Towards toxin PEGylation: The example of rCollinein-1, a snake venom thrombin-like enzyme, as a PEGylated biopharmaceutical prototype.

PEGylation was firstly described around 50 years ago and has been used for more than 30 years as a strategy to improve the drugability of biopharmaceuticals. However, it remains poorly employed in toxinology, even though it may be a promising strategy to empower these compounds in therapeutics. This work reports the PEGylation of rCollinein-1, a recombinant snake venom serine protease (SVSP), able to degrade fibrinogen and inhibit the hEAG1 potassium channel. We compared the functional, structural, and immunogenic properties of the non-PEGylated (rCollinein-1) and PEGylated (PEG-rCollinein-1)

Charge variant analysis of protein-based biopharmaceuticals using two-dimensional liquid chromatography hyphenated to mass spectrometry.

The profile of charge variants represents an important critical quality attribute of protein-based biopharmaceuticals, in particular of monoclonal antibodies, and must therefore becontrolled. In this work, 2D-LC methods for charge variant analysis were developed using a strong cation-exchange chromatography (SCX) as first dimension (1D) separation. Non-porous SCX (3 µm) particle columns and different mobile phases were evaluated using a test mixture of some standard proteins of different size and pI (comprising myoglobin, bovine serum albumin, cytochrome c, lysozyme and β-lactoglobulin) and tw

Biopharmaceutical Monotargeting versus 'Universal Targeting' of Late-Onset Alzheimer's Disease Using Mixtures of Pleiotropic Natural Compounds.

A five-year close reading of the scientific literature on late-onset Alzheimer's disease (AD) has prompted the invention of a novel therapeutic method that biomechanistically targets the targetable disease-process targets of AD with one or another mixture of non-toxic pleiotropic natural compounds. The featured mixture herein is comprised of curcumin, resveratrol, and EGCG. The mixture's targets include central pathological elements of AD (including amyloid, tau, synaptic dysfunction, oxidative stress, mitochondrial dysfunction, and aberrant neuroinflammation), modifiable risk factors, comorbi

Medium-chain fatty acid-enabled nanostructured lipid carriers for improved β-carotene biopharmaceutical properties and antioxidant efficacy.

OBJECTIVE: This study aimed to enhance the in vitro digestion and permeability of β-carotene (BC), a lipophilic antioxidant with therapeutic potential in oxidative stress-related disorders, by developing a nanostructured lipid carrier (NLC) system based on medium-chain fatty acids (MCFAs). SIGNIFICANCE: Although BC exhibits strong antioxidant activity, its poor solubility and low gastrointestinal absorption limit clinical use. This study addresses these barriers by formulating a novel medium-chain fatty acid-based NLC using ethanol injection method that improves BC's solubility, permeability,

Triamcinolone acetonide loaded-cationic nano-lipoidal formulation for uveitis: Evidences of improved biopharmaceutical performance and anti-inflammatory activity.

Topical administration of corticosteroids is the cornerstone treatment of anterior uveitis, but poor corneal penetration and retention cause hindrance in their therapeutic utility. The conventional eye drops are less valuable in conditions where inflammation reaches deeper regions of the eye. Therefore, there is a clear need for an effective drug delivery system, which can increase corticosteroid penetration after topical application. To address this, cationic nanostructured lipid carriers of the drug triamcinolone acetonide (cTA-NLC) were prepared. The cTA-NLC were prepared by a hot microemul

Nose-to-brain delivery of empagliflozin-loaded nanostructured lipid carriers incorporated in-situ gel: Biopharmaceutical evaluation for Alzheimer's disease.

Alzheimer's therapy remains limited by poor drug targeting and multifactorial pathology. The therapeutic potential of SGLT-2 inhibitors like empagliflozin (EGZ) is constrained by poor brain bioavailability. Current study investigates the potential of EGZ-nanostructured lipid carrier (ENLC) for brain delivery via nasal route. The ENLC were prepared using hot melt emulsification technique followed by probe sonication and optimized using Box-Behnken design. ENLC were incorporated into poloxamer 407-chitosan in situ gel (ENPCG) to improve nasal retention, controlled release, and direct brain trans

Integrated analytical characterization and multiscale modeling of supramolecular interactions in lumefantrine-curcumin coamorphous systems with enhanced biopharmaceutical and anticancer performance.

Poor aqueous solubility, limited physical stability of amorphous forms, and suboptimal systemic absorption remains major challenges for lipophilic small molecule drugs. In this study, lumefantrine (LMF) and curcumin (CUR) were engineered into stoichiometry-controlled coamorphous systems (CAMS) using an interaction-driven solid-state design strategy. Integrated analytical characterization using powder X-ray diffraction, differential scanning calorimetry, and attenuated total reflectance Fourier transform infrared spectroscopy confirmed the formation of homogeneous single-phase coamorphous matri

Biopharmaceutical Characterization of Grapiprant Within the BCS Framework Under Canine-Relevant Conditions: Solubility and Caco-2 Permeability Assessment.

