Recovery of noise-induced changes in the dark cells of the quail tegmentum vasculosum.

Morphologic changes in the tegmentum vasculosum (TV) of adult quail after high intensity sound exposure were studied. Quail were continuously exposed to 115 dB SPL, 1500 Hz pure tone in a sound field for 12 h and either sacrificed immediately (0 day), 1, 2, 3, 4, 6 or 10 days later. Serial sections through the basilar papilla at 100 micron intervals from base to apex were obtained for study with light microscopy and TEM. Significant morphologic changes were found within the TV of quail sacrificed on days 0-4. On a quantitative scale, the majority of recovery occurred within the first 24 h. Aft

Altered membrane properties of cerebral vascular smooth muscle following subarachnoid hemorrhage: an electrophysiological study. I. Changes in resting membrane potential (Em) and effect on the electrogenic pump potential contribution to Em.

Subarachnoid hemorrhage was produced experimentally in cats by intracisternal injection of non-heparinized autologous arterial blood obtained by cardiac puncture under ketamine and xylazine anesthesia. Cats were sacrificed at varying time intervals between 30 min and 7 days post ictus. Measurements of resting membrane potential were recorded from smooth muscle cells of the basilar artery. These measurements were obtained by impalement from the adventitial surface of isolated but otherwise intact segments of the artery using glass microelectrodes with tip sizes less than 0.1 micron. The resting

Middle and inner ear modelling: from microCT images to 3D reconstruction and coupling of models.

We present finite element (FE) modeling approaches of ear mechanics including 3-dimensional (3D) reconstruction of the human middle and inner ear. Specifically, we demonstrate a semi-automatic methodology for the 3D reconstruction of the inner ear structures, a FE harmonic response model of the middle ear to predict the stapes footplate frequency response, a 2D FE slice model of the cochlea for the coupled response at the micromechanical level for either acoustic or electrical excitation and a coupled FE middle ear model with a simplified cochlea box model to simulate the basilar membrane velo…

Type II collagen-induced inner ear disease: critical evaluation of the guinea pig model.

Guinea pigs systematically challenged with type II collagen were examined electrophysiologically, histologically, and immunologically for inner ear lesions. Some functional, morphologic, and immunologic changes could be demonstrated. Evidence of possible immunoglobulin deposition in the basilar membrane, the perivascular region, as well as in other inner ear tissue were present. Immunologic and histopathologic simulations to inner ear diseases in humans and to the inner ear antigen-induced animal model were found. The results are discussed in light of the original model introduced by Yoo et al…

Surgical excision of meningiomas involving the clivus: preoperative and intraoperative features as predictors of postoperative functional deterioration.

Neurological deterioration is commonly seen after surgical excision of clival meningiomas; however, an understanding of the risk factors associated with postoperative deterioration can lead to improvements in outcome. In 75 patients with clival meningiomas operated on over a 7-year period, the following data were studied; preoperative variables such as presenting Karnofsky scale score, age, sex, and prior operations or radiation therapy. Radiological findings on magnetic resonance imaging or arteriography, such as the development of the arachnoidal cleavage plane between tumor and the brain st

Role of plasmin, thrombin, and antithrombin III as etiological factors in delayed cerebral vasospasm.

The fibrinolytic enzyme plasmin at 0.25 units/ml produced a contraction of isolated canine basilar arteries that developed slowly and was sustained for at least 2 hours. Plasmin and thrombin (1 unit/ml) acted synergistically to enhance the contractile response. In contrast to plasmin, the marked contraction elicited by thrombin ended within 1 hour, and afterward the artery was completely tachyphylactic to thrombin. Fibrin clot, fibrinopeptides, and fibrin degradation products did not prolong significantly the effect of thrombin or prevent the tachyphylaxis. Plasmin and thrombin may occupy a co

Autoimmune aspects of pemphigus vulgaris and mucosal pemphigoid.

Oral mucosal lesions of ulcerative, desquamative, and bullous character are major problems for clinicians and patients alike. Underlying such processes is the autoimmune reaction manifesting as specific subsets of immunopathology. Responsible for the maintenance of mucosal integrity within the oral cavity are numerous adhesion proteins which operate at the cell-to-cell and cell-to-connective-tissue-matrix level. Two of the better-understood conditions characterized by altered or disordered levels of adherence are the pemphigus and the pemphigoid groups of lesions. Autoantibodies in pemphigus v

The Greenwood function shows close alignment with pitch perceived by cochlear implant patients with long, flexible electrode arrays and fine-structure stimulation.

