Motor and sensory centers for the innervation of mandibular and sublingual salivary glands: a horseradish peroxidase study in the dog.

Horeradish peroxidase was injected at multiple sites in the mandibular and sublingual salivary glands in order to label the preganglionic salivatory neurons in the brain stem. The same injections resulted in retrograde labeling of the sympathetic and sensory neurons that project to these glands. Labeled fusiform and multipolar salivatory neurons were found ipsilaterally in the lateral reticular formation of the medulla where they extended over the rostral four-fifths of the facial nucleus and the caudal one-third of the dorsal nucleus of the trapezoid body. The vast majority of the small and m

Autonomic neuropathy of the adrenal gland of rats after alloxan diabetes.

Six alloxan-diabetic male rats with blood glucose levels between 250 and 420% were sacrificed. Their adrenal glands were extracted and studied by Holmes technique and the modified Gross Bielschowsky stain. Degenerative changes in the nerve fibres ending in the cortex and those reaching the medulla were noticed together with chromatolysis of the chromaffin cells. These findings are similar to the autonomic neuropathies described by other workers in the digestive tract and urinary bladder. This work gives histological evidence of the biochemical findings of reduced plasma catecholamines in longs

Sphincter of Oddi pressure in chagasic patients with megaesophagus.

Autonomic denervation is found throughout the entire length of the digestive tract in Chagas' disease. Anatomic evidence of myenteric ganglia reduction in chagasic gallbladders has been noted; however, the sphincter of Oddi has not been studied. The purpose of this study is twofold: first, to determine sphincter of Oddi pressure in 11 patients with chronic Chagas' disease and megaesophagus, and to compare the results with those obtained in 27 control subjects; and second, to evaluate the effect of cholecystokinin-octapeptide on sphincter of Oddi pressure in both groups of patients. Sphincter o

Central origins of cranial nerve parasympathetic neurons in the rat.

The location of central neurons that contribute preganglionic parasympathetic axons to cranial nerves VII, IX, and X in rats has been identified using horseradish peroxidase (HRP) tracing methods. Collectively, these neurons form an uninterrupted dorsal column that extends over the entire length of the medulla. The cephalic end of this column turns ventrally with neurons scattered in the parvicellular reticular formation between the rostral pole of the nucleus of the solitary tract (NST) and the facial motor nucleus. Applying HRP crystals to the cut cervical vagus labels neurons in the classic

[A comparative study of the laparoscopic appearance and anatomy of the autonomic nervous in normal males].

OBJECTIVE: To further understand the anatomical basis of pelvic autonomic nerve preservation. METHODS: Autopsy of five adult male donated cadavers was performed. Meanwhile, ten videos of laparoscopic total mesorectal excision for male mid-low rectal cancer admitted from January to June 2012 were observed and studied. Anatomical features of pelvic autonomic nerve were compared between autopsy and laparoscopic appearance. RESULTS: Autopsy observations indicated that:the abdominal aortic plexus was situated upon the sides and front of the aorta, between the origins of the superior and inferior

Brain-to-stomach transfer of α-synuclein via vagal preganglionic projections.

Detection of α-synuclein lesions in peripheral tissues is a feature of human synucleinopathies of likely pathogenetic relevance and bearing important clinical implications. Experiments were carried out to elucidate the relationship between α-synuclein accumulation in the brain and in peripheral organs, and to identify potential pathways involved in long-distance protein transfer. Results of this in vivo study revealed a route-specific transmission of α-synuclein from the rat brain to the stomach. Following targeted midbrain overexpression of human α-synuclein, the exogenous protein was capable

Localization of preganglionic neurons of the accessory ciliary ganglion in the midbrain: HRP and WGA-HRP studies in the cat.

Localization of preganglionic neurons of the accessory ciliary ganglion (ACG), including ectopic intraocular ganglion cells, was investigated in the cat with the aid of horseradish peroxidase (HRP) and HRP-conjugated wheat germ agglutinin (WGA-HRP) methods. When HRP or WGA-HRP was injected into the anterior and posterior chambers of the eye, no retrogradely labeled cells were found in the visceral oculomotor nuclei, although most neurons of the ACG and the main ciliary ganglion (CG) were intensely labeled. When a microsyringe needle was inserted into the ciliary body, the tracer diffused into

The effects of delta-opioid receptor agonists on synaptic transmission in hamster submandibular ganglion.

The effects of agonists for delta-opioid receptors on synaptic transmission in hamster submandibular ganglion were studied in vitro, using an intracellular recording technique. [Met5]enkephalinamide (17 microM), [D-Ser2¿Leu-enkephalin-Thr (14 microM) and [D-Pen2, D-Pen5]enkephalin (10 microM), delta selective agonists caused reversible depression of the fast EPSP amplitude in 10 of 30 cells tested. The inhibitory actions of [D-Pen2, D-Pen5]enkephalin were antagonized by ICI 174,864, a delta selective antagonist. Naloxon a non-selective antagonist and ICI 174,864 both also potentiated the fast

Is the central inspiratory activity responsible for pCO2-dependent drive of the sympathetic discharge?

