In recent work, Parry et al. provide a detailed visualization of Microsporidia MB sporogeny (E. R. S. Parry, R. Pevsner, B. C. Poulton, D.-K. Purusothaman, et al., mSphere 10:e00851-24, 2025, https://doi.org/10.1128/msphere.00851-24). Their findings reveal that Microsporidia MB is localized to mosquito germline tissues and undergoes octosporogony within developing oocytes, suggesting both vertical and potential horizontal transmission routes. The identification of spores with hallmark microsporidian structures strengthens the case for environmental dissemination via egg deposition. These resul

Anopheles stephensi Liston, 1901 (Diptera: culicidae) is a competent vector of Plasmodium falciparum (Haemosporida: plasmodiidae) malaria, and its expansion in the African continent is of concern due to its viability in urban settings and resistance to insecticides. To enhance its genetic tractability, we determined the utility of a ~2 kb An. stephensi lipophorin (lp) promoter fragment in driving transgene expression. Lipophorin genes are involved in lipid transport in insects, and an orthologous promoter in An. gambiae (AGAP001826) was previously demonstrated to successfully express a transge…

Heterozygous carriers of haemoglobin S and C (HbAS and HbAC) have a reduced risk of severe malaria but are not protected from Plasmodium falciparum infection, suggesting that the protection involves acquired immunity. During a blood meal, female Anopheles mosquitoes inject saliva that can elicit a host antibody response, which can serve as a proxy for exposure to Plasmodium infection. Previous studies have shown that the peptide gSG6-P1 of An. gambiae saliva is antigenic and highly Anopheles specific. Here, we used plasma samples from 201 Ghanaian children with wild-type haemoglobin (HbAA), Hb…

BACKGROUND: Ivermectin (22,23-dihydroavermectin B1a: H2B1a) is an endectocide used to treat worm infections and ectoparasites including lice and scabies mites. Furthermore, survival of malaria transmitting Anopheles mosquitoes is strongly decreased after feeding on humans recently treated with ivermectin. Currently, mass drug administration of ivermectin is under investigation as a potential novel malaria vector control tool to reduce Plasmodium transmission by mosquitoes. A "post-ivermectin effect" has also been reported, in which the survival of mosquitoes remains reduced even after ivermect…

Current studies on Anopheles anticholinesterase insecticides are focusing on identifying agents with high selectivity towards Anopheles over mammalian targets. Acetylcholinesterase (AChE) from electric eel is often used as the bioequivalent enzyme to study ligands designed for activity and inhibition in human. In this study, previously identified derivatives of a potent AChE, donepezil, that have exhibited low activity on electric eel AChE were assessed for potential AChE-based larvicidal effects on four African malaria vectors; An. funestus, An. arabiensis, An. gambiae and An. coluzzii. This …

Anopheles funestus is one of the major malaria vectors in Africa. As with the other main vectors, insecticide resistance in this species threatens existing vector control strategies. Unfortunately, scientific investigations, which could improve understanding of this vector species or lead to the development of new control strategies, are currently limited by difficulties in laboratory rearing of the species. In an attempt to optimise laboratory-rearing conditions for An. funestus, the effect of an artificial blood-feeding system for adults, different larval diet doses, and a range of other rea…

BACKGROUND: Several studies carried out in Benin have shown the involvement of Anopheles gambiae sensu lato (s.l.), the Anopheles funestus group in malaria transmission, but none of them reported the contribution of the Anopheles nili group to the transmission of this disease. The current study investigated the question through an entomological cross-sectional survey performed in Northern Benin.
METHODS: Mosquito samplings were performed in September and October 2017 in 4 villages located in two districts: Bambaba and Wodara (Kérou district) and, Péhunco 2 and Béké (Péhunco district). The col…

Anopheles sinensis is an important malaria vector in China and other Southeast Asian countries, and the emergence of insecticide resistance in this mosquito poses a serious threat to the efficacy of malaria control programs. The recently published An. sinensis genome and transcriptome provide an opportunity to understand the molecular mechanisms of insecticide resistance. Analysis of the An. sinensis genome revealed 174 detoxification genes, including 93 cytochrome P450s (P450s), 31 glutathione-S-transferases (GSTs), and 50 choline/carboxylesterases (CCEs). The gene number was similar to that …

Anopheles gambiae s.s. Giles (Diptera: Culicidae), the primary African malaria vector, has been characterized at the subspecies level in Madagascar, where only the molecular form S and haplotype gIA occur. The haplotype gIC proposed by other authors was not observed amongst the 35 mosquito genomes sequenced. These S/gIA characteristics are also found on the Comoros archipelago and in continental Africa.

Spontaneous oxidation of 3-hydroxykynureine (3-HK), a metabolic intermediate of the tryptophan degradation pathway, elicits a remarkable oxidative stress response in animal tissues. In the yellow fever mosquito Aedes aegypti the excess of this toxic metabolic intermediate is efficiently removed by a specific 3-HK transaminase, which converts 3-HK into the more stable compound xanthurenic acid. In anopheline mosquitoes transmitting malaria, xanthurenic acid plays an important role in Plasmodium gametocyte maturation and fertility. Using the sequence information provided by the Anopheles gambiae…

Oviposition preference of the Australasian coastal malaria vector Anopheles farauti s.s. for water of varying salinity was determined in the laboratory to help understand the distribution and control of this species in the field. Numbers of eggs laid showed an inverse relationship with salinity; of 5 NaCl concentrations most eggs were laid in distilled water but some were laid in 3.17% NaCl (the salinity of seawater). The association of An. farauti with coastal areas occurs in spite of an aversion to salt water by ovipositing females. Factors other than salinity must be the primary determinant…

The utility of light traps for sampling Anopheles fluviatilis, the major malaria vector in the hill tracts of Koraput district, Orissa, was assessed. A modified version of the CDC miniature light trap was used in the study. The data generated from light trap collections were analysed and compared with daytime resting, man-biting and night resting collections carried out in parallel. The number per trap recorded in cattle sheds was significantly higher than that recorded in human dwellings. A similar pattern was observed in hand catches of indoor resting population though the difference was not…
