BACKGROUND: Polycystic kidney disease (PKD) is a genetic disorder affecting millions of people worldwide that is characterized by fluid-filled cysts and leads to end-stage renal disease (ESRD). Liposarcomas are common soft-issue sarcomas arising predominantly in deep soft tissue and the retroperitoneum with varied mortality and recurrence rates, largely dependent on histologic type. We present a rare case of dedifferentiated adamantinoma. The 48-year-old male patient was initially diagnosed with classic adamantinoma in the right fibula and underwent curettage. Two years postoperatively, the patient presented with progressive swelling and pain in the same region. In previous work we described a novel culture technique using a cholera toxin and PMA-free medium (Mel-mix) for obtaining pure melanocyte cultures from human adult epidermis. In Mel-mix medium the cultured melanocytes are bipolar, unpigmented and highly proliferative. Dedifferentiated liposarcoma was first described in 1979, and its definition has been recently extended. We found an undiagnosed anterior mediastinal mass in a 56- year-old man for whom we performed a complete tumor resection through median sternotomy. Microscopic examination revealed that the mass was primarily a low-grade, dedifferentiated liposarcoma, with a partial high-grade area. Three cases of "dedifferentiated" chordoma arising in the sacrococcygeal region are presented. In all three cases, the "dedifferentiated" component arose de novo in conjunction with conventional chordoma.
