Occupational exposure to cadmium and kidney dysfunction.

Investigations were carried out in an alkaline battery factory. The study group consisted of 102 persons and the control group of 85 persons. Cadmium in blood (Cd-B) and cadmium in urine (Cd-U), as well as beta 2-microglobulin (B2-M), retinol binding protein (RBP), amino acids in urine were determined. Exposure to cadmium was high; Cd-B and Cd-U concentrations were higher than recommended, 10 micrograms/l and 10 micrograms/g creat. In 65% and 56% of workers, respectively. Excretion of B2-M and RBP in urine was higher than the accepted upper limits of 380 and 130 micrograms/g creat. in about 20…

Biological indicators of cadmium exposure and toxicity.

The increasing environmental and occupational exposure of populations to cadmium creates the need for biological indicators of cadmium exposure and toxicity. The advantages and disadvantages of monitoring blood cadmium, urinary, fecal, hair, and tissue cadmium, serum creatine, beta 2-microglobulin, alpha 1-anti-trypsin and other proteins, and urinary amino acids, enzymes, total proteins, glucose, beta 2-microglobulin, retinol-binding protein, lysozyme, and metallothionein are discussed. It is concluded that urinary cadmium, metallothionein and beta 2-microglubulin may be used together to asses…

Effects of acetaminophen on cadmium metabolism in mice.

Acetaminophen (ACM) administration to mice of the (C57BL/6 X DBA/2)F1 strain produced a typical hepatic centrilobular necrosis similar to that observed in rodents and humans. To determine the effects of this drug-induced necrosis on cadmium (Cd) metabolism, mice were given a sublethal dose of CdCl2 . 2.5 H2O containing 109CdCl2 and maintained for a period of time sufficient for Cd-metallothionein (Cd-MT) to be synthesized and distributed. Subsequent administration of ACM ip or po evoked a marked redistribution of Cd from livers to kidneys of mice, and increased the amount of Cd excreted in uri…

Chelation of cadmium with BAL and DTPA in rats.

Cadmium is unique among non-essential metals in several of its toxicological effects because of its long biological half life, slow excretion and delayed action on the kidneys. Although cadmium poisoning in humans is uncommon, there are reports of chronic cadmium poisoning in workmen and also in others in certain polluted areas. At present there are no suitable methods either to measure the body burden of cadmium or to treat cadmium poisoning. The therapeutic effects of various chelating agents including 2,3-dimercaptopropanol (BAL) and its soluble glycosides have been studied without much suc…

Overexpression of Tetrahymena Cysteine Synthetase 1 Promotes Cadmium Removal by Biosynthesizing Cadmium Sulfide Quantum Dots in Escherichia coli.

Heavy metal cadmium causes significant contamination in aquatic ecosystems. The biomineralization of cadmium represents a vital biological mechanism for handling cadmium stress in diverse microorganisms. To improve the biomineralization capacity of cadmium by microorganisms in aquatic environments, Tetrahymena cysteine synthetase 1 (TtCsa1) was overexpressed in E. coli. The tolerance of E. coli/pET-28a-TtCSA1 to cadmium was enhanced by expressing TtCsa1. Upon addition of cysteine, E. coli/pET-28a-TtCSA1 generated more H2S, which reacted with Cd2+ to form CdS quantum dots (QDs), resulting in a …

Three-year assessment of cadmium exposure and bone mineral density changes in cadmium-contaminated areas in northwestern Thailand.

Chronic cadmium exposure is linked to bone loss, but its effect on the short-term progression of bone mineral density (BMD) in previously exposed populations is not well understood. This study aimed to investigate the association between baseline urinary cadmium (U-Cd) levels and the subsequent three-year change in BMD in a chronically exposed cohort in northwestern Thailand. We conducted a three-year longitudinal study of 393 residents (40 men, 353 women) aged 35 and older. Baseline (2019) U-Cd was the primary exposure, and BMD at the calcaneus was measured in 2019 and 2022. Dual-energy X-ray…

Nrf2 deficiency aggravates hepatic cadmium accumulation, inflammatory response and subsequent injury induced by chronic cadmium exposure in mice.

