Human exposure to potentially carcinogenic compounds.

Average exposures to suspected carcinogens were surveyed for the population in Finland. Alcohol, food and tobacco constitute the largest sources of exposure. Occupational populations may be more exposed than the population at large, by many orders of magnitude. For example, occupational exposure to airborne asbestos and trichloroethylene may be 10 000 times higher than exposure outside workplaces. Application of exposure data to health surveillance and epidemiological research is discussed.

Carcinogenicity of benzene hexachloride and its isomers.

Technical BHC and the alpha, beta, gamma and delta isomers of BHC are carcinogenic for the liver of mice and rats. Mice given the isomers of BHC developed carcinomas and hyperplastic nodules of the liver as early as 24 weeks. Several strains of mice (dd, ICR-JLC, CL1, DDY, IRC, DBA/Z, C3H/HEN, C57BL/6 were susceptible. Male mice were more susceptible to hepatic carcinogenesis than female mice and they appeared to be more susceptible to alpha-BHC. The incidence of neoplasms of the liver was increased when beta-, gamma-, or delta-BHC were each given together with alpha-BHC. PCB-5 promoted the in

Consideration of the possible carcinogenicity of some pesticides.

The sophistication, sensitivity, and the predictability of carcinogenesis bioassays using small rodents is reviewed. Bioassays were conducted using both sexes of mice (B6C3F1:C57BL/6xC3H F1 hybrid) and rats (Osborne-Mendel or Fisher 344). Chemicals tested included three acaracides, two fungicides, two fumigants, four herbicides, twelve chlorinated insecticides, and five organo-phosphorus insecticides. The tumor response of each test strain and sex to the above chemicals is discussed, as well as the spontaneous tumor incidence of each strain. The results suggest caution when interpreting the da

Evaluation of the toxicity and carcinogenicity of hair dyes.

The chronic toxicological and carcinogenic potential following skin painting in mice was evaluated for nine oxidative and three nonoxidative hair dyes. Groups of male and female Swiss mice were treated one time weekly for at least 20 mo with one dose level of each dye. The oxidative dyes were mixed 1:1 with 6% hydrogen peroxide before treatment and the three semipermanent formulations were applied without dilution. Control groups were shaved only and received no applications. Body weights and survival rates did not differ between appropriate male and female treatment and control groups. Absolu

Chemical structure, reactivity, and carcinogenicity of halohydrocarbons.

This review summarizes studies concerning the covalent binding of [14C]TCE to rat liver microsomal protein and exogenous DNA, in vitro, the enhancement of this binding by inducers of mixed-function oxidases, and inhibition of binding by inhibitors of these enzymes. Furthermore, recent studies on this type of binding in various strains of mice and rats of both sexes and using microsomal preparations from various organs are briefly reviewed. Other work reviewed here concerns the synthesis of TCE epoxide and its reaction with nucleophiles since it is believed that TCE epoxide is the activated car

[Carcinogenic risks associated with radiation pollution].

1. The cancerogenic pollution by non-ionizing radiations is limited to the case of solar ultraviolet, whose activity at ground level may be increased as a consequence of the stratospheric depletion of ozone, itself produced by certain chemical pollutants: nitrogen oxydes from supersonic aircrafts, freon. 2. As regards ionizing radiations, the discussion is focused on the fundamental problem of the "threshold", aand on the means by which one may obtain some quantitative data related to carcinogenesis by small radiation doses in Man. A new concept, that of a "practical threshold" is proposed. 3.

The carcinogenicity of pesticides used in New Zealand.

AIM: To estimate the number and weight of pesticides used in New Zealand agriculture that are classified as known or suspected human carcinogens. METHODS: The yearly usage of active ingredients was extracted from the most recent (2005) report on pesticide use. For each active ingredient, the carcinogenicity classification of three regulatory agencies (New Zealand Environmental Protection Authority [EPA NZ], US Environmental Protection Agency, European Chemicals Agency) was extracted. The International Agency for Research on Cancer (IARC) Monograph Programme's classifications were also conside…

[Advances in non-carcinogenic toxicity of trichloroethylene].

Trichloroethylene (TCE) is a widely used organic solvent and an important industrial material. Due to mass production and use, and improper waste disposal, TCE has become a common environmental contaminant, so there is a wide range of occupationally and environmentally exposed population. Occupational and environmental exposure to TCE can produce toxic effects on multiple organs and systems. This paper is a review of the immunotoxicity, reproductive toxicity, neurotoxicity, teratogenic effect and other non-carcinogenic toxic effects of TCE from the aspects of epidemiological study, experimenta

Inhalation carcinogenicity of dichloromethane in rats and mice.

The carcinogenicity of inhaled dichloromethane (DCM) was examined by exposing groups of 50 F344/DuCrj rats and 50 Crj: BDF1 mice of both sexes to 0, 1000, 2000, or 4000 ppm (w/w) DCM-containing aerosol for 2 years. Inhalation of DCM resulted in increased incidences of subcutis fibromas, mammary gland fibroadenoma, and peritoneum mesotheliomas in male rats; mammary gland fibroadenomas in female rats; and bronchiolar-alveolar adenomas and carcinomas in the lung and hepatocellular adenomas and carcinomas in male and female mice. These results clearly indicate that inhaled DCM is carcinogenic in F

Genotoxicity and carcinogenicity risk of carbon nanotubes.

Novel materials are often commercialized without a complete assessment of the risks they pose to human health because such assessments are costly and time-consuming; additionally, sometimes the methodology needed for such an assessment does not exist. Carbon nanotubes have the potential for widespread application in engineering, materials science and medicine. However, due to the needle-like shape and high durability of multiwalled carbon nanotubes (MWCNTs), concerns have been raised that they may induce asbestos-like pathogenicity when inhaled. Indeed, experiments in rodents supported this hy

Effects of carcinogenic metals on gene expression.

Six metals and/or their compounds have been recognized as carcinogens: arsenic, beryllium, cadmium, chromium, cobalt and nickel. With the exception of arsenic, the main rote of exposure is inhalation and the main target organ is the lung. Arsenic is exceptional because it also produces tumors of skin and lung after oral uptake. With the exception of hexavalent chromium, carcinogenic metals are weak mutagens, if at all, and their mechanisms of carcinogenicity are still far from clear. A general feature of arsenic, cadmium, cobalt and nickel is their property to enhance the mutagenicity and carc…

Carcinogenicity, mutagenicity and teratogenicity of manganese compounds.

Manganese, an essential trace element, is one of the most used metals in the industry. Recently, several new manganese compounds have been introduced as fungicide, as antiknock agent in petrol and as contrasting agent in nuclear magnetic resonance tomography. Manganese displays a somewhat unique behaviour with regard to its toxicity. It is relatively non-toxic to the adult organism except to the brain where it causes Parkinson-like symptoms when inhaled even at moderate amounts over longer periods of time. Relatively high doses of manganese affect DNA replication and repair in bacteria and cau

Carcinogenic metals and NF-kappaB activation.

Epidemiological and animal studies suggest that several metals and metal-containing compounds are potent mutagens and carcinogens. These metals include chromium, arsenic, vanadium, and nickel. During the last two decades, chemical and cellular studies have contributed enormously to our understanding of the mechanisms of metal-induced pathophysiological processes. Although each of these metals is unique in its mechanism of action, some common signaling molecules, such as reactive oxygen species (ROS), may be shared by many of these carcinogenic metals. New techniques are now available to reveal…
