A Small-Molecule Inhibitor of Human DNA Polymerase η Potentiates the Effects of Cisplatin in Tumor Cells.

Translesion DNA synthesis (TLS) performed by human DNA polymerase eta (hpol η) allows tolerance of damage from cis-diamminedichloroplatinum(II) (CDDP or cisplatin). We have developed hpol η inhibitors derived from N-aryl-substituted indole barbituric acid (IBA), indole thiobarbituric acid (ITBA), and indole quinuclidine scaffolds and identified 5-((5-chloro-1-(naphthalen-2-ylmethyl)-1H-indol-3-yl)methylene)-2-thioxodihydropyrimidine-4,6(1H,5H)-dione (PNR-7-02), an ITBA derivative that inhibited hpol η activity with an IC50 value of 8 μM and exhibited 5-10-fold specificity for hpol η over repli

BCL6 represses CHEK1 and suppresses DNA damage pathways in normal and malignant B-cells.

BCL6 is a transcriptional repressor protein that is expressed in a developmentally regulated fashion during B-cell maturation. Specifically, BCL6 is required for formation of germinal centers in response to T-cell dependent antigen activation. Germinal center B-cells feature the ability to tolerate rapid proliferation and simultaneous genetic recombination. Genetic lesions that cause constitutive expression of BCL6 are commonly associated with diffuse large B-cell lymphomas (DLBCL). Recent studies show that BCL6 contributes to the germinal center phenotype by directly repressing genes involved

NER enzymes maintain genome integrity and suppress homologous recombination in the absence of exogenously induced DNA damage in Pseudomonas putida.

In addition to its prominence in producing genetic diversity in bacterial species, homologous recombination (HR) plays a key role in DNA repair and damage tolerance. The frequency of HR depends on several factors, including the efficiency of DNA repair systems as HR is involved in recovery of replication forks perturbed by DNA damage. Nucleotide excision repair (NER) is one of the major DNA repair pathways involved in repair of a broad range of DNA lesions generally induced by exogenous chemicals or UV-irradiation and its functions in the cells not exposed to DNA-damaging agents have attracted

Identification of the DNA repair defects in a case of Dubowitz syndrome.

Dubowitz Syndrome is an autosomal recessive disorder with a unique set of clinical features including microcephaly and susceptibility to tumor formation. Although more than 140 cases of Dubowitz syndrome have been reported since 1965, the genetic defects of this disease has not been identified. In this study, we systematically analyzed the DNA damage response and repair capability of fibroblasts established from a Dubowitz Syndrome patient. Dubowitz syndrome fibroblasts are hypersensitive to ionizing radiation, bleomycin, and doxorubicin. However, they have relatively normal sensitivities to m

The molecular basis of nonmelanoma skin cancer: new understanding.

Nonmelanoma skin cancers, including basal cell carcinomas and squamous cell carcinomas, represent the most common malignant neoplasms in humans. Although many environmental and genetic factors contribute to the development of skin cancers, the most important is chronic exposure to UV radiation in sunlight. We now appreciate that the role of UV in the development of nonmelanoma skin cancers is 2-fold. First, UV radiation causes mutations in cellular DNA. Failure to repair these genetic alterations ultimately leads to unrestrained growth and tumor formation. Second, UV radiation has profound eff

Biofilm and planktonic lifestyles differently support the resistance of the desert cyanobacterium Chroococcidiopsis under space and Martian simulations.

When Chroococcidiopsis sp. strain CCMEE 057 from the Sinai Desert and strain CCMEE 029 from the Negev Desert were exposed to space and Martian simulations in the dried status as biofilms or multilayered planktonic samples, the biofilms exhibited an enhanced rate of survival. Compared to strain CCMEE 029, biofilms of strain CCME 057 better tolerated UV polychromatic radiation (5 × 10(5) kJ/m(2) attenuated with a 0.1% neutral density filter) combined with space vacuum or Martian atmosphere of 780 Pa. CCMEE 029, on the other hand, failed to survive UV polychromatic doses higher than 1.5 × 10(3) k

Photothermal targeting of lactate metabolism reverses radioresistance via disrupting the immunosuppressive barrier.

