Parity and dietary energy regulate IgG and non-IgG bioactive proteins in colostrum of beef cattle.

Bovine colostrum provides crucial immune and developmental support to newborn calves, yet the roles of non-immunoglobulin bioactive proteins remain underexplored. The objective of this study was to characterize the colostrum proteome and evaluate the effects of parity, prepartum dietary metabolizable energy, and colostral immunoglobulin G (IgG) concentration on its protein composition. Colostrum samples from primiparous and multiparous Angus × Simmental beef cows fed low-, control-, or high-metabolizable-energy diets during the prepartum period were analyzed by quantitative proteomics. We iden…

Differential Expressions Between Colostrum and Mature Milk in Primiparous Holstein Cows with Integrations of Lipidomic and Proteomics.

In the current study, by integrating the analyses of lipidomics and proteomics, the compositional differences between colostrum and mature milk of Holstein cows were systematically investigated. We have identified a total of 1863 lipid metabolites and 1690 proteins. A small portion of them were highly differentiated between colostrum and mature milk. Among them, 23 highly differentiated lipid metabolites and 17 proteins were screened to run correlation analysis. The results showed that they are highly correlated; therefore, they could serve as biomarkers to distinguish colostrum from mature mi…

Effects of breed and colostrum replacer quantity on lymphocyte populations in the blood of Holstein and Holstein × Angus calves.

The objective of this study was to explore the effects of breed and colostrum replacer (CR) quantity fed at birth on lymphocyte populations in the peripheral blood of Holstein (H) and Holstein × Angus (C) crossbred calves. Male H (n = 10) and C (n = 10) calves were enrolled immediately after birth and randomly assigned to one of 2 quantities of CR: low (2.5 g of IgG/kg of birth BW; n = 4 for H and 6 for C) or high (5.0 g of IgG/kg of birth BW; n = 6 for H and 4 for C). Calves were tube fed CR meals at 0 and 12 h and then fed milk replacer at an average of 12% of BW for 21 d, followed by an ave…

Bovine colostrum for prevention of recurrent urinary tract infections in susceptible children: A randomized controlled trial.

BACKGROUND: Recurrent urinary tract infections (UTIs) in children with congenital anomalies of the kidney and urinary tract (CAKUT) are a major cause of deterioration of kidney functions and possibly, renal scarring. Rising antimicrobial resistance necessitates alternative preventive strategies. Bovine colostrum (BC), a bioactive prebiotic, has emerged as a promising adjuvant therapy in upper respiratory and gastrointestinal (GIT) infections. Given the shared mucosal immune mechanisms, the urinary tract may represent a suitable target for this preventive strategy to reduce UTI recurrence. AIM…

The influence of colostrum-derived maternal cells on neonatal lamb immune responses to routine management practices.

Colostrum is essential for newborn ruminants because it delivers vital nutrients, boosts immune function, and promotes growth. Studies suggest that maternal cells found in colostrum enhance the immune development of newborns, resulting in lasting health benefits. This study examines the impact of maternal colostral cells on lamb growth and husbandry-related mucosal health challenges. Initial assessments of ovine colostrum samples (n = 9) identified an effective method for removing maternal cells from colostrum. This method was utilized in an ewe-twin lamb model across two lambing seasons durin…

Evaluation of Selected Pro- and Anti-Inflammatory Adipokines in Colostrum from Mothers with Gestational Diabetes Mellitus.

Adipokines related to gestational diabetes mellitus (GDM) are an emerging area of interest. The aim of this study was to evaluate the associations between GDM and adipokine levels in human milk. This was an observational cohort study targeting mothers with gestational diabetes, which evaluated the association of maternal hyperglycemia severity, classified as GDM-G1 (diet treatment) and GDM-G2 (insulin treatment), with colostral adipokines involved in pro- and anti-inflammatory processes. Colostrum was collected from hyperglycemic (N = 34) and normoglycemic (N = 26) mothers, and adipokine level…

Effect of low colostrum intake on gastrointestinal development and uterine and cervical morphometrical architecture in the neonatal gilt.

To assess the importance of natural variation in colostrum intake on piglet gastrointestinal and reproductive development, two equally sized female piglets from each of 27 litters were selected, one with low (average 226 g) and one with high (average 401 g) colostrum intake. At weaning (23 d of age), piglets were euthanised to perform macromorphological measurements on ileum, colon, cervix and uterus tissues, and to obtain tissue samples from the cervix and uterus for histology. Sections of uterine and cervical preparations were analysed using digital image analysis. Despite being selected for…

Effect of oropharyngeal colostrum therapy on neonatal sepsis in preterm neonates: A systematic review and meta-analysis.

Various studies have shown that oropharyngeal colostrum application (OPCA) is beneficial to preterm neonates. We performed a systematic review and meta-analysis to assess whether OPCA reduces the incidence of culture-proven neonatal sepsis in preterm neonates. Randomized controlled trials comparing OPCA with placebo or standard care in preterm neonates were included. Medline, Embase, Web of Science, Cumulated Index to Nursing and Allied Health Literature, Scopus, and CENTRAL were searched for studies published up to June 15, 2023. We used the Cochrane Risk of Bias tool, version 2, for risk of …

Oropharyngeal application of colostrum or mother's own milk in preterm infants: a systematic review and meta-analysis.

