Cytokines in the pathogenesis of cancer cachexia.

PURPOSE OF REVIEW: The aim of the present review is to summarize and update the role of different cytokines in the pathogenesis of cancer cachexia and to provide therapeutic strategies based on cytokine action. RECENT FINDINGS: Cancer cachexia is a syndrome characterized by a marked weight loss, anorexia, asthenia and anemia. The cachectic state is invariably associated with the presence and growth of the tumor and leads to a malnutrition status due to the induction of anorexia or decreased food intake. In addition, the competition for nutrients between the tumor and the host leads to an acce…

Veno-occlusive disease: cytokines, genetics, and haemostasis.

Hepatic veno-occlusive disease (VOD) is a major cause of morbidity and mortality following high dose cytotoxic therapy for stem cell transplantation (SCT). Pre-existing liver damage, SCT-related therapy, and genetic polymorphisms all appear to increase the risk of developing VOD. Studies of biological markers during SCT suggest that cytokines, haemostasis, and hepatic drug metabolism via the glutathione pathway are all involved in the pathogenesis of VOD. Until recently, treatment options were limited and experimental therapies directed at the pathogenesis of the disease were mostly unsuccessf…

Cytokines--central factors in alcoholic liver disease.

Many processes related to the consumption or breakdown of alcohol that contribute to alcohol-induced liver disease are mediated by small proteins known as cytokines, which are produced and secreted by liver cells and many other cells throughout the body. Through a variety of actions, cytokines regulate certain biochemical. processes in the cells that produce them as well as in neighboring cells. For example, in case of an infection, they attract white blood cells to the tissues, triggering an inflammatory response. In the liver, persistent cytokine secretion resulting in chronic inflammation l…

Role of cytokines in postmenopausal bone loss.

It is increasingly recognized that changes in cytokine activity are involved in the pathophysiology of postmenopausal bone loss in many ways. Estrogen may affect skeletal cytokine activity by diverse mechanisms such as direct interference with cytokine gene transcription, regulation of major signaling molecules, changes in lymphocyte function, and central effects on the hypothalamic-pituitary-adrenal axis. This article summarizes our current knowledge of these interactions and discusses the possible implications with respect to the prevention and treatment of postmenopausal osteoporosis.

S-Adenosylmethionine, cytokines, and alcoholic liver disease.

Hepatic deficiency of S-adenosylmethionine (AdoMet) is a critical acquired metabolic abnormality in alcoholic liver disease (ALD) and in many experimental models of hepatotoxicity. Subnormal AdoMet, elevated serum tumor necrosis factor (TNF), and endotoxemia (LPS) are hallmarks of ALD and experimental liver injury. AdoMet deficiency is attributed to its subnormal synthesis, but mechanisms for increased TNF are not known. AdoMet deficiency may affect the critical balance of proinflammatory (e.g., TNF) and antiinflammatory [e.g., interleukin (IL)-10] cytokines. Rats maintained on a choline-defic…

Cytokines in severe respiratory syncytial virus bronchiolitis.

Bronchiolitis caused by respiratory syncytial virus (RSV) infection is an important cause of severe lung disease in infants, and increasing evidence suggests that it is immunologically mediated. Experiments in mice suggest that this may be due to differential T-cell activation producing either type 1 or type 2 cytokines. We investigated this hypothesis in man by studying 24 infants ventilated with severe RSV bronchiolitis and by measuring messenger RNA (mRNA) for interleukin-4 (IL-4) and interferon-gamma (IFN-gamma), by polymerase chain reaction, in nasopharyngeal aspirates (NPAs) and bronchoa…

[Production of cytokines from blood donor leukocytes].

Cytokine production by donor blood leukocytes was investigated in vitro. LPS stimulation led to the production of IL-1 beta, IL-6, and TNF-alpha, if their spontaneous production was below the threshold sensitivity of the test method, and to a statistically negligible increase of production if spontaneous production was pronounced. The concentrations of cytokines produced depended on the nature of inductors and their doses. The inductor modulating effect can be used for stimulation and inhibition of cytokine production.

Epidermal cytokines in murine cutaneous irritant responses.

Investigations on cytokines in skin irritancy or non-immunological irritant contact dermatitis (ICD) should improve our understanding of their complex mechanism. Numerous studies showed, however, that similar epidermal cytokines have been detected in irritant and allergic reactions, suggesting a lack of specific cytokines that clearly differentiate allergic from irritant reactions even though the pathomechanisms between allergic contact dermatitis (ACD) and ICD are distinguished. Recent data, however, indicate that some mediators may be restricted to allergic responses (contact hypersensitivit…

Osteoarthritis and cartilage: the role of cytokines.

