Natural products (NPs) have attracted increasing attention due to their structural diversity, abundance, and low environmental impact, making them valuable for novel fungicide development. In this study, a series of coumarin-7-N/O acethydrazide derivatives were synthesized by incorporating acetic acid moieties into the coumarin scaffold. The in vitro antifungal activities of these compounds were evaluated against six common plant pathogenic fungi. Compound B13 demonstrated significant antifungal effects, with EC50 values of 3.24, 1.89, 1.70, 0.36, 4.27, and 1.50 μg/mL against Botrytis cinerea,

Fungal diseases remain a major constraint on global crop production, driving the search for fungicides with broader efficacy and improved selectivity. Confronting the dual challenges of structural rigidity and declining antifungal performance in conventional succinate dehydrogenase inhibitors (SDHIs), this study developed novel syringic acid derivatives. Notably, W11 exhibited remarkable broad-spectrum potency against eight phytopathogenic fungi and two oomycete species, with EC50 values of 0.679, 0.424, and 0.384 μg/mL against Gibberella zeae, Alternaria solani, and Colletotrichum fructicola,

The root microbiome includes fungal pathogens capable of colonizing multiple plant hosts, yet the underlying genetic determinants remain unknown. Here we report that Plectosphaerella cucumerina is a core member of the Arabidopsis thaliana root microbiota, which displays pathogenic potential across multiple hosts. Using a collection of 72 Plectosphaerella isolates and whole-genome sequencing, we observed subtle phenotypic and genotypic variation associated with fungal phylogeny but not host plant identity. Transcriptome profiling of a P. cucumerina isolate in roots of diverse plants revealed co

This study evaluates the chemical profile, physicochemical stability, and antifungal potential of the essential oil extracted from Schinus terebinthifolia Raddi residues (EOStR) against banana anthracnose, using a confirmed Colletotrichum musae isolate in the in vitro assays. Hydrodistillation yielded 6% ± 1 (m m-1) of EOStR, mainly composed of δ-3-carene (28.1%), limonene (22.5%), and α-phellandrene (17.1%), with hydrogenated monoterpenes representing over 93% of the oil. A stable microemulsion containing 10% EOStR was formulated, showing no phase separation, predictable colloidal rearrangeme

Rising atmospheric CO2 has significant implications for crop productivity and food security. Based on studies in C3 plants, elevated CO2 (eCO2) can shape plant-pathogen interactions, although the outcomes are often variable. The question of how eCO2 influences immunity and disease development in C4 plants, such as the globally important cereal crop maize (Zea mays L.), has not been systematically examined. We challenged maize plants grown under ambient CO2 (aCO2, 420 ppm) and eCO2 (550 ppm) with bacterial, viral, fungal, and oomycete pathogens. Plants grown in eCO2 were more susceptible to sug

Soybean (Glycine max), a crucial global crop, experiences yearly yield reduction due to diseases such as anthracnose (Colletotrichum truncatum) and root rot (Fusarium spp.). The use of fungicides, which have traditionally been employed to control these phytopathogens, is now facing challenges due to the emergence of fungicide-resistant strains. Streptomyces bacillaris S8 strain S8 is previously known to produce valinomycin t through a nonribosomal peptide synthetase (NRPS) pathway. The objective of this study was to evaluate the antifungal activity of S. bacillaris S8 against C. truncatum and

This study integrated transcriptome and hormone profiling of Colletotrichum camelliae-infected tea leaves to identify the methyl esterase (MES) gene family. Genome-wide analysis identified 17 MES genes, with CsMES4 as a crucial candidate localized in the nucleus and cell membrane. Through prokaryotic expression, purification, in vitro enzymatic assays, and transient silencing experiments in tea plants, we demonstrated that CsMES4 hydrolyze methyl salicylate (MeSA), methyl indole-3-acetate (MeIAA), and methyl jasmonate (MeJA) to produce salicylic acid, indole-3-acetic acid, and jasmonic acid, r

