Human neurospheres as three-dimensional cellular systems for developmental neurotoxicity testing.

BACKGROUND: Developmental neurotoxicity (DNT) of environmental chemicals is a serious threat to human health. Current DNT testing guidelines propose investigations in rodents, which require large numbers of animals. With regard to the "3 Rs" (reduction, replacement, and refinement) of animal testing and the European regulation of chemicals [Registration, Evaluation, and Authorisation of Chemicals (REACH)], alternative testing strategies are needed in order to refine and reduce animal experiments and allow faster and less expensive screening. OBJECTIVES: The goal of this study was to establish

Antiphospholipid antibodies inhibit the migration and invasion of trophoblast cells by suppressing the JNK/C-Jun/MMP1 signaling pathway.

Источник пометил публикацию как отозванную

BACKGROUND: Antiphospholipid syndrome (APS) is an autoimmune disease primarily manifested by recurrent thrombosis and pregnancy-related complications. The migration and invasion abilities of trophoblast cells play a crucial role in maintaining normal pregnancy. It is now increasingly recognized that adverse pregnancy outcomes in APS are associated with the disruption of trophoblast function by aPL (antiphospholipid antibodies), rather than thrombotic occlusion of the placental vasculature. Therefore, this article aimed to explore the potential mechanisms by which aPL affect trophoblast cell fu

piRNA DQ690018 increased bone mass by promoting the proliferation and osteogenic differentiation of BMSCs.

BACKGROUND: The proliferation, migration, and osteogenic differentiation of bone mesenchymal stem cells (BMSCs) play vital roles in maintaining bone mass and metabolism. BMSC base cell therapy has become a competitive treatment option for osteoporosis. PIWI-interacting RNAs (piRNAs) are critical regulators of cellular processes. METHODS: Small RNA sequencing and qPCR were used to detect piRNA expression during BMSC proliferation. CCK-8, clone formation, and EdU assays were used to detect the effect of piRNA DQ690018 on BMSC proliferation. Wound healing and Transwell assays were used to assess

Upregulation of E-cadherin by the combination of methionine restriction and HDAC2 intervention for inhibiting gastric carcinoma metastasis.

Invasion and metastasis are the leading causes of death in individuals with malignant tumors, including gastric cancer. In this study, we aim to explore the effect and related mechanisms of methionine restriction (MR) on gastric carcinoma metastasis. In the MR cell model, gastric carcinoma cells are cultured in the MR medium, and in the animal model, BALB/c nude rodents are administered with a methionine-free diet after receiving injections of MKN45 cells into the caudal vein. Transwell assay is used to detect cell invasion and migration. Chromatin immunoprecipitation is performed to investiga

GW4064 inhibits migration and invasion through cathepsin B and MMP2 downregulation in human bladder cancer.

The ability of bladder cancer to invade and metastasize often leads to poor prognosis in bladder cancer patients. The aim of this study was to evaluate the effect of the farnesoid X receptor (FXR) agonist GW4064 on the migration and invasion of human bladder cancer cells. Long-term exposure to GW4064 decreased the colony formation of RT4 and T24 cells. The wound healing migration assay revealed an inhibitory effect of GW4064 on both of these bladder cancer cell lines. In addition, integrin β3 expression and myosin light chain phosphorylation were decreased after GW4064 treatment. Immunocytoche

Evaluation of developmental toxicants and signaling pathways in a functional test based on the migration of human neural crest cells.

BACKGROUND: Information on the potential developmental toxicity (DT) of the majority of chemicals is scarce, and test capacities for further animal-based testing are limited. Therefore, new approaches with higher throughput are required. A screening strategy based on the use of relevant human cell types has been proposed by the U.S. Environmental Protection Agency and others. Because impaired neural crest (NC) function is one of the known causes for teratologic effects, testing of toxicant effects on NC cells is desirable for a DT test battery. OBJECTIVE: We developed a robust and widely appl

Up-regulation of stromal cell-derived factor-1 enhances migration of transplanted neural stem cells to injury region following degeneration of spiral ganglion neurons in the adult rat inner ear.

