Targeting injury-induced transglutaminase-2 activity with a cementoin-SLPI fusion protein: a novel therapeutic strategy for alkali-induced corneal injury.

BACKGROUND: Chemical injury of the cornea, particularly alkali-induced corneal injuries, triggers a self-amplifying cascade of inflammation, extracellular matrix disruption, and pathological neovascularization that often resists conventional therapy. Transglutaminase-2 (TG2) is a calcium-dependent enzyme whose activation in injured tissues orchestrates crosslinking of matrix proteins and modulation of inflammatory signaling, positioning it as a mechanistic hub in tissue damage. MAIN TEXT: Here we review the molecular and cellular mechanisms underpinning TG2's role in corneal injury, focusing …

BRD4 inhibition mitigates acute and chronic corneal injury following topical nitrogen mustard exposure.

Corneal chemical injury, including mustard vesicant injury, remains a clinical challenge with limited therapeutic options. To enable effective mechanistic and therapeutic studies, we developed a mouse model of nitrogen mustard (NM)-induced corneal injury using a 3D-molded, cap-shaped filter paper conforming to the corneal surface to generate reproducible injuries. Multimodal in vivo imaging and histological assessment revealed an injury course characterized by acute epithelial disruption, stromal edema, and inflammation, followed by transient partial recovery and chronic stromal remodeling wit

Correlation between tear cytokine profiles and corneal injury degree in patients with dry eye syndrome.

BACKGROUND: Dry eye syndrome (DES) is a very common ocular surface disorder that is closely related to corneal damage. However, the relationship between tear cytokines in patients with DES and the severity of corneal damage remains unclear. METHODS: Patients with DES of different corneal injury degrees (mild, moderate, severe) were enrolled. The baseline data of each group of patients were compared. The levels of tears cytokines were detected by the multiplex Luminex assay. The Kruskal-Wallis H test and the Mann-Whitney U test were used to compare the levels of cytokines. Independent risk fac

Topical application of Cas9 ribonucleoproteins inhibits corneal neovascularization in a mouse model of alkali burn injury.

Corneal neovascularization is a sight-threatening condition for which current treatments such as anti-VEGF agents are limited by invasiveness and side effects. We present the first non-viral, CRISPR/Cas9-based gene therapy delivered via topical eye drops that penetrates the cornea and inhibits pathological neovascularization. Cas9 ribonucleoproteins (RNPs) targeting the Vegfa gene were complexed with a liposomal carrier (lipofectamine) and administered to mice after alkali burn injury to the cornea. This approach achieved approximately 2% gene editing at the Vegfa locus in vivo, which signific

Discovery of Trolox amide derivatives as potent membrane-targeted ferroptosis inhibitors for corneal injury therapy.

Vitamin E and its water-soluble analogue Trolox are potent radical scavengers but require high concentrations to inhibit ferroptosis effectively. The discovery of a Trolox amide derivative 1 showing 95-fold greater potency inspired the systematic synthesis of a focused library. Most amide derivatives demonstrated significantly enhanced potency, with IC50 values in the sub-micromolar range. Notably, compound 20, which features an indazole moiety, emerged as the most potent inhibitor, exhibiting a 556-fold improvement in activity (IC50 = 0.036 μM) over Trolox and a favorable safety index. Early myeloid cell infiltration and subset-specific macrophage responses in murine corneal nerve injury.

PURPOSE: Corneal nerve fibers and resident macrophages form a specialized microenvironment essential for tissue integrity and recovery after injury. This study aims to elucidate the early immune dynamics following corneal nerve injury, focusing on myeloid cell infiltration and resident macrophage subset shifts. METHODS: Using a murine circular nerve-cut model, we tracked immune responses for 21 days, with a focus on the first 12 h post-injury. Confocal imaging was used to assess corneal nerve density, while flow cytometry quantified infiltrating and resident immune cell populations. Transcrip

Snowflake-inspired and blink-driven flexible piezoelectric contact lenses for effective corneal injury repair.

The cornea is a tissue susceptible to various injuries and traumas with a complicated cascade repair process, in which conserving its integrity and clarity is critical to restoring visual function. Enhancing the endogenous electric field is recognized as an effective method of accelerating corneal injury repair. However, current equipment limitations and implementation complexities hinder its widespread adoption. Here, we propose a snowflake-inspired, blink-driven flexible piezoelectric contact lens that can convert mechanical blink motions into a unidirectional pulsed electric field for direc

Topical application of calcitonin gene-related peptide as a regenerative, antifibrotic, and immunomodulatory therapy for corneal injury.

