Insights into Macrophage Autophagy in Latent Tuberculosis Infection: Role of Heat Shock Protein 16.3.

Tuberculosis (TB) is a major bacterial infectious disease worldwide that is predominantly caused by Mycobacterium tuberculosis (Mtb). The comorbidity of multiple drug-resistant TB strains with HIV and diabetes is widespread. In the presence of these diseases, host immunity is weakened, allowing the recovery of dormant bacilli and leading to recurrent TB infection. As an important component of the host innate and adaptive immune responses, macrophage autophagy plays a significant role in protecting the host against TB. However, dormant bacilli can escape from autophagosomes and/or suppress auto

Over expression of GroESL in Cupriavidus necator for heterotrophic and autotrophic isopropanol production.

We previously reported a metabolic engineering strategy to develop an isopropanol producing strain of Cupriavidus necator leading to production of 3.4gL-1 isopropanol. In order to reach higher titers, isopropanol toxicity to the cells has to be considered. A toxic effect of isopropanol on the growth of C. necator has been indeed observed above a critical value of 15gL-1. GroESL chaperones were first searched and identified in the genome of C. necator. Native groEL and groES genes from C. necator were over-expressed in a strain deleted for PHA synthesis. We demonstrated that over-expressing gro

[Demonstration of Chlamydophila pneumoniae and Chlamydia stress protein, HSP-60 in heart muscle tissue].

There has been much attention to the association between Chlamydophila pneumoniae and athrosclerosis since C. pneumoniae was demonstrated in macropharges, and vascular smooth muscle cells of atheroma tissues. There are few data demonstrating whether C. pneumoniae is present in other tissues than atherosclerotic tissues. We surveyed samples of heart tissue from 10 patients with ischemic heart disease and 10 patients with other disease by immunohistochemical staining with monoclonal antibodies against C. pneumoniae and Chlamydia stress protein 60 (C. HSP-60). In all, 11 out of 20 (55.0%) samples

Molecular phylogenetic analysis of Orientia tsutsugamushi based on the groES and groEL genes.

DNA sequences encoding the GroES and GroEL proteins of Orientia tsutsugamushi were amplified by the PCR and sequenced. Pairwise alignment of full-length groES and groEL gene sequences indicated high sequence similarity (90.4-100% and 90.3-100%) in O. tsutsugamushi, suggesting that these genes are good candidates for the molecular diagnosis and phylogenetic analysis of scrub typhus. Comparisons of the 56-kD type-specific antigen (TSA) protein gene and the groES and groEL genes showed that genotypes based on the 56-kD TSA gene were not related to a cluster containing the groES and groEL genes in

A multiplex approach to molecular detection of Brucella abortus and/or Mycobacterium bovis infection in cattle.

A multiplex amplification and detection platform for the diagnosis of Mycobacterium bovis and Brucella abortus infection simultaneously in bovine milk and nasal secretions was developed. This system (designated the bovine pathogen detection assay [BPDA]-PCR) consists of duplex amplification of species-specific targets (a region of the BCSP31K gene of B. abortus and a repeat-sequence region in the hsp65 gene of M. bovis, respectively). This is followed by a solid-phase probe capture hybridization of amplicons for detection. On the basis of spiking experiments with normal milk, the analytical se

Diabetes-prone NOD mice are resistant to Mycobacterium avium and the infection prevents autoimmune disease.

It was recently proposed that the diabetes genes of non-obese diabetic (NOD) mice are linked to the Bcg gene that is associated with resistance to infection by mycobacteria; however, it has not been established whether NOD mice are resistant or susceptible to the infection, although there are previous investigations on response of NOD mice to other intracellular parasites (e.g. Kaye et al., Eur. J. Immunol. 22: 357-364). We have investigated here this question, as well as the consequences of mycobacterial infection on the natural history of murine diabetes. Female NOD mice were intraperitoneal

Mycobacterial infection induces the secretion of high-mobility group box 1 protein.

High-mobility group box protein 1 (HMGB1) is a non-histone nuclear protein that acts as a pro-inflammatory cytokine and is released by monocytes and macrophages. Necrotic cells also release HMGB1 at the site of tissue damage which induces a variety of cellular responses, including the expression of pro-inflammatory mediators. This study investigated the secretion of HMGB1 in mycobacterial infection by macrophages in vitro and in the lungs of infected guinea pigs. We observed that infection by mycobacterium effectively induced HMGB1 release in both macrophage and monocytic cell cultures. Cultur

A novel mutation in the HSPD1 gene in a patient with hereditary spastic paraplegia.

A mutation in the HSPD1 gene has previously been associated with an autosomal dominant form of spastic paraplegia in a French family. HSPD1 encodes heat shock protein 60, a molecular chaperone involved in folding and quality control of mitochondrial proteins. In the present work we have investigated 23 Danish index patients with hereditary spastic paraplegia (HSP) for mutations in the HSPD1 gene. One patient was found to be heterozygous for a c.1381C > G missense mutation encoding the mutant heat shock protein 60 p.Gln461Glu. The mutation was also present in two unaffected brothers, but absent

Identification of nocardia species by restriction endonuclease analysis of an amplified portion of the 16S rRNA gene.