Radiotherapy (RT) is frequently compromised by radioresistance and insufficient immunogenicity. Multiomics analysis here identifies a significant association between glycolysis and DNA damage repair, linking them to RT resistance and poor patient survival. To address this, we develop a mild thermia-guided radiosensitizer, GQD@PDA/Au, which reprograms glycolytic metabolism upon photothermal stimulation to inhibit lactate accumulation. Reduced lactate levels aggravate DNA damage and dsDNA breaks, thereby enhancing tumor radio-immunogenicity by activating the cGAS-STING pathway, inducing tumor IC

DNA damage and cell death induced by exposure to ultra-high dose rate low-dose pulsed X-rays emitted from a kilojoule plasma focus device.

BACKGROUND: FLASH radiotherapy, characterized by ultra-high dose rates (> 40 Gy/s), potentially spares normal tissues while maintaining antitumor efficacy (the FLASH effect). While electron and proton FLASH are explored, pulsed X-ray sources like plasma focus devices offer unique possibilities. Previous work has reported hyper-radiosensitivity in colorectal cancer cells exposed to ultra-high-dose-rate pulsed X-rays from a kilojoule plasma focus (PF) device, without significant effects on non-cancerous cells. This study further investigates the biological effects of ultra-high-dose-rate (~ 10⁷

Different radiation dose rate as radioprotection and the cross effect with chromium using in vivo somatic cells of Drosophila.

Ionizing radiation plays a key role in the adaptation of an individual organism to environmental pollution, at the same time, it has biological effects that depend on radiation intensity or dose rate (DR). Although the effect of DR has been studied in vitro, the phenomenon known as the inverse effect of DR, which indicates as it decreases that the induction of damage is greater, has not been widely studied in vivo. The present study is aimed to test 0.5 and 1 Gy in somatic cells of the wing of D. melanogaster, administered at 5.4 or 34.3 Gy/h and from 0.037 to 0.3 mM of CrO3 as conditioning tr

EFHD2 drives lactate-mediated DNA damage repair and immunosuppression via HMGB1 and HIF-1α to confer radioresistance in colorectal cancer.

Radioresistance in colorectal cancer (CRC) remains a critical clinical challenge, with underlying mechanisms involving deoxyribonucleic acid (DNA) damage repair, metabolic reprogramming, and immune evasion. Here, we employed multi-omics approaches, including ribonucleic acid (RNA) sequencing, proteomic profiling, and lactyl-proteomics of radioresistant CRC cell models, combined with co-immunoprecipitation, molecular docking, spatial transcriptomics, and single-cell RNA sequencing. Functional validation included production of an EF-hand domain family member D2 (EFHD2) lysine 9 (K9) lactylation-

Low dose radiation induces new protein synthesis that promotes cancer radiotherapy resistance.

Radiotherapy (RT) is a cornerstone of cancer treatment; however, its efficacy is frequently hampered by its adverse effects on normal tissues. By studying the effects of high-dose radiotherapy (HDRT) and low-dose radiotherapy (LDRT), we found that cancer cells adapt distinct responses to these doses to reduce cytotoxicity. Upon HDRT, cancThese authors contributed equally to this worker cells initiate a strong DNA damage response (DDR) to gain resistance through rapid production and/or activation of proteins for cell cycle arrest and DNA damage repair. In contrast, LDRT has a milder effect on t

Mitochondrial p32-mediated regulation of p-TBK1 affects the radiosensitivity of colorectal cancer.

PURPOSE: Many colorectal cancer (CRC) patients respond poorly to radiotherapy due to radioresistance. Understanding the molecular mechanisms underlying this resistance is crucial. It was demonstrated that p32, a mitochondrial protein translation regulator, is related to cancer development. However, its specific function and mechanism in CRC, has not yet been investigated. This study aims to explore the role of p32 in CRC and its impact on radiotherapy sensitivity. METHODS: Cell viability was evaluated by MTT and EdU assay. Mitochondrial DNA (mtDNA) leakage was quantified by RT-qPCR. Radiosens

XPA protein as a limiting factor for nucleotide excision repair and UV sensitivity in human cells.