Prolonged cadmium (Cd) exposure leads to Cd accumulation and oxidative damage in the liver. Nuclear factor erythroid-derived 2-like 2 (NRF2) plays a vital role in preventing acute hepatic toxicity of Cd. However, the participation of NRF2 in chronic liver injury, especially in the context of chronic Cd exposure, has rarely been investigated. Here, we explored the involvement of NRF2 in Cd-induced liver injury using Nrf2 knockout (Nrf2-KO) mice chronically exposed to Cd in drinking water (100 or 200 ppm) for up to 24 weeks. We found that absence of Nrf2 exacerbated the Cd-induced liver fibrosis…

Athyrium yokoscense, a cadmium-hypertolerant fern, exhibits two cadmium stress mitigation strategies in its roots and aerial parts.

Athyrium yokoscense is hypertolerant to cadmium (Cd) and can grow normally under a high Cd concentration despite Cd being a highly toxic heavy metal. To mitigate Cd stress in general plant species, Cd is promptly chelated with a thiol compound and is isolated into vacuoles. Generated active oxygen species (ROS) in the cytoplasm are removed by reduced glutathione. However, we found many differences in the countermeasures in A. yokoscense. Thiol compounds accumulated in the stele of the roots, although a long-term Cd exposure induced Cd accumulation in the aerial parts. Synchrotron radiation-bas…

Cadmium sulfide nanoparticle biomineralization and biofilm formation mediate cadmium resistance of the deep-sea bacterium Pseudoalteromonas sp. MT33b.

Cadmium (Cd) is a common toxic heavy metal in the environment, and bacteria have evolved different strategies to deal with Cd toxicity. Here, a bacterium designated Pseudoalteromonas sp. MT33b possessing strong Cd resistance was isolated from the Mariana Trench sediments. Supplement of cysteine significantly increased bacterial Cd resistance and removal rate. Biofilm formation was demonstrated to play a positive role toward bacterial Cd resistance. Transcriptome analysis showed the supplement of cysteine effectively prevented Cd2+ from entering bacterial cells, promoted saccharide metabolism a…

Betulinic acid protects mice from cadmium chloride-induced toxicity by inhibiting cadmium-induced apoptosis in kidney and liver.

Cadmium exposure is closely associated with a variety of diseases including cancers and the accumulation of cadmium has been long recognized as a public health problem. It is therefore of high importance to find methods to reduce cadmium accumulation in the human body. Herein, we report that administration of betulinic acid (BA) protects mice from cadmium chloride (CdCl2)-induced toxicity by inhibiting cadmium-induced apoptosis in both kidney and liver. Mice were given oral doses of 3 mg/kg, 10 mg/kg and 30 mg/kg of BA daily for ten consecutive days, and were injected with one dose of 1 mg/kg …

Renal function after reduction in cadmium exposure: an 8-year follow-up of residents in cadmium-polluted areas.

BACKGROUND AND OBJECTIVE: Long-term exposure to cadmium (Cd) causes renal dysfunction, but the change in renal function with exposure is unknown. We assessed the evolution of Cd-induced renal effects after a reduction in dietary exposure to Cd in rice. METHODS: Four hundred twelve residents in previously Cd-polluted and nonpolluted areas were examined twice, in 1998 and in 2006. Changes in blood Cd, urinary Cd, and kidney function [N-acetyl-β-d-glucosaminidase (NAG), β2-microglobulin, and albumin in urine] were measured. RESULTS: In the most polluted area, mean blood Cd was 8.9 μg/L and 3.3 …

Relation between dietary cadmium intake and biomarkers of cadmium exposure in premenopausal women accounting for body iron stores.

BACKGROUND: Cadmium is a widespread environmental pollutant with adverse effects on kidneys and bone, but with insufficiently elucidated public health consequences such as risk of end-stage renal diseases, fractures and cancer. Urinary cadmium is considered a valid biomarker of lifetime kidney accumulation from overall cadmium exposure and thus used in the assessment of cadmium-induced health effects. We aimed to assess the relationship between dietary cadmium intake assessed by analyses of duplicate food portions and cadmium concentrations in urine and blood, taking the toxicokinetics of cadm…