Selenium treatment has been shown to enhance plant resilience against both biotic and abiotic stresses; however, whether it can improve citrus resistance to bacterial diseases remains unexplored. In this study, we systematically identified sulfate transporter (Sultr) genes in the Citrus sinensis genome that are involved in selenate uptake. Analysis of gene structure, conserved motifs, and protein domains revealed a high degree of conservation among CsSultrs. Collinearity analysis between C. sinensis and Arabidopsis thaliana Sultr genes suggested possible evolutionary duplication and divergence

BACKGROUND: Colletotrichum fructicola is a predominant anthracnose species in Camellia oleifera, causing various adverse effects. Traditional intercropping Vernicia fordii with C. oleifera may enhance anthracnose resistance, but the mechanism remains elusive.
METHODS: We utilized UPLC-MS/MS and acid-base detection to identify the major antimicrobial alkaloid components in the V. fordii leaf extract. Subsequently, by adding different concentrations of V. fordii leaf extract for cultivating C. fructicola, with untreated C. fructicola as a control, we investigated the impact of the V. fordii lea

Glomerella leaf spot (GLS), a fungal disease caused by Colletotrichum fructicola, severely affects apple quality and yield, yet few resistance genes have been identified in apple (Malus domestica Borkh.). Here we found a transcription factor MdWRKY17 significantly induced by C. fructicola infection in the susceptible apple cultivar "Gala." MdWRKY17 overexpressing transgenic "Gala" plants exhibited increased susceptibility to C. fructicola, whereas MdWRKY17 RNA-interference plants showed opposite phenotypes, indicating MdWRKY17 acts as a plant susceptibility factor during C. fructicola infectio

Essential oil-based products with broad plant disease control claims are commercially available and may be a practical alternative to copper fungicides for crop protection in organic mango orchards. We evaluated the disease control efficacy and crop safety of thyme oil, savory oil, and tree tea oil through replicated in vitro, in vivo (detached leaf and potted trees), and field assays. Three Colletotrichum species associated with mango anthracnose were tested in vitro, whereas only C. siamense was used for in vivo assays. Within the range of concentrations tested in vitro (62.5 to 2,000 μl act

Our previous work demonstrated the values of 2-(2-oxazolinyl)aniline (NHPhOx) in achieving scaffold-distinct antifungal amides. Herein, amide-free antifungal leads are disclosed, featuring the replacement of N-phenylamide by pyrazolo[1,5-a]pyrimidine. Pyrazolo[1,5-a]pyrimidine oxazolines and analogs were designed and synthesized via N-distribution inspired hybridization of pharmaceutically important pyrazolo[1,5-a]pyrimidine and oxazolines. In vitro evaluation showed that compound D10 stands out as a novel antifungal candidate against agriculturally important Colletotrichum capsici and Pyricul

Glomerella leaf spot (GLS), a fungal disease caused by Colletotrichum fructicola, severely affects apple (Malus domestica) quality and yield. In this study, we found that the transcription factor MdWRKY71 was significantly induced by C. fructicola infection in the GLS-susceptible apple cultivar Royal Gala. The overexpression of MdWRKY71 in apple leaves resulted in increased susceptibility to C. fructicola, whereas RNA interference of MdWRKY71 in leaves showed the opposite phenotypes. These findings suggest that MdWRKY71 functions as a susceptibility factor for the apple-C. fructicola interacti

Infection by many fungi activates a variety of calcium dependent defenses in the hosts, slowing or suppressing the attacker and limiting the efficacy of mycoherbicides. The calcium requirement for fungal growth is so low that it could only be implied based on fungi containing calcium-dependent signaling enzymes. Analytical grade media contain <2 microM calcium, and the addition of specific chelators does not affect fungal growth. Hydrophobic derivatives of the calcium-specific chelator BAPTA designed to traverse plant cuticles were synthesized in order to chelate calcium internally during fung

Two strains of aerobic and mesophilic microorganisms were isolated from palm-tree plantation sand. They grew on insoluble polysaccharides: mannan, cellulose, chitin as only source of carbon. This lytic activity was used for the purification of the two strains. The morphology of the organisms and the presence of LL-diaminopimetic acid in their cell-wall are characteristic of the genus Streptomyces. Investigations lead to: 1) the characterization of their specific polysaccharidase activity toward insoluble and natural beta-and alpha-glycans (mannan, cellulose, chitin, pectine and starch) and the