Nucleotide excision repair (NER) acts on a variety of DNA lesions, including damage induced by many chemotherapeutic drugs. Cancer therapy with such drugs might be improved by reducing the NER capacity of tumors. It is not known, however to what extent any individual NER protein is rate-limiting for any step of the repair reaction. We studied sensitivity to UV radiation and repair of DNA damage with regard to XPA, one of the core factors in the NER incision complex. About 150,000-200,000 molecules of XPA protein are present in NER proficient human cell lines, and no XPA protein in the XP-A cel

Cellular responses to radiation and risk of breast cancer.

Mutations in the ataxia telangiectasia gene (ATM) result in an abnormal p53-mediated cellular response to DNA damage produced by ionising radiation. This deficiency is believed to contribute to the radiosensitivity and high cancer risk seen in ataxia telangiectasia (AT) patients and AT heterozygotes. Epidemiological studies have demonstrated that relatives of AT patients are particularly predisposed to breast cancer. This observation, together with the finding that a relatively high proportion of breast cancer patients display an abnormal severe reaction of normal tissues following radiotherap

Cell injury and repair resulting from sleep loss and sleep recovery in laboratory rats.

STUDY OBJECTIVES: Increased cell injury would provide the type of change in constitution that would underlie sleep disruption as a risk factor for multiple diseases. The current study was undertaken to investigate cell injury and altered cell fate as consequences of sleep deprivation, which were predicted from systemic clues. DESIGN: Partial (35% sleep reduction) and total sleep deprivation were produced in rats for 10 days, which was tolerated and without overtly deteriorated health. Recovery rats were similarly sleep deprived for 10 days, then allowed undisturbed sleep for 2 days. The plasm

The genotoxic stress-inducible gene GEI1(Orf19.3109) is a repressor of virulence in Candida albicans.

Transcriptional reprogramming under genotoxic stress is essential for the survival and pathogenesis of Candida albicans. While many DNA damage response (DDR) genes are well-characterized, the functions of pathogen-specific, stress-inducible genes remain poorly understood. Here, we characterized GEI1 (Orf19.3109), a gene significantly induced by genotoxic agents and oxidative stress, which lacks a clear ortholog in Saccharomyces cerevisiae. Interestingly, although GEI1 expression is triggered by DNA damage stress, its deletion does not impair genotoxin tolerance. Instead, the gei1Δ mutant shows

Liquid Chromatography-Mass Spectrometry Screening of Cyclophosphamide DNA Damage In Vitro and in Patients Undergoing Chemotherapy Treatment.

DNA alkylating drugs have been used as frontline medications to treat cancer for decades. Their chemical reaction with DNA leads to the blockage of DNA replication, which impacts cell replication. While this impacts rapidly dividing cancerous cells, this process is not selective and results in highly variable and often severe side effects in patients undergoing alkylating-drug based therapies. The development of biomarkers to identify patients who effectively respond with tolerable toxicities vs patients who develop serious side effects is needed. Cyclophosphamide (CPA) is a commonly used chem

Management Pearls on the Treatment of Actinic Keratoses and Field Cancerization.

Field cancerization (FC) is a chronic disease involving multiple clinical and subclinical actinic keratoses (AK) on large photo-exposed surfaces with multifocal areas of dysplasia and precancerous changes. Patients and treatment must be properly monitored and managed to avoid aggravation and progression of the disease. Management of actinic keratoses includes lesion-directed treatments, such as cryotherapy and field-directed therapies. Field-directed therapies may have the potential to address subclinical damage, reduce AK recurrence rates and potentially reduce the risk of squamous cell carci

MINERVA: A CubeSat for demonstrating DNA damage mitigation against space radiation in C. elegans by using genetic modification.

The ideas of deep-space human exploration, interplanetary travel, and space civilizations are becoming a reality. However, numerous hindrances remain standing in the way of accomplishing these feats, one of which is space ionizing radiation. Space ionizing radiation has become the most hazardous health risk for long-term human space exploration, as it can induce chromosomal damage and epigenetic changes. The Minerva mission aims to demonstrate cutting-edge technology to inhibit DNA damage against deep-space radiation exposure by using genetic modification. The concept of the experiment is to t
