Vasomotor effects of facial nerve stimulation: noncholinergic vasodilation in the eye.

The effect of facial nerve stimulation on ocular blood flow was studied in rabbits. The intracranial part of the facial nerve was stimulated electrically and the regional blood flow was measured with labelled microspheres. Effects on the intraocular pressure were determined in a separate series of experiments. Stimulation increased choroidal blood flow by about 200%. The blood flow of the iris and the ciliary processes also increased. The blood flow of the eyelids and the nictitating membrane increased by approximately 1000%, and the blood flow of Harder's gland increased by about 200%. The bl

Alterations in excitatory amino acid-mediated regulation of midbrain dopaminergic neurones induced by chronic psychostimulant administration and stress: relevance to behavioural sensitization and drug addiction.

Repeated, intermittent administration of the psychostimulants d-amphetamine and cocaine, as well as other drugs of abuse, leads to an enduring augmentation of certain behavioural responses (e.g. locomotor activity) produced by these drugs. This behavioural sensitization has been the subject of considerable interest due to its potential relevance to drug addiction. Repeated administration of d-amphetamine also leads to an enhancement in the ability of electrical stimulation of the prefrontal cortex to induce burst firing in midbrain dopaminergic (DA) neurones. This hyper-responsiveness probably

Negative allosteric modulation of mGlu7 disrupts fear memory reconsolidation and glutamatergic signaling in rat and human brain tissue.

Anxiety- and stress-related disorders are amongst the most frequent neurological disorders, and efficient treatment is lacking. Metabotropic glutamate receptors (mGlu) have emerged as promising targets for intervention. Of particular interest is mGlu7, because of its expression in the lateral amygdala (LA), a region critical for fear learning. In the present study we examined the effects of the highly specific negative allosteric modulator of mGlu7 (ADX71743) on fear memory reconsolidation. Our investigation unveils that infusion in rats of ADX71743 in the LA or subcutaneously disrupts the rec

Type II Cells in the Human Carotid Body Display P2X7 Receptor and Pannexin-1 Immunoreactivity.

The carotid body is a peripheral chemoreceptor that consists of clusters of chemoreceptive type I cells, glia-like type II cells, afferent and efferent nerves, and sinusoidal capillaries and arterioles. Cells and nerves communicate through reciprocal chemical synapses and electrical coupling that form a "tripartite synapse," which allows for the process of sensory stimuli within the carotid body involving neurotransmission, autocrine, and paracrine pathways. In this network there are a variety of neurotransmitters and neuromodulators including adenosine 5'-triphosphate (ATP). Carotid body cell

Microglial infiltration mediates cognitive dysfunction in rat models of hypothalamic obesity via a hypothalamic-hippocampal circuit involving the lateral hypothalamic area.

This study aimed to explore the mechanism underlying cognitive dysfunction mediated by the lateral hypothalamic area (LHA) in a hypothalamic-hippocampal circuit in rats with lesion-induced hypothalamic obesity (HO). The HO model was established by electrically lesioning the hypothalamic nuclei. The open field (OP) test, Morris water maze (MWM), novel object recognition (NOR), and novel object location memory (NLM) tests were used to evaluate changes in cognition due to alterations in the hypothalamic-hippocampal circuit. Western blotting, immunohistochemical staining, and cholera toxin subunit

Regulation of synaptic transmission at the photoreceptor terminal: a novel role for the cation-chloride co-transporter NKCC1.

The Na(+)-K(+)-2Cl(-) co-transporter type 1 (NKCC1) is localized primarily throughout the outer plexiform layer (OPL) of the distal retina, a synaptic lamina that is comprised of the axon terminals of photoreceptors and the dendrites of horizontal and bipolar cells. Although known to play a key role in development, signal transmission and the gating of sensory signals in other regions of the retina and in the CNS, the contribution of NKCC1 to synaptic transmission within the OPL is largely unknown. In the present study, we investigated the function of NKCC1 at the photoreceptor-horizontal cell

Neurotransmitter Release Can Be Stabilized by a Mechanism That Prevents Voltage Changes Near the End of Action Potentials from Affecting Calcium Currents.