Background: The Biopharmaceutics Classification System (BCS) classifies drug substances according to their aqueous solubility and intestinal permeability; however, its application to veterinary drugs should account for species-specific gastrointestinal physiology. Grapiprant is a selective prostaglandin E2 receptor subtype 4 (EP4) antagonist approved as an oral tablet to control pain and inflammation associated with osteoarthritis in dogs, but its properties within the BCS framework remain unclear. Methods: This study evaluated the equilibrium solubility of grapiprant across pH conditions rele…

Quantitative and Qualitative Evaluation of Microorganism Profile Identified in Bioburden Analysis in a Biopharmaceutical Facility in Brazil: Criteria for Classification and Management of Results.

Microbiological contamination may cause microbial proliferation and consequently additional problems for pharmaceutical companies through production stoppage, product contamination, investigations of process deviations, out-of-specification results, and product disposal. This is one of the major concerns of the regulatory health agencies. Microbiological load (bioburden) may represent a potential risk for patients if the sterilization process is not effective and/or due to the production of toxins. Although bioburden can be eliminated by terminal sterilization or filtration processes, it is im

Biopharmaceutical, preclinical pharmacokinetic and pharmaco-dynamic investigations of an orally administered novel 3-nbutylphthalide prodrug for ischemic stroke treatment.

Ischemic stroke (IS) has been contributing in leading causes of disability and death worldwide and the cases are still increasing. In China, naturally sourced compound 3-n-butylphthalide (NBP) is widely applied in clinical practice for IS treatment with established evidences of efficacy and safety. However, NBP is an oily liquid at room temperature and has no active brain targeting ability, quite limiting its broader application in clinical practice. Via intravenous injection (i.v.) a prodrug compound (DB1) we previously developed deriving from NBP had dramatically enhanced the pharmacological

Intestinal perfusion with mesenteric blood sampling in wild-type and knockout mice: evaluation of a novel tool in biopharmaceutical drug profiling.

In the present study, we successfully downscaled, for the first time, the in situ intestinal perfusion technique with mesenteric blood sampling from rat to mouse. To evaluate the feasibility of this approach, we assessed the apparent permeability (P(app)) of mouse intestine for a set of marker compounds [atenolol, paracellular transport; metoprolol, transcellular transport; talinolol, P-glycoprotein (P-gp)-mediated efflux] in both wild-type and P-gp-deficient mice. In wild-type mice, the observed P(app) values for atenolol (1.8 +/- 0.3 x 10(-6) cm/s) and metoprolol (50.2 +/- 20.1 x 10(-6) cm/s

High-performance liquid chromatographic determination of bupivacaine in plasma samples for biopharmaceutical studies and application to seven other local anaesthetics.

A sensitive analytical procedure for bupivacaine dosing in plasma samples by reversed-phase high-performance liquid chromatography is described. After a two-step extraction, the analysis was performed using a C18 column and a mobile phase of 0.01 M sodium dihydrogen-phosphate (pH 2.1)-acetonitrile (80:20, v/v). The extraction yield of bupivacaine from plasma was 73.5 +/- 5.1% (mean +/- S.D., n = 10). The within-day and between-day reproducibilities at a concentration of 100 ng/ml were 2.1% and 5.6%, respectively (n = 10). Calibration curves were linear (r2 = 0.9996) between 5 and 1000 ng/ml. T

Microphysiological modeling of the Blood-CSF barrier for biopharmaceutical transport and sequestration: evaluation of PEG-induced accumulation and vacuolation in the choroid plexus epithelium.

BACKGROUND: It has long been established that the central nervous system (CNS) is a highly privileged space, with the blood-brain barrier (BBB) acting as the gatekeeper that allows or denies access to the brain by nutrients, drugs, and toxins. The BBB, however, is not the only barrier at play. The barrier between blood and cerebrospinal fluid (CSF) also has a critical role in guarding and feeding the CNS, but despite its importance, the blood-CSF barrier (BCSFB) remains relatively unexplored compared to the BBB, particularly with regard to models that do not rely on animals. METHODS: Given th

Algal Surface-Inspired Chitosan-Modified Monodisperse Porous Polystyrene Microspheres: Pore Size Regulation and Adsorption Mechanism for Biopharmaceutical Wastewater.

Monodisperse porous polystyrene microspheres (PPS) and their derivatives were successfully fabricated via a two-step swelling polymerization approach combined with secondary cross-linking technology. Inspired by the surface morphology of algae, PPS and derivatives were subsequently modified using chitosan with different viscosities (CS-1 and CS-2), thereby constructing chitosan-functionalized PPS. By precisely adjusting the amounts of vinyl chloroacetate (VCA) and the cross-linker 1,4-bis(chloromethyl)benzene (XDC), the pore size of PPS and derivatives could be finely tuned within the range of

Biopharmaceutical studies on drug/conjugated-metabolite interactions. III. Effect of acetaminophen sulfate and its positional isomers on the pharmacokinetics of acetaminophen in rats.

The effect of three positional isomers, o-, m- and p-acetylaminophenyl sulfate (AOAPS, AMAPS and APAPS (acetaminophen sulfate), respectively), on the pharmacokinetics of acetaminophen (APAP) was investigated in rats. All of the intravenously administered positional isomers were rapidly eliminated from plasma, and approximately 80% of the dose was excreted in an unchanged form in the urine within 4 h, while biliary excretions represented a small percent of the doses. Following the intravenous bolus injection of APAP, plasma elimination of APAP was accelerated and the distribution volume of APAP
