Soma-germ contact across the basement membrane in the ovary.

Epithelial cells interact with other cells and environments at their apical side, while the basement membrane typically impedes such interaction at the basal surface. One notable instance is communication between soma and germ cells within the ovary across numerous bilaterian taxa. This contact underlies proper oogenesis and subsequent embryogenesis. Throughout the history of morphology and cell biology, there has been an emphasis on this heterocellular interaction primarily occurring at the apical side of epithelial cells. Contrary to this long-standing understanding, we uncover that ovarian …

Efficacy and Safety of Rituximab in Antiglomerular Basement Membrane Disease.

INTRODUCTION: Anti-glomerular basement membrane (GBM) disease is caused by pathogenic antibodies usually targeting the noncollagenous domain of the α3 chain of type IV collagen and frequently presents as rapidly progressive glomerulonephritis and diffuse alveolar hemorrhage (DAH). Rapid reduction of these antibodies is imperative for kidney survival and the mainstay of therapy is the combination of plasma exchange (PLEX), glucocorticoids, and cyclophosphamide. Rituximab has been postulated as a potential treatment for anti-GBM disease; however, data on efficacy and safety are lacking. METHODS…

Defining invasion in breast cancer: the role of basement membrane.

Basement membrane (BM) is an amorphous, sheet-like structure separating the epithelium from the stroma. BM is characterised by a complex structure comprising collagenous and non-collagenous proteoglycans and glycoproteins. In the breast, the thickness, density and composition of the BM around the ductal lobular system vary during differing development stages. In pathological conditions, the BM provides a physical barrier that separates proliferating intraductal epithelial cells from the surrounding stroma, and its absence or breach in malignant lesions is a hallmark of invasion and metastases.…

Thickening of the basement membrane as a diagnostic sign of mycosis fungoides.

The epidermal basement membrane (BM) is readily identified on skin biopsy specimens stained with periodic acid-Schiff (PAS) and PAS with diastase (PAS-D). Thickening of BM can be evidenced in several inflammatory and tumoral conditions. We noticed that most of our biopsy specimens of mycosis fungoides (MF) showed thickening of the BM. We decided to retrospectively study BM thickness in 27 biopsy specimens of MF and compare them with 27 cutaneous biopsy specimens of inflammatory diseases. We studied PAS and PAS-D stains in all cases and we measured BM thickness with an ocular micrometer. Cases …

Ultrastructural characterization of damage in the basement membrane of facial melasma.

The pathogenesis of melasma is not fully understood, and the role of skin basement membrane zone (BMZ) alterations in disease development and the maintenance of hypermelanogenesis are also poorly known. We performed a comparative study to characterize the ultrastructural alterations that occur in BMZ in melasma and adjacent normal skin, as well as we discuss the implications of these changes in the physiopathology of the disease. Pairs of facial skin biopsies (2 mm) from 10 women with melasma and normal skin (< 2 cm apart) were processed by Transmission Electronic Microscopy or immunohistochem…

The presence of thin glomerular basement membranes in various glomerulopathies.

Results of 61 cases of various glomerulopathies with thin glomerular basement membranes are presented. The largest group of 31 cases consisted of mesangial glomerulonephritis. The second largest group consisted of 19 patients with small glomerular lesions described as non-specific. This group stood out in both clinical presentations and in the higher diversity of lesions within the lamina densa of the basement membrane. The results of measurements of the lamina densa in various glomerulopathies were compared to those obtained in control groups consisting of thin basement membrane syndrome and …

Retinal basement membrane abnormalities and the retinopathy of Alport syndrome.

PURPOSE: To determine the effects of X-linked and autosomal recessive Alport syndrome on retinal basement membranes and how these result in the characteristic perimacular dot-and-fleck retinopathy, lozenge, and macular hole. METHODS: The type IV collagen chains present in the normal retina were determined immunohistochemically. Ten patients with Alport syndrome underwent retinal photography and optical coherence tomography to determine the thickness of the internal limiting membrane (ILM) by segmentation analysis, the layers affected by the retinopathy, and any correlates of the lozenge and m…

Incidence and outcome of antiglomerular basement membrane disease in Chinese.

BACKGROUND: Antiglomerular basement membrane (anti-GBM) disease is an uncommon disease, especially among Asian population. Many reports and studies on this condition in the Caucasian population are available, but little information exists on anti-GBM disease in Asians. To study the incidence and clinical characteristics of anti-GBM disease among Chinese patients, we reviewed our experience of anti-GBM disease in our hospital (Queen Mary Hospital, Hong Kong) from 1992 to 2003. METHODS: All patients who were admitted for acute renal impairment, which was caused by crescentic glomerulonephritis …

Human skin basement membrane in health and in autoimmune diseases.

Skin basement membrane zone (BMZ) is an ultrastructurally defined area situated between the outer layer of skin, the epidermis, and the inner layer of skin, the dermis. The major function of skin BMZ is to serve as an adherent connection between the epidermis and the dermis. Heritable skin diseases characterized by genetic mutations that result in defective BMZ protein production, such as junctional and dystrophic epidermolysis bullosa, manifest clinically as skin fragility and subepidermal blister formation inducible by minor trauma. Many distinct, yet interconnected BMZ components, have been…

Microangiopathy, the vascular basement membrane and Alzheimer's disease: a review.