UNLABELLED: At chemical synapses, presynaptic action potentials (APs) activate voltage-gated calcium channels, allowing calcium to enter and trigger neurotransmitter release. The duration, peak amplitude, and shape of the AP falling phase alter calcium entry, which can affect neurotransmitter release significantly. In many neurons, APs do not immediately return to the resting potential, but instead exhibit a period of depolarization or hyperpolarization referred to as an afterpotential. We hypothesized that presynaptic afterpotentials should alter neurotransmitter release by affecting the elec

The Coupling between Ca2+ Channels and the Exocytotic Ca2+ Sensor at Hair Cell Ribbon Synapses Varies Tonotopically along the Mature Cochlea.

The cochlea processes auditory signals over a wide range of frequencies and intensities. However, the transfer characteristics at hair cell ribbon synapses are still poorly understood at different frequency locations along the cochlea. Using recordings from mature gerbils, we report here a surprisingly strong block of exocytosis by the slow Ca2+ buffer EGTA (10 mM) in basal hair cells tuned to high frequencies (∼30 kHz). In addition, using recordings from gerbil, mouse, and bullfrog auditory organs, we find that the spatial coupling between Ca2+ influx and exocytosis changes from nanodomain in

The Alzheimer's disease risk gene BIN1 regulates activity-dependent gene expression in human-induced glutamatergic neurons.

Bridging Integrator 1 (BIN1) is the second most important Alzheimer's disease (AD) risk gene, but its physiological roles in neurons and its contribution to brain pathology remain largely elusive. In this work, we show that BIN1 plays a critical role in the regulation of calcium homeostasis, electrical activity, and gene expression of glutamatergic neurons. Using single-cell RNA-sequencing on cerebral organoids generated from isogenic BIN1 wild type (WT), heterozygous (HET) and homozygous knockout (KO) human-induced pluripotent stem cells (hiPSCs), we show that BIN1 is mainly expressed by olig

Simulating the impact of photoreceptor loss and inner retinal network changes on electrical activity of the retina.

Objective.A major reason for poor visual outcomes provided by existing retinal prostheses is the limited knowledge of the impact of photoreceptor loss on retinal remodelling and its subsequent impact on neural responses to electrical stimulation. Computational network models of the neural retina assist in the understanding of normal retinal function but can be also useful for investigating diseased retinal responses to electrical stimulation.Approach.We developed and validated a biophysically detailed discrete neuronal network model of the retina in the software package NEURON. The model inclu

A Single Quasi-Vertical van der Waals Heterostructure Memristor as Opto-Reconfigurable Artificial Intelligence Hardware.

The explosive advancement of artificial intelligence (AI) has driven vast amounts of unstructured data that directly emerges at sensory interaction interfaces. Memristors with superior computing ability are among the most promising computing blocks. However, conventional vertical Metal/Resistive layer/Metal (V-MRM) memristors tend to generate intense electric fields, suffer from limited sensory response, inflexible, and rigid functional modes, as well as long-standing variability and overshoot issues. Here, we present an unconventional two-terminal quasi-vertical (QV) van der Waals heterostruc

Dynamin-1 deletion enhances post-tetanic potentiation and quantal size after tetanic stimulation at the calyx of Held.

KEY POINTS: Post-tetanic potentiation (PTP) is attributed mainly to an increase in release probability (Pr ) and/or readily-releasable pool (RRP) in many synapses, but the role of endocytosis in PTP is unknown. Using the calyx of Held synapse from tissue-specific dynamin-1 knockout (cKO) mice (P16-20), we report that cKO synapses show enhanced PTP compared to control. We found significant increases in both spontaneous excitatory postsynaptic current (spEPSC) amplitude and RRP size (estimated by a train of 30 APs at 100 Hz) in cKO over control during PTP. Actin depolymerization blocks the incre

Synaptic plasticity in neurodegenerative diseases evaluated and modulated by in vivo neurophysiological techniques.