Neural stem cell (NSC) transplantation into the cochlea is widely used for the treatment of spiral ganglion neuron (SGN) degenerative disease and injury in the animal models, but the migration of the transplanted NSCs to the injury region is difficult and the mechanism is still unclear. In this study, we aimed to validate whether the SGN-degenerated cochlear microenvironment plays a role in the NSC migration and investigated whether stromal cell-derived factor-1 (SDF-1) was involved in the NSCs migration. Using a rat SGN degeneration model, we demonstrated that the transplanted NSCs are more l

Phenotypic and functional comparison of rat enteric neural crest-derived cells during fetal and early-postnatal stages.

In our previous study, we showed that with increasing time in culture, the growth characteristics of enteric neural crest-derived cells (ENCCs) change, and that the proliferation, migration and neural differentiation potential of these cells in vitro notably diminish. However, there are no studies on the developmental differences in these characteristics between fetal and early-postnatal stages in vitro or in vivo. In this study, we isolated fetal (embryonic day 14.5) and postnatal (postnatal day 2) ENCCs from the intestines of rats. Fetal ENCCs had greater maximum cross-sectional area of the

Ablation of nasal-associated lymphoid tissue does not affect focal ischemic brain injury in mice.

Stroke is a devastating disease with a strong inflammatory component. It has been shown that part of this response is mediated by IL17+ γδT cells. γδT cells constitute a lymphocyte population with innate features that mainly populates epithelial surfaces including skin, intestine, and airways. We have shown that in the context of stroke, T cells migrate from the small intestine to the meninges but whether they can migrate from other epithelial surfaces is still unknown. Because of its proximity, one possible source of stroke-associated IL17+ γδT cells could be the Nasal-Associated Lymphoid Tis

Expression of CD73 slows down migration of skin dendritic cells, affecting the sensitization phase of contact hypersensitivity reactions in mice.

BACKGROUND: Application of haptens to the skin induces release of immune stimulatory ATP into the extracellular space. This "danger" signal can be converted to immunosuppressive adenosine (ADO) by the action of the ectonucleotidases CD39 and CD73, expressed by skin and immune cells. Thus, the expression and regulation of CD73 by skin derived cells may have crucial influence on the outcome of contact hypersensitivity (CHS) reactions. OBJECTIVE: To investigate the role of CD73 expression during 2,4,6-trinitrochlorobenzene (TNCB) induced CHS reactions. METHODS: Wild type (wt) and CD73 deficient

Ultrasonic Stimulation of Mouse Skin Reverses the Healing Delays in Diabetes and Aging by Activation of Rac1.

Chronic skin-healing defects are one of the leading challenges to lifelong well-being, affecting 2-5% of populations. Chronic wound formation is linked to age and diabetes and frequently leads to major limb amputation. Here we identify a strategy to reverse fibroblast senescence and improve healing rates. In healthy skin, fibronectin activates Rac1 in fibroblasts, causing migration into the wound bed, and driving wound contraction. We discover that mechanical stimulation of the skin with ultrasound can overturn healing defects by activating a calcium/CamKinaseII/Tiam1/Rac1 pathway that substit

Metainflammation alters neutrophil function and migration in vivo in response to tissue injury.

Overnutrition and the consumption of Western-type diets lead to chronic low-grade systemic inflammation (ie metainflammation) and a dysfunctional immune response. Although neutrophils are affected by metainflammation, mechanistic evidence regarding the direct effects of dietary fat exposure on neutrophil function and migration in vivo, particularly in response to injury, remains limited. Here, we investigated how metainflammation induced by a high-cholesterol diet (HCD) influences neutrophil function and migration following tissue injury. We employed a tailfin transection model in juvenile zeb

Ocular toxoplasmosis: the role of retinal pigment epithelium migration in infection.

