Protective Role of Apigenin-PEGylated Chitosan Nanoparticles Against Pesticide Deltamethrin-Induced Liver Injury via Antioxidant Activation, NF-κB Inhibition, and Bcl-2/Bax/Caspase-3 Signaling Modulation.

The present investigation evaluated the liver-protective effect of apigenin encapsulated in PEGylated chitosan nanoparticles (APG-PCHNPs) against hepatotoxicity induced by pesticide deltamethrin (DLM) in rats. Sixty rats were randomly allocated into six groups: a control group, groups treated with APG (20 mg/kg BW) or APG-PCHNPs (20 mg/kg BW), a group exposed to DLM (5 mg/kg BW), and two combination treatment groups receiving DLM (5 mg/kg BW) plus either APG or APG-PCHNPs at 20 mg/kg BW for 35 days. DLM administration significantly decreased serum total protein and its fractions. Simultaneousl

Integrating network pharmacology with ex-vivo analysis to assess the effect of IL-2 in halting breast cancer: involvement of Treg/CTLA-4/Blimp-1/caspase-3.

The T lymphocytes have a vital role in tumor immunosurveillance within the tumor microenvironment (TME) but on the other hand, the TME adopts several mechanisms that cause inhibition and apoptosis to effector T cells leading to tumor immune evasion. The controversial role of interleukin-2 (IL-2) was demonstrated by promoting the activation and proliferation of different immune cells such as NK cells, effector and regulatory T cells (T regs). Thus, ex-vivo approach was used to investigate IL-2 anti-tumor effect on breast cancer cells isolated from Egyptian patients after mastectomy via modulati

Innate detection of Salmonella replication triggers caspase-8-dependent apoptosis via TLR-driven TNF signaling and NLRC4-mediated sensing of the SPI-2 Type III secretion system.

Salmonella enterica comprises over 2500 serovars that are responsible for over 90 million annual infections and 100,000 deaths worldwide. Despite this diversity, our understanding of innate immune responses to Salmonella is based on extensive study of a few serovars, primarily Typhimurium, including strains that cannot replicate within primary murine macrophages. Non-replicating Salmonella trigger caspase-1 and -11-dependent pyroptosis. Whether the innate immune system distinguishes between replicating and non-replicating intracellular Salmonella is poorly defined. Here we demonstrate that rep

Engagement of fatty acids with Toll-like receptor 2 drives interleukin-1β production via the ASC/caspase 1 pathway in monosodium urate monohydrate crystal-induced gouty arthritis.

OBJECTIVE: The concept that intraarticular crystals of uric acid by themselves trigger episodes of painful gouty arthritis is inconsistent with the clinical reality. Patients with large deposits of monosodium urate monohydrate (MSU) crystals (tophi) do not necessarily experience gouty attacks. In fact, it is the excessive consumption of food or alcohol that elicits the inflammation of the acute gout attack. The aim of this study was to identify the precise mechanism that initiates flares of gouty arthritis. METHODS: Human peripheral blood mononuclear cells (PBMCs) and murine macrophages were

A Distinct c-Raf-MEK1/2-p38 MAPK-Caspase Signaling Pathway Mediates TNF-α-Induced Rapid and Non-Lytic Neutrophil Extracellular Trap Formation in Human Neutrophil-Like Cells.

Neutrophil extracellular traps (NETs) contribute to host defense by trapping pathogens. However, excessive or dysregulated NET formation is implicated in the pathogenesis of various conditions, including thrombosis, autoimmune diseases, and cancer. Despite their clinical significance, the specific intracellular signaling pathways underlying NET formation in response to proinflammatory cytokines, including tumor necrosis factor-α (TNF-α), remain incompletely defined. Thus, in the present study, we aimed to elucidate the mechanisms underlying TNF-α-induced NET formation in human neutrophil-like

Natural indoles, indole-3-carbinol (I3C) and 3,3'-diindolylmethane (DIM), attenuate staphylococcal enterotoxin B-mediated liver injury by downregulating miR-31 expression and promoting caspase-2-mediated apoptosis.

Staphylococcal enterotoxin B (SEB) is a potent superantigen capable of inducing inflammation characterized by robust immune cell activation and proinflammatory cytokine release. Exposure to SEB can result in food poisoning as well as fatal conditions such as toxic shock syndrome. In the current study, we investigated the effect of natural indoles including indole-3-carbinol (I3C) and 3,3'-diindolylmethane (DIM) on SEB-mediated liver injury. Injection of SEB into D-galactosamine-sensitized female C57BL/6 mice resulted in liver injury as indicated by an increase in enzyme aspartate transaminase

Evaluation of the effects of silver nanoparticles and ciprofloxacin on apoptotic gene expression (BAX, Bcl-2, and caspase-3) in NALM-6 acute lymphoblastic leukemia progenitor B cells.

Acute lymphoblastic leukemia (ALL), classified as a lymphoid neoplasm under the WHO 5th Edition Haematolymphoid Tumours system, is characterized by uncontrolled proliferation of immature B-cell precursors. Nanotechnology-based therapies are promising for hematologic malignancies because they modulate cellular pathways and improve drug delivery without directly targeting apoptotic proteins. This study investigates the effects of silver nanoparticles (Ag-NPs), known for their anti-cancer and oxidative stress-mediated cytotoxic properties, and ciprofloxacin, a fluoroquinolone with reported anti-l

CAIX-HIF-1α Inhibition by 2-(4-Chlorophenyl)-3-(4-Ethoxyphenyl) Thiazolidin-4-one (BCS12) Activates Caspase Cascade and PARP Cleavage, Driving Mitochondrial Apoptosis in Hypoxic Breast Cancer.

INTRODUCTION: Hypoxia is a hallmark of aggressive breast cancers, particularly Triple- Negative Breast Cancer (TNBC), where stabilization of Hypoxia-Inducible Factor-1α (HIF-1α) upregulates Carbonic Anhydrase IX (CAIX), thereby enabling tumor pH homeostasis, proliferation, and resistance to therapy. Selective CAIX inhibition, therefore, offers a promising therapeutic strategy. In this study, a thiazolidine derivative was identified as a selective inhibitor of CAIX with the use of integrated computational, in vitro, and in vivo studies to develop targeted therapy against TNBC. METHODS: A serie

Cluster of differentiation 147 (CD147) expression is linked with thiram induced chondrocyte's apoptosis via Bcl-2/Bax/Caspase-3 signalling in tibial growth plate under chlorogenic acid repercussion.

Tibial dyschondroplasia (TD) is a metabolic disease of young poultry that affects bone andcartilage's growth. It mostly occurs in broilers due to thiram toxicity in the feed. In this disease, tibial cartilage is not yet ripe for ossification, but it also results in lameness, death, and moral convictions of commercial poultry due to numerous apoptotic changes on cell level. These changes serve a cardinal role in this situation. Many potential problems indicate that chlorogenic acid (CGA) performs an extensive role in controlling apoptosis's perception. However, the actual role of CGA in TD affe