Several studies demonstrated in experimental models and in humans synaptic plasticity impairment in some neurodegenerative and neuropsychiatric diseases such as Parkinson's disease, Alzheimer's disease, Huntington's disease, and schizophrenia. Recently new neurophysiological tools, such as repetitive transcranial magnetic stimulation and transcranial direct current stimulation, have been introduced in experimental and clinical settings for studying physiology of the brain and modulating cortical activity. These techniques use noninvasive transcranial electrical or magnetic stimulation to modul

Synaptic behaviour in the myenteric plexus of the guinea-pig gastric antrum.

1. Intracellular recording methods were used to study the synaptic behaviour of neurones in the myenteric plexus of the guinea-pig gastric antrum. Synaptic potentials occurred spontaneously or were evoked by focal electrical stimulation of interganglionic fibre tracts. Synaptic events consisted of fast and slow excitatory postsynaptic potentials (EPSPs) and inhibitory postsynaptic potentials (IPSPs). 2. Fast EPSPs with durations less than 20 ms were evoked in every antral neurone in a population sample of 370 cells. Most of the ganglion cells received multiple inputs from axons entering the in

Al-Stoichiometry-Engineered Defect Profiles in InAlZnO Nanofibers for Balanced Charge Transport and Optical Plasticity.

Metal oxide optoelectronic synapses are promising for neuromorphic vision, but combining a low dark current with sustained photoinduced plasticity remains challenging. Here, Al stoichiometry is engineered in electrospun In1AlxZn0.08O (x = 0.007, 0.011, and 0.015) nanofiber networks to tune sub-bandgap defects, carrier transport, and photocarrier relaxation. Optical, spectroscopic, electrical, and synaptic measurements identify 1.1 atomic % Al as optimal. Strong Al-O bonding suppresses excess oxygen-vacancy-related donor states while preserving the In 5s transport network. The optimized device

Designing Light-Sensitive Organic Semiconductors with Azobenzenes for Photoelectrochemical Transistors as Neuromorphic Platforms.

Organic neuromorphic electronics aim to emulate the adaptive behavior of biological synapses using soft, biocompatible materials capable of analog and stimulus-responsive modulation. While azobenzene-based semiconductors provide reversible light-induced switching, their application in mixed ionic-electronic conductors for neuromorphic systems remains largely unexplored. In this study, photoresponsive organic photoelectrochemical transistors (OPECTs) are engineered by functionalizing PEDOT:PSS with azobenzene derivatives bearing nitro or fluorine substituents. These modifications alter the elec

Some neural effects of adenosin.

The possible neural effects of adenosine were investigated by using electrophysiological techniques at the level of some central and peripheral synapses. The evoked potentials in the somatosensorial cerebral cortex are influenced according to both the type of administration and the level of the electrical stimulation. While the local application does not induce significant alterations, the intrathalamic injections and the perfusion of the IIIrd cerebral ventricle do change the distribution of activated units at the level of different cortical layers especially during the peripheral stimulation

Endplate contributions to the safety factor for neuromuscular transmission.

The neuromuscular junction (NMJ), for most extremity and axial skeletal muscle fibers, with the exception of extraocular, middle ear, and some facial and pharyngeal muscles, is a 'slave' synapse that is designed to activate the muscle fiber every time the nerve terminal is activated. The fidelity of the NMJ hinges upon the electrical depolarization produced by activation of acetylcholine receptors (AChRs), called the endplate potential (EPP), being larger than is needed to trigger an action potential (AP) in the skeletal muscle fiber. The safety factor (SF) is a measure of how much larger the

The effects of selective electrical stimulation of the rat cochlea on hippocampal field potentials.

The hippocampus is a brain structure well known for its importance to spatial learning and memory. As such it is assumed to use multisensory integration in order to generate mathematical maps of the spatial environment. Auditory inputs to the hippocampus have been described to a limited extent. Although it has been demonstrated that natural auditory stimulation can evoke local field potentials (LFPs) and single neuron responses in the hippocampus, and that noise trauma and tinnitus can cause maladaptive hippocampal plasticity, to the best of our knowledge no one has selectively, electrically s
