days after ulcer induction for 14 consecutive days. Our study found that treatment with PLC resulted in a reduction of the gastric ulcer area, a faster rate of ulcer healing, and stimulated mucosal restoration. Additionally, the treatment with PLC r…

Surface Tuning of La0.5Sr0.5CoO3 Perovskite Catalysts by Acetic Acid for NOx Storage and Reduction.

Selective dissolution of perovskite A site (A of ABO3 structure) was performed on the La1 - xSrxCoO3 catalysts for the NOx storage and reduction (NSR) reaction. The surface area of the catalysts were enhanced using dilute HNO3 impregnation to dissolve Sr. Inactive SrCO3 was removed effectively within 6 h, and the catalyst preserved the perovskite framework after 24 h of treatment. The tuned catalysts exhibited higher NSR performance (both NOx storage and NO-to-NO2 oxidation) under lean-burn and fuel-rich cycles at 250 °C. Large amounts of NOx adsorption were due to the increase of nitrate/nitr

Novel wine yeast with mutations in YAP1 that produce less acetic acid during fermentation.

Acetic acid, a byproduct formed during yeast alcoholic fermentation, is the main component of volatile acidity (VA). When present in high concentrations in wine, acetic acid imparts an undesirable 'vinegary' character that results in a significant reduction in quality and sales. Previously, it has been shown that saké yeast strains resistant to the antifungal cerulenin produce significantly lower levels of VA. In this study, we used a classical mutagenesis method to isolate a series of cerulenin-resistant strains, derived from a commercial diploid wine yeast. Four of the selected strains showe…

Glacial acetic acid treatment and atypical endocervical glandular cells: an analysis of 92 cases.

Glacial acetic acid (GAA) treatment minimizes the risk that bloody ThinPrep (Hologic, Marlborough, MA) Papanicolaou (Pap) test samples will be unsatisfactory for diagnosis. In our experience, GAA treatment also adversely affects the morphologic appearance of endocervical glandular cells. We analyzed a series of 92 consecutive GAA-treated Pap tests interpreted as atypical endocervical cells (AECs) and compared these with a control group of 130 samples with AECs in Pap tests that had not been treated with GAA to determine if GAA treatment increases the false-positive diagnosis of AECs. By using …

Tipepidine enhances the antinociceptive-like action of carbamazepine in the acetic acid writhing test.

Several antidepressants have been used to treat severe pain in clinics. Recently, we reported that the centrally acting non-narcotic antitussive (cough suppressant drug), tipepidine produces an antidepressant-like effect in the forced swimming test, although the mechanism of action appears to be quite different from that of known antidepressants. In the present study, we investigated whether a combination of tipepidine and carbamazepine acts synergistically to induce an antinociceptive effect in the acetic acid-induced writhing test in mice. Prior to studying the combination of tipepidine and …