Out of 27 cats anesthetized with chloralose-urethane mixture, paralyzed, vagotomized and artificially ventilated, phrenic nerve response to systemic hypercapnia (7-8 vol.% CO2/O2 mixture) was accompanied by an increase in blood pressure and sympathetic discharge in 19 cats. Out of these 19 cats, 12 were totally debuffered and in the remaining 7 cats one carotid sinus nerve was left intact. Single unit activity in the sympathetic cervical nerve and spontaneous mass activity in the cervical, splanchnic, renal sympathetic and phrenic nerves were recorded. Evoked response in the phrenic nerve was

Can natural ways to stimulate the vagus nerve improve seizure control?

The vagus nerve (VN) is the longest cranial nerve, innervating the neck, thorax and abdomen, with afferent fibers transmitting a range of interoceptive stimuli and efferent fibres to somatic structures and autonomic preganglions. Over the last few decades, electrical stimulation of the VN using implanted devices (VNS) has been developed leading to its approval for the treatment of epilepsy and depression. More recently, non-invasive devices to stimulation the VN have been developed. The VN has many functions and the activity that is most amenable to assessment is its effect in controlling the

Transient changes in spinal cord glial cells following transection of preganglionic sympathetic axons.

Following peripheral nerve injury, retrograde signals originating from the injury site may activate intrinsic factors in the injured neurons, possibly leading to regenerative growth. Retrograde influences from peripheral injury sites may lead to the activation of glial cells in the vicinity of the centrally located cell bodies of the injured neurons. Few studies have examined changes in the spinal cord intermediolateral cell column (IML), which houses sympathetic preganglionic cell bodies, following injury to distal axons in the cervical sympathetic trunk (CST). The goal of the present study w

Effects of inhibitors of enzymatic and cellular pH-regulating systems on central sympathetic chemosensitivity.

Previous studies in cats using isolated NaCl-CO2 perfusion of the lower brainstem demonstrated an intrinsic chemosensitivity of sympathoexcitatory bulbospinal neurones within the rostroventrolateral medulla (RVLM). In the present experiments, the effects of inhibitors of enzymatic and cellular systems, known to be involved in pH regulation, were investigated. Isolated perfusion of the lower brainstem with CO2-enriched solutions was performed and preganglionic sympathetic nerve activity (SNA) was recorded. Drugs were locally injected into the left RVLM with glass micropipettes. Perfusion of the

The distribution of oxytocin- and neurophysin-stained fibers in the spinal cord of the rat and monkey.

Immunohistochemical methods have been used to describe the distribution of neurophysin I- and oxytocin-stained fibers in the spinal cord. In albino rats of either sex, such fibers descend through the dorsal part of the lateral funiculus to the caudal end of the cord. Fibers leave the lateral funiculus to innervate the marginal zone of the dorsal horn at all levels, and the intermediolateral column at thoracic, lumbar and sacral levels. Some fibers course medially through the intermediate gray to end in the central gray, which is innervated at all levels, and also appears to be innervated by fi

Targeted ablation of cardiac sympathetic neurons reduces the susceptibility to ischemia-induced sustained ventricular tachycardia in conscious rats.

The Cardiac Arrhythmia Suppression Trial demonstrated that antiarrhythmic drugs not only fail to prevent sudden cardiac death, but actually increase overall mortality. These findings have been confirmed in additional trials. The "proarrhythmic" effects of most currently available antiarrhythmic drugs makes it essential that we investigate novel strategies for the prevention of sudden cardiac death. Targeted ablation of cardiac sympathetic neurons may become a therapeutic option by reducing sympathetic activity. Thus cholera toxin B subunit (CTB) conjugated to saporin (a ribosomal inactivating

Paradoxical effects of continuous high dose gabapentin treatment on autonomic dysreflexia after complete spinal cord injury.

Spinal cord injury (SCI) can have profound effects on the autonomic and cardiovascular systems, notably with injuries above high-thoracic levels that result in the development of autonomic dysreflexia (AD) characterized by volatile hypertension in response to exaggerated sympathetic reflexes triggered by afferent stimulation below the injury level. Pathophysiological changes associated with the development of AD include sprouting of both nociceptive afferents and ascending propriospinal 'relay' neurons below the injury, as well as dynamic changes in synaptic inputs onto sympathetic preganglion

Potentiation of NMDA currents by pituitary adenylate cyclase activating polypeptide in neonatal rat sympathetic preganglionic neurons.

Whole cell patch-clamp recordings were made from sympathetic preganglionic neurons (SPNs) in the intermediolateral cell column of thoracolumbar spinal cord slices of 12- to 16-day-old rats, and the effects of pituitary adenylate cyclase activating polypeptide (PACAP)-38 on N-methyl-D-aspartate (NMDA)- and kainate (KA)-induced inward currents were examined. PACAP, in concentrations (10-30 nM) that caused no significant change of holding currents, reversibly increased NMDA-induced currents but not KA-induced currents. At higher concentrations (>30 nM), the peptide produced a sustained inward cur

Selected changes in spinal cord morphology after T4 transection and olfactory ensheathing cell transplantation.

Spinal cord transection at T4 results in severe damage of the nervous tissue, with impairment of motor, sensory and autonomic functions. Transplantation of olfactory ensheathing cells (OECs) has the potential to improve these functions through a number of mechanisms, which include facilitation of regeneration and neuroprotection. For cardiovascular functions, we have previously shown that OECs reduce the duration of autonomic dysreflexia, without evidence of regeneration. To further understand the mechanisms underpinning this improvement, we have studied changes in selected morphological featu