The pathogenesis of osteoarthritis involves multiple etiologies, including mechanical, biochemical, and genetic factors that contribute to the imbalance in the synthesis and destruction of articular cartilage. It is now well documented that interleukin-1 and tumor necrosis factor-alpha are the predominant proinflammatory and catabolic cytokines involved in disease initiation and progression. Other proinflammatory cytokines may amplify or modulate this process, whereas anti-inflammatory cytokines, which are often detected, paradoxically, in osteoarthritis tissues, may counteract the tissue dest…

Intestinal nematode parasites, cytokines and effector mechanisms.

Laboratory models of intestinal nematode infection have played an important role in developing our understanding of the immune mechanisms th … ells of the Th2 type gain ascendancy then a protective immune response ensues, mediated by Th2 type cytokines and the effector mechanisms they control. In contrast, if an inappropriate Th1 type response predominates the ability to expel infection is compromised. Equally important is the observation that multiple potential effector mechanisms are stimulated …

[Analysis of contact dermatitis by cytokines. I].

The study of the participation of T cells and the kinetic studies of cytokines by the RT-PCR method in the challenge phase of contact hypersensitivity reaction (CHR) were performed using skin tissues collected at regular intervals from mice sensitized with 2,4-dinitro-1-fluorobenzene (DNFB). The results obtained from these studies and the further analyses using anti-CD4 and CD8 antibodies showed that the revelation of phenotype of lymph node cells at the reaction site and T lymphocytes at the CHR site were found to play important roles. Furthermore, we examined cytokines formed at the CHR site…

Circulating levels of cytokines in poststreptococcal glomerulonephritis.

Cellular immune mechanisms are important in the pathogenesis of acute glomerulonephritis, as underlined by recent demonstrations of cytokine activity in the urine and in the renal tissue of some of these patients. Therefore, we decided to study circulating levels of IL-6, TNF alpha and PDGF measured by ELISA in 12 patients with acute poststreptococcal glomerulonephritis (PSGN) on admission, at the time of discharge from the hospital, 45 days and 3 months later. We also studied 9 patients with acute streptococcal pharyngitis without nephritis, during acute infection, 8 and 21 days later and 20

Hypothesis: compartmentalization of cytokines in intraabdominal infection.

BACKGROUND: Although the proximal role of systemic cytokines in the infectious-inflammatory cascades is well recognized, the magnitude and meaning of its intraperitoneal levels in peritonitis have received little attention. We hypothesized that in peritonitis a significant and clinically relevant cytokine-mediated inflammatory response is compartmentalized in the peritoneal cavity. METHODS: MEDLINE was used to search the literature for all articles dealing with experimental, primary, and secondary bacterial peritonitis and cytokines. RESULTS: Bacterial peritonitis is associated with an immen…

Use of cytokines in clozapine-induced agranulocytosis.

OBJECTIVE: To report and review the use of cytokines for the treatment of clozapine-induced neutropenia. METHOD: Case report and review of literature. RESULTS: Cytokines, granulocyte colony-stimulating factor (G-CSF) and granulocyte-macrophage colony-stimulating factor (GM-CSF), appear to shorten the duration of clozapine-induced neutropenia. CONCLUSIONS: G-CSF or GM-CSF therapy should be considered in patients with profound neutropenia of prolonged duration (high-risk neutropenia).

Immunmodulating cytokines induce term and preterm parturition.

The aim of our study was to investigate if cytokines, which are able to cause preterm delivery is case of intraamniotic infection, also participate in the mechanisms of normal term labor. Therefore we estimated cytokine concentrations in cercicovaginal secretions of 96 patients, divided into four different collectives. In collective A (women with spontaneous term labor) cytokine concentrations raised from a median level of 278 pg/ml for Il-1 beta and 263 pg/ml for Il-6 in early term labor to a median level of 3351 pg/ml for Il-1 beta and 39,442 pg/ml for Il-6 at complete cervical dilatation. Activating mechanism of CNTF and related cytokines.

Ciliary neurotrophic factor (CNTF) shares structural and functional properties with members of the hematopoietic cytokine family. It is composed of a four-helix bundle structure and shares the transmembrane signal transducing proteins, glycoprotein-130 (gp130) and leukemia inhibitory factor receptor (LIF-R). Structure-function analysis showed that the gp130-interactive proteins bind in a similar manner to that of growth hormone (site I and II). In addition, gp130-interactive proteins and granulocyte colony-stimulating factor (G-CSF) utilize another binding site (site III) at the boundary betwe