Calcitonin gene-related peptide (CGRP) is a multifunctional neuropeptide abundantly expressed by corneal nerves. Using a murine model of corneal mechanical injury, we found CGRP levels in the cornea to be significantly reduced after injury. Topical application of CGRP as an eye drop three times daily accelerates corneal epithelial wound closure, reduces corneal opacification, and prevents corneal edema after injury in vivo. We then used a series of in vitro and in vivo techniques to investigate the mechanisms underlying CGRP's functions. CGRP promotes corneal epithelial cell migration, prolife

Involvement of endoplasmic reticulum stress in trigeminal ganglion corneal neuron injury in dry eye disease.

Dry eye disease (DED) is a multifactorial disease with a high prevalence worldwide. Uncomfortable corneal sensations severely affect daily life in DED patients. Hence, corneal neuron injury is a vital pathogenesis in DED. Notably, endoplasmic reticulum stress (ERS) plays a role in peripheral neuron injury. However, the role of ERS in DED corneal neuron injury is still far from being clear. In this study, we established an environmental DED (eDED) model in vivo and a hyperosmotic DED model in vitro. Subsequently, trigeminal ganglion (TG) corneal neurons were retrograde labeled by WGA-Alexa Fluo

URP20 improves corneal injury caused by alkali burns combined with pathogenic bacterial infection in rats.

Corneal alkali burns often occur in industrial production and daily life, combined with infection, and may cause severe eye disease. Oxidative stress and neovascularization (NV) are important factors leading to a poor prognosis. URP20 is an antimicrobial peptide that has been proven to treat bacterial keratitis in rats through antibacterial and anti-NV effects. Therefore, in this study, the protective effect and influence mechanism of URP20 were explored in a rat model of alkali burn together with pathogenic bacteria (Staphylococcus aureus and Escherichia coli) infection. In addition, human um

Ablation of Sphingosine Kinase 1 Protects Cornea from Neovascularization in a Mouse Corneal Injury Model.

The purpose of this study was to investigate the role of sphingosine kinase 1 (SphK1), which generates sphingosine-1-phosphate (S1P), in corneal neovascularization (NV). Wild-type (WT) and Sphk1 knockout (Sphk1-/-) mice received corneal alkali-burn treatment to induce corneal NV by placing a 2 mm round piece of Whatman No. 1 filter paper soaked in 1N NaOH on the center of the cornea for 20 s. Corneal sphingolipid species were extracted and identified using liquid chromatography/mass spectrometry (LC/MS). The total number of tip cells and those positive for ethynyl deoxy uridine (EdU) were quan

Sulfur mustard corneal injury is associated with alterations in the epithelial basement membrane and stromal extracellular matrix.

Sulfur mustard (SM; bis(2-chloroethyl) sulfide) is a highly reactive bifunctional alkylating agent synthesized for chemical warfare. The eyes are particularly sensitive to SM where it causes irritation, pain, photophobia, and blepharitis, depending on the dose and duration of exposure. In these studies, we examined the effects of SM vapor on the corneas of New Zealand white male rabbits. Edema and hazing of the cornea, signs of acute injury, were observed within one day of exposure to SM, followed by neovascularization, a sign of chronic or late phase pathology, which persisted for at least 28

Effects of mesenchymal stem/stromal cells on cultures of corneal epithelial progenitor cells with ethanol injury.

PURPOSE: Mesenchymal stem/stromal cells (MSCs) facilitate the regeneration of injured tissue. Our group has previously shown that human MSCs (hMSCs) or hMSC-derived factors suppress excessive inflammatory response in the cornea following chemical injury in vivo. We here investigated direct effects of hMSC-derived factors on cultures of chemically injured human corneal epithelial progenitor cells (hCEP), independent of systemic anti-inflammatory effects that hMSCs have been shown to have in vivo. METHODS: We injured hCEP by incubation in 20% ethanol for 30 seconds, and cultured the cells in fr

ZO-1 reorganization and myofibroblast transformation of corneal endothelial cells after freeze injury in the cat.

Corneal endothelial wound healing following scrape injury in the rabbit and cat is characterized by cell spreading and maintenance of a normal endothelial phenotype consisting of apically-localized, circumferential microfilament bands and cell border-associated ZO-1, a tight junction protein and marker for endothelial differentiation. In contrast, after freeze injury in the rat and rabbit endothelial cells develop basally organized microfilament bundles (stress fibers), and appear to proliferate and form a multilayered zone at the wound margin. The purpose of the present study was to determine