Identification of clinical isolates of Nocardia to the species level is important for defining the spectrum of disease produced by each species and for predicting antimicrobial susceptibility. We evaluated the usefulness of PCR amplification of a portion of the Nocardia 16S rRNA gene and subsequent restriction endonuclease analysis (REA) for species identification. Unique restriction fragment length polymorphism (RFLP) patterns were found for Nocardia sp. type strains (except for the N. asteroides type strain) and representative isolates of the drug pattern types of Nocardia asteroides (except

Protective-Antigen Scaffold Nanoparticle Vaccine Overcomes Carrier-Induced Suppression and Immunosenescence to Protect against Pseudomonas aeruginosa Pneumonia.

Multidrug-resistant Pseudomonas aeruginosa (PA) causes severe infections, with severe burden especially in older adults. Vaccines remain effective and are urgently needed despite immunosenescence. Self-assembling nanoparticles can enhance vaccine immunogenicity, but their translational use is constrained by the useless anticarrier immunity and carrier-induced epitope suppression (CIES). Herein, we developed a protective-antigen-based nanoparticle platform in which the PA chaperonin GroEL, an intrinsically self-assembling protective antigen, serves as the scaffold for a fusion immunogen reGroEL

CryoEM Strategies for Elucidating the Closed State Architecture of CCT.

Cryo-electron microscopy (CryoEM) has emerged as a state-of-the-art technique in structural biology, thanks to advances in sample preparation, high-voltage electron microscopy, direct electron detectors, and sophisticated image processing software. In this study, we investigate the eukaryotic chaperonin CCT, a ~1 MDa hetero-oligomeric complex essential for the folding of key substrates such as actin, tubulin, and WD40 family members. While several open-state structures of CCT have been resolved, the highest-resolution reconstructions have been obtained in the closed state. This is primarily du

Metronidazole and ether derivatives target Helicobacter pylori via simultaneous stress induction and inhibition.

Metronidazole is a front-line drug for the treatment of Helicobacter pylori infections. However, its mode of action and cellular targets are poorly defined, and higher dosing and combination therapies are required to overcome resistance. Here we performed activity-based protein profiling with tailored metronidazole probes and identified chaperonin HpGroEL and thiol peroxidase HpTpx as prominent targets, the latter being essential for H. pylori survival under oxidative stress. Alkynylated ether probes exhibited enhanced antibacterial potency compared with the parent drug in vitro, including act

CCT7 Regulates TRAF6-Mediated Autophagy to Promote Posttraumatic Joint Contracture.

Posttraumatic joint contracture (PTJC) is a prevalent complication of joint injury, characterised by marked reductions in both active and passive joint motion. Previous studies have indicated that the chaperonin-containing T-complex polypeptide (CCT) is involved in fibrotic processes. Thus, this study aims to explore the role of CCT7 in PTJC and clarify its underlying regulatory mechanisms. We found that CCT7 expression was significantly upregulated in PTJC joint tissues and in fibroblasts stimulated with TGF-β1. Knockdown of CCT7 markedly reduced the expression of α-SMA and COL-I and suppress

The CCT6A paradigm: biomarker potential and therapeutic challenges in oncology and beyond.

Chaperonin Containing TCP1 Subunit 6A (CCT6A), a structurally conserved core subunit of the eukaryotic CCT/TRiC complex, orchestrates ATP-dependent protein folding, cytoskeletal dynamics, and stress-responsive adaptation, reflecting its indispensable role in cellular homeostasis. Emerging evidence highlights CCT6A as a multifaceted oncogenic driver across diverse malignancies (such as colorectal cancer and hepatocellular carcinoma), through metabolic reprogramming, stabilization of oncoproteins, and immunosuppression. Its overexpression correlates with aggressive phenotypes, therapeutic resist

Prefoldin subunit 6 of Plasmodium falciparum binds merozoite surface protein-1.

Malaria is a human disease caused by eukaryotic protozoan parasites of the Plasmodium genus. Plasmodium falciparum (Pf) causes the most lethal form of human malaria and is responsible for widespread mortality worldwide. Prefoldin is a heterohexameric molecular complex that binds and delivers unfolded proteins to chaperonin for correct folding. The prefoldin PFD6 is predicted to interact with merozoite surface protein-1 (MSP-1), a protein well known to play a pivotal role in erythrocyte binding and invasion by Plasmodium merozoites. We previously found that the P. falciparum (Pf) genome contain

Evolution and activation mechanism of the flavivirus class II membrane-fusion machinery.

The flavivirus envelope glycoproteins prM and E drive the assembly of icosahedral, spiky immature particles that bud across the membrane of the endoplasmic reticulum. Maturation into infectious virions in the trans-Golgi network involves an acid-pH-driven rearrangement into smooth particles made of (prM/E)2 dimers exposing a furin site for prM cleavage into "pr" and "M". Here we show that the prM "pr" moiety derives from an HSP40 cellular chaperonin. Furthermore, the X-ray structure of the tick-borne encephalitis virus (pr/E)2 dimer at acidic pH reveals the E 150-loop as a hinged-lid that open

TCP1γ Subunit Is Indispensable for Growth and Infectivity of Leishmania donovani.

T-complex protein-1 (TCP1) is a ubiquitous group II chaperonin and is known to fold various proteins, such as actin and tubulin. In Leishmania donovani, the γ subunit of TCP1 (LdTCP1γ) has been cloned and characterized. It forms a high-molecular-weight homo-oligomeric complex that performs ATP-dependent protein folding. In the present study, we evaluated the essentiality of the LdTCP1γ gene. Gene replacement studies indicated that LdTCP1γ is essential for parasite survival. The LdTCP1γ single-allele-replacement mutants exhibited slowed growth and decreased infectivity in mouse macrophages comp