Our aim was to study the migration of retinal pigmented epithelium (RPE) into the retinal layer during infection of C57BL/6 mice with Toxoplasma gondii. Eyes from infected and non-infected animals were analyzed on the 60th day of infection by light and transmission electron microscopy. Non-infected eyes showed a normal morphology. In contrast, we observed free parasites in the retinal vasculature, the presence of mononuclear inflammatory infiltrate (MNII) and parasites in the vasculature of choroids in infected eyes. No inflammatory infiltrate was observed; RPE cells were identified near the M

Influence of transsphincteric fluid flow on spike burst rate of the opossum sphincter of Oddi.

In this study, we evaluated the effect of transsphincteric fluid flow on the rate of spike bursts in the opossum sphincter of Oddi (SO). For chronic studies in awake animals, bipolar electrodes were implanted on the SO, gastric antrum, duodenum, and jejunum. In group 1 animals, surgery was limited to electrode implantation. Subsequently, these animals underwent a cholecystectomy. Group 2 animals had electrode implantation without further surgery. In group 3 animals, bile flow was diverted around the SO, whereas in group 4 animals, pancreatic juice as well as bile was diverted around the SO. Mu

Neurons that migrate from the olfactory epithelium in the chick express luteinizing hormone-releasing hormone.

In several mammalian species, luteinizing hormone-releasing hormone (LHRH) neurons have been shown to migrate from nasal regions to the brain during early development. Using immunocytochemistry, we have identified LHRH containing neurons in developing chick embryos. In embryonic day 4 (E4) and E5 animals, a small group of LHRH immunoreactive (IR) neurons were found just ventral to the olfactory pit. LHRH-IR neurons were also found within the immunoreactive (IR) neurons were found just ventral to the olfactory pit. LHRH-IR neurons were also found within the nasal epithelium. In E6 and E7 animal

Cell-mediated immune responses to chlamydial antigens in guinea pigs injected with inactivated chlamydiae.

Cell-mediated immunity (CMI) to chlamydial antigens was readily induced in guinea pigs by a single injection of Betaprone-inactivated chlamydiae in complete Freund adjuvant. The CMI was measured in vivo by delayed hypersensitivity skin tests, and in vitro by inhibition of migration of peritoneal exudate cells and by proliferation of lymph node lymphocytes. There was an overall correlation between in vivo and in vitro responses. Of the in vitro assays, migration inhibition reflected the state of sensitization, as judged by skin tests, more uniformly than lymphocyte stimulation. Extensive inter-

Long-term influence of bone marrow-derived mesenchymal stem cells on liver ischemia-reperfusion injury in a rat model.

BACKGROUND: The aim of this study was to evaluate the long-term usefulness of intraportal injection of the bone marrow-derived mesenchymal stem cells (BM-MSCs) in limitation of experimentally induced ischemia-reperfusion injury (IRI) in a rat model. MATERIAL AND METHODS: Twenty Wistar rats were divided into 3 groups: donor group (n=5), study group (n=10), and control group (n=5). IRI was performed using a modified hanging-weight system after left portal triad occlusion in study group animals. Isolated autologous BM-MSCs were labeled with fluorochrome PKH-26 then intraportally injected into th

The molecular biology of furin-like proprotein convertases in vascular remodelling.

Vascular smooth muscle cell (VSMC) proliferation and migration represent key features in atherosclerosis and restenosis. The proprotein convertases (PCs) furin and PC5 are highly expressed in human atheroma and are putatively involved in vascular lesion formation via the activation of precursor proteins, essential for cell proliferation and migration. In vitro assays have identified these PCs to govern cell functions via endoproteolytic cleavage of key substrates, including pro-integrins and pro-matrix metalloproteinases. In vivo gene expression studies of furin/PC5 and their substrates demons

Endotoxin induced migration of leukocytes from blood to milk.

Guinea pigs were separated from young on days 5 to 7 of lactation. They were anesthetized with ether and were infused intramammary via the teat canal with either sterile saline (.5 ml) or Escherichia coli endotoxin (026:B6:500 microgram/.5 ml). Each animal served as its own control by having sterile saline in one gland and endotoxin in the other. Animals were sacrificed 4, 6, 8, and 12 h later to determine the time of maximum migration of polymorphonuclear leukocytes from blood to milk. A control group of animals having had no intramammary infusion was sacrificed. Tissues were prepared for obs