CONTEXT: Many preterm neonates often cannot be fed enterally and hence do not receive the benefits of colostrum. Oropharyngeal application of colostrum is a novel way of harnessing the immunological benefits of colostrum. Randomized controlled trials (RCTs) investigating the efficacy of this approach have shown variable results. OBJECTIVE: The aim of this systematic review was to synthesize available data on the effect of oropharyngeal application of colostrum or mother's own milk (CMOM) in preterm infants. DATA SOURCES: Six electronic databases (MEDLINE, Embase, CINAHL, Scopus, Web of Scien…

Bovine colostrum to supplement the first feeding of very preterm infants: The PreColos randomized controlled trial.

BACKGROUND & AIMS: Gut immaturity leads to feeding difficulties in very preterm infants (<32 weeks gestation at birth). Maternal milk (MM) is the optimal diet but often absent or insufficient. We hypothesized that bovine colostrum (BC), rich in protein and bioactive components, improves enteral feeding progression, relative to preterm formula (PF), when supplemented to MM. Aim of the study is to determine whether BC supplementation to MM during the first 14 days of life shortens the time to full enteral feeding (120 mL/kg/d, TFF120). METHODS: This was a multicenter, randomized, controlled tri…

Label-free quantitative proteomics analysis reveals the fate of colostrum proteins in the intestine of neonatal calves.

The contribution of intestinally absorbed colostral immunoglobulins to the transmission of passive immunity is widely reported in neonatal calves. However, changes in the colostral proteome in the gastrointestinal digesta remain unclear. Therefore, this study aimed to investigate changes in colostral proteome affected by gastrointestinal proteases in neonatal calves. Twenty-one neonatal Holstein calves were used in this study, including 18 colostrum-fed calves slaughtered at 8 (CI, n = 6), 24 (CII, n = 6), and 36 h (CIII, n = 6) postpartum and 3 milk-fed calves slaughtered 24 h postpartum (MI,…

Mycobacterium bovis DNA detection in colostrum as a potential indicator of vaccination effectiveness against bovine tuberculosis.

Bovine tuberculosis (bTB) remains a problem on many dairy farms in Mexico, as well as a public health risk. We previously found a high frequency of Mycobacterium bovis DNA in colostrum from dairy cows using a nested PCR to detect mpb70. Since there are no reliable in vivo tests to determine the effectiveness of booster Mycobacterium bovis BCG vaccination against bTB, in this work we monitored M. bovis DNA in colostrum by using this nested PCR. In order to decrease the risk of adverse reactions in animals likely containing viable M. bovis, a single application of BCG and a subunit vaccine (EEP-…

[Clinical and epidemiological determinants of sodium and potassium levels in the colostrum of breastfeeding mothers with and without hypertension in Brazil].

This study aimed to determine whether maternal hypertension and other clinical and epidemiological determinants have an impact on sodium and potassium levels in the 48-hour colostrum of breastfeeding mothers. The study included 105 randomly selected breastfeeding mothers, of whom 72 (68.8%) had normal blood pressure and 33 (31.4%) were hypertensive. Colostrum was collected in-hospital in the morning, and sodium and potassium concentrations were measured using a flame photometer. When sodium and potassium concentrations were compared to the variables age, parity, family history of hypertension,…

Effects of dexamethasone on lymphoid tissue in the gut and thymus of neonatal calves fed with colostrum or milk replacer.

An increased susceptibility to disease in neonatal calves may be attributable to high glucocorticoid levels that influence immune reactions. We tested whether dexamethasone (DEXA) administration influences the proliferation, apoptosis, and number of B- and T-lymphocytes in Peyer's patches (PP) and thymus in calves fed colostrum (C) or a milk-derived formula. All calves were subcutaneously administered bovine colostrum-derived immunoglobulin G and fed chicken-egg derived immunoglobulins that protected against rotavirus and pathogenic Escherichia coli. The DEXA (30 microg/kg of BW daily) was inj…

Inhibition of enteropathogenic Escherichia coli (EPEC) adherence to HEp-2 cells by bovine colostrum and milk.

BACKGROUND: enteropathogenic Escherichia coli (EPEC) is the main etiological agent of infantile diarrhea in Brazil and other developing countries. Human milk IgA protects newborn intestinal mucosa by inhibiting bacterial adhesion to epithelial cells and this effect is shown by in vitro assays of EPEC adhesion to HEp-2 cultured cells. Bovine milk, if effective in promoting this protection, could be an useful tool in the absence of the natural breastfeeding, in high-risk nurseries or in hospital infections. METHODS: the effect of colostrum, milk, and serum from dairy cows on the adherence to EP…

Evaluation of colostrum and whey-derived gamma globulins for prevention of cryptosporidiosis in artificially infected neonatal calves.

Thirty colostrum-deprived Friesian male calves were used to study the effect of colostrum and whey-derived gammaglobulins on the clinical events and oocyst-excretion pattern following artificial infection with a fresh, field-sourced strain of Cryptosporidium parvum. The calves were raised naive and free from contact with C. parvum from birth until infection at either 10 or 17 days of age. The two age groups comprised three sub-groups that were each given a single treatment of colostrum, whey-derived gammaglobulins or whole milk (controls) at 5 hours after birth. Blood samples taken both before…

Effect of colostrum intake on metabolic rate and plasma glucose in the neonatal pig in relation to environmental temperature.

The effects of feeding sow colostrum on oxygen consumption (VO2), heat production (HP), respiratory quotient (RQ) and rectal temperature (RT) were examined in 26 newborn piglets in a thermoneutral (32 degrees C) and in a cold (18 degrees C) environment. Colostrum (32 g/kg body weight) was given by gavage at 3 h of age. A control group received a similar amount of saline. VO2, HP, RQ and RT were measured for approximately 1 h prior to force-feeding and 2 h after feeding. The response of plasma glucose to ingestion of colostrum or saline was recorded in 18 additional piglets kept at 32 or 18 deg…