The present review focuses on the vascular basement membrane (VBM) and its relationship to the lesions of Alzheimer's disease (AD). Examination of the fine structure of the microvasculature reveals AD-associated VBM alterations, which include both thickening and vacuolization. Immunocytochemistry confirms that all three intrinsic VBM components [collagen type IV, laminin, and heparan sulfate proteoglycan (HSPG)] outline the capillary bed, which is pathologically altered in AD patients (microangiopathy). Ultrastructural analyses of AD tissue samples demonstrate that HSPG's normal staining patte…

Gingival basement membrane antigens in desquamative lesions of the gingiva.

Desquamative gingival lesions are a clinical manifestation of several entities where histologic alterations in the gingival basement membrane are frequent. Normal and lesional gingival specimens from six patients with desquamative lesions of the gingiva associated with cicatricial pemphigoid or lichen planus and from one normal subject were examined by indirect immunofluorescence for immunologic alterations in basement membrane antigens. In four out of five desquamative gingival lesions from patients with lichen planus, type IV collagen and EBA basement membrane antigens were interrupted, thic…

The use of monoclonal antibodies in skin basement membrane research.

Recent experiments have indicated that it is possible to uncover new antigens within both the basement membrane zone and the epidermis of normal adult human skin by producing murine anti-human monoclonal antibodies following immunization with human skin preparations. Already one such monoclonal antibody has defined a biochemical defect that may be important in the pathogenesis of one of the more severe blistering skin diseases, dystrophic epidermolysis bullosa. It is likely that further attempts at hybridoma production using basement membrane extracts of skin will lead not only to the identifi…

Epithelial and mesothelial basement membranes in diabetic patients and dogs.

The influence of diabetes on the thickness of basement membrane of the ciliary epithelium, lens epithelium, and corneal mesothelium (endothelium) was investigated post mortem in eyes from 27 human subjects and 30 dogs. Ten of the human subjects were diabetic, 10 were non-diabetic matched for age and sex, and seven were non-diabetic subjects who had been diagnosed as hypertensive 2-8 years before death. The dogs comprised three prospectively identified groups: 10 were alloxan diabetic for 5 years and kept in poor glucose control, 10 were alloxan diabetic for 5 years and kept in good glucose con…

Characterization of glomerular basement membrane components within pediatric glomerular diseases.

BACKGROUND: Disruptions in gene expression associated with the glomerular basement membrane (GBM) could precipitate glomerular dysfunction. Nevertheless, a comprehensive understanding of the characterization of GBM components within pediatric glomerular diseases and their potential association with glomerular function necessitates further systematic investigation. METHODS: We conducted a systematic analysis focusing on the pathological transformations and molecular attributes of key constituents within the GBM, specifically Collagen IV α3α4α5, Laminin α5β2γ1, and Integrin α3β1, across prevale…

A case report of atypical anti-glomerular basement membrane disease.

BACKGROUND: Anti-glomerular basement membrane (anti-GBM) disease is characterized by crescentic necrotizing glomerulonephritis, with linear deposits of immunoglobulin G (IgG) in the GBM. Classic anti-GBM disease is clinically associated with rapidly progressive glomerulonephritis with or without pulmonary hemorrhage. Some patients have a better renal prognosis and milder symptoms than those with classic anti-GBM disease, which is termed atypical anti-GBM disease. CASE PRESENTATION: A 43-year-old Japanese woman was admitted to our hospital complaining of hematuria that had persisted for more t…

Anti-glomerular basement membrane disease in children: a brief overview.

Anti-glomerular basement membrane disease (Anti-GBM), previously known as Goodpasture syndrome, is an extremely rare cause of rapidly progressive glomerulonephritis and chronic kidney disease stage 5 (CKD5) in children. It is associated with acute pulmonary haemorrhage and it has a poor prognosis. It is classified as an autoimmune, small-vessel vasculitis caused by autoantibody formation against the alpha-3 chain in type IV collagen found in the glomerular basement membrane. Evidence of anti-GBM antibodies in serum or histologically are required for diagnosis. Treatment in children is based on…

Human Cytomegalovirus Interactions with the Basement Membrane Protein Nidogen 1.

In 2000, we reported that human cytomegalovirus (HCMV) induced specific damage on chromosome 1. The capacity of the virus to induce DNA brea … via two separate pathways highlighted the virus' emphasis on NID1 elimination. NID1 is an important basement membrane protein secreted by many cell types, including the endothelial cells (ECs) lining the vasculature. We found that ss NID1 was also reduced in infected ECs and hypothesized that virus-induced removal of NID1 might offer HCMV a means of increased distr …

[Review on anti-glomerular basement membrane disease or Goodpasture's syndrome].

Anti-glomerular basement membrane (anti-GBM) disease or Goodpasture's syndrome is a small vessel vasculitis affecting the capillary beds of kidneys and lungs. It is an autoimmune disease mediated by autoantibodies targeting the glomerular and alveolar basement membranes, leading to pneumorenal syndrome. It is a rare, monophasic and severe disease, associating rapidly progressive glomerulonephritis and alveolar hemorrhage. The presence of antineutrophil cytoplasmic antibodies (ANCA) is reported in 20 to 60% of cases. Management should be prompt and combine plasma exchange with systemic corticos…