INTRODUCTION: The natural, tonotopic frequency distribution of the inner ear is typically described by the Greenwood function, which logarithmically projects the audible frequency spectrum onto the intracochlear basilar membrane. Recent developments in cochlear implant (CI) programming aim to improve sound quality and music perception through consideration of the frequency distribution as described by the Greenwood function when assigning frequency bands to the individual contacts of the electrode array. This approach is commonly referred to as anatomy-based fitting (ABF). However, empirical v…

Finite element model of the stapes-inner ear interface.

Since 1958, stapedotomy has been the method of choice for middle ear surgeons who operate on patients suffering from otosclerosis, especially stiffening of the interface between the stapes footplate of the middle ear and the oval window, which is a part of the cochlea of the inner ear. Later, many surgeons started to use the Schuknecht prosthesis, which consists of cartilage and is inserted into the complete opened oval window during stapedectomy. Our study shows that basilar membrane (BM) displacement is increased with an increasing stapes footplate area by a numerical simulation including th…

Trauma to the inner ear in birds, due to acoustic overstimulation or ototoxic aminoglycosides, can lead to hair cell loss which is followed … ell population, mainly so-called short hair cells residing on the abneural mobile part of the avian basilar membrane. Uncoupling of the tectorial membrane from the hair cells during sound overexposure may serve as a protection mechanism. The rapid functional recovery after sound trauma appears not to be associated with regeneration of the lost hair cells, but with …

Effects of endolymphatic and perilymphatic application of salicylate in the pigeon. II: Fine structure of auditory hair cells.

Large doses of salicylate are known to cause reversible ototoxic effects including fine structural alterations of the auditory hair cells in mammals. To investigate possible fine structural correlates of salicylate effects on pigeon auditory hair cells, the basilar papillae following perilymphatic or endolymphatic application of salicylate were fixed and processed for transmission electron microscopy. The pigeon auditory hair cells possessed organelles typically described in avians. A single or multi-layered array of cisternae along the cytoplasmic side of the lateral plasma membrane, i.e. sub

Temporal sequence of viral antigen expression in the cochlea induced by cytomegalovirus.

Injection of guinea-pig cytomegalovirus into seronegative guinea pig cochleas results in viral labyrinthitis. The location of infected cells 1-8 days following inoculation was examined using an immunohistochemical assay for viral antigens. Mesothelial cells below the basilar membrane and those lining the scala tympani, especially below the osseous spiral lamina, were the first cells to express antigen. This occurred 2 days following inoculation. Infection then spread rapidly toward the brain and through the perilymphatic duct to involve inflammatory cells, mesothelial cells of Reissner's membr…

Nonlinear response to a click in a time-domain model of the mammalian ear.

In this paper, a state-space implementation of a previously developed frequency-domain model of the cochlea is coupled to a lumped parameter model of the middle ear. After validation of the time-domain model by comparison of its steady-state response to results obtained with a frequency-domain formulation, the nonlinear response of the cochlea to clicks is investigated. As observed experimentally, a compressive nonlinearity progressively develops within the first few cycles of the response of the basilar membrane (BM). Furthermore, a time-frequency analysis shows that the instantaneous frequen…

Coupled oscillator mechanism extends the amplifying effect of outer hair cells to high frequencies.

Outer hair cells (OHCs) are essential for the sensitivity and frequency specificity of the mammalian ear. To perform this function, OHCs need to amplify the motion of the basilar membrane, which is much stiffer than themselves. OHCs must overcome this impedance mismatch for their amplifying function particularly at high frequencies, where the mismatch is the largest. This issue could be solved by the existence of multiple modes of motion. Here, systems of two coupled oscillators are examined as the simplest of such cases. It is found that some of these model systems indeed make OHCs function a…

Molecular profile of vascular ion channels after experimental subarachnoid hemorrhage.

Cerebral vasospasm is a transient, delayed constriction of cerebral arteries that occurs after subarachnoid hemorrhage (SAH). Smooth muscle cells show impaired relaxation after SAH, which may be caused by a defect in the ionic mechanisms regulating smooth muscle membrane potential and Ca(2+) permeability. We tested this hypothesis by examining changes in expression of mRNA and protein for ion channels in the basilar arteries of dogs after SAH using quantitative real-time polymerase chain reaction (PCR) and western blotting. SAH was associated with a significant reduction in basilar artery diam
