2,2'-Dithiodipyridine (PySSPy) is a typical pyridine disulfide, which has been widely used in industry and biomedical research, and frequently identified as the photo-degradation product of ubiquitous metal pyrithiones. Although many disulfides can moderately react with physiological oxidants such as H2O2, it was rarely reported that the highly-reactive hydroxyl radicals (•OH) could be produced from the reaction of disulfides with H2O2. Here we unexpectedly found that PySSPy can readily react with H2O2 over a wide pH range to continuously produce •OH and lead to appreciable DNA damage. Compare…

To elucidate how a functional disulfide bond controls protein activity, it is essential that the redox state of the bond in the population of protein molecules is known. A differential cysteine alkylation and mass spectrometry technique is described that affords precise quantification of protein disulfide bond redox state. The utility of the technique is demonstrated by quantifying the redox states of the eight disulfide bonds in human coagulation factor VIII.

Carbon disulfide (CS2) exhibits unique chemical properties and multifunctional reactivity that render it indispensable in the synthesis of diverse organic molecules. Despite its simple structure, CS2 exhibits unique chemical properties that have established it as a versatile and indispensable reagent in organic synthesis. The electrophilic nature of the carbon center, combined with the nucleophilicity of the sulfur atoms, enables CS2 to participate in a wide array of reactions, making it a key sulfur source for the introduction of sulfur functionalities into organic molecules. The ambident nat

Carbon disulfide (CS2) is an environmental contaminant, which is deadly hazardous to the workers under chronic or acute exposure. However, the toxicity mechanisms of CS2 are still unclear due to the scarcity of biocompatible donors, which can release CS2 in cells. Here we developed the first bioorthogonal CS2 delivery system based on the "click-and-release" reactions between mesoionic 1,3-thiazolium-5-thiolates (TATs) and strained cyclooctyne exo-BCN-OH. We successfully realized intracellular CS2 release and investigated the causes of CS2-induced hepatotoxicity, including oxidative stress, pro

The cystine transporter solute carrier family 7 member 11 (SLC7A11) (also known as xCT) promotes glutathione synthesis and counters oxidative stress-induced cell death, including ferroptosis, by importing cystine. Also, SLC7A11 plays a crucial role in tumor development. However, recent studies have uncovered an unexpected role of SLC7A11 in promoting disulfidptosis, a novel form of regulated cell death induced by disulfide stress. In this review, we examine the opposing roles of SLC7A11 in regulating redox homeostasis and cell survival/death, summarize current knowledge on disulfidptosis, and
