Influence of tick-borne encephalitis and West Nile viruses on the chromosomes of pig kidney cells.

Changes of the mitotic index (MI) were studied in pig kidney (PS) cells infected either with tick-borne encephalitis (TBE) virus strains Hypr and Ir-32, or with West Nile (WN) virus prototype strain, strains K-99 and E-13. The cell division was arrested by the infection (metaphase barrier). The effect of TBE virus strains was manifested by the appearance of lagging chromosomes while the action of WN virus strains by colchicine--like metaphases. The prototype TBE virus strain Hypr affected the chromosomes and mitotic apparatus of PS cells less markedly than did the freshly isolated Ir-32 strain

Development of a triplex RT-qPCR assay for simultaneous quantification of Japanese encephalitis, Murray Valley encephalitis, and West Nile viruses for environmental surveillance.

The co-circulation of mosquito-borne Japanese encephalitis virus (JEV), Murray Valley encephalitis virus (MVEV), and West Nile virus (WNV) has impacted human and animal health in multiple countries worldwide. To facilitate early warnings and surveillance of the presence of these viral infectious agents in the environment, a triplex reverse transcription-quantitative PCR (RT-qPCR) was developed for simultaneous quantification of JEV, MVEV, and WNV in potential hotspots such as piggery and urban wastewater and environmental water samples. The performance of the developed triplex RT-qPCR assay wa

Cross-protection between West Nile and Japanese encephalitis viruses in red-winged blackbirds (Agelaius phoeniceus).

Similar to West Nile virus (WNV), Japanese encephalitis virus (JEV) has a history of intercontinental spread, and birds are important for the maintenance and transmission of both of these closely related viruses. We examined viremic and serologic responses of blackbirds (Agelaius phoeniceus), with and without immunity to WNV, following experimental inoculation with two strains of JEV. Japanese encephalitis (JE) viremia was detected in only one of 16 (6.3%) WNV-immune birds, while all 16 nonimmune birds had detectable JE viremia. Two weeks after JEV inoculation, all birds without pre-existing W

Phylogenetic and virulence analysis of tick-borne encephalitis viruses from Japan and far-Eastern Russia.

We have previously reported that tick-borne encephalitis (TBE) is endemic in a specific area of Hokkaido, Japan. In Oshima, the southern part of Hokkaido, TBE virus was isolated from sentinel dogs, ticks and rodents in 1995 and 1996. To identify when these TBE viruses emerged in Hokkaido, the times of divergence of TBE virus strains isolated in Oshima and far-eastern Russia were estimated. TBE virus was isolated in Khabarovsk in 1998 and the nucleotide sequences of viral envelope protein genes of isolates from Oshima and Khabarovsk were compared. From the synonymous substitution rate of these

Antigenic analysis of tick-borne encephalitis complex viruses by time-resolved fluoroimmunoassay with monoclonal antibodies.

The antigenic structure of 5 strains of tick-borne encephalitis (TBE) virus and 7 other viruses of the TBE complex was examined by the highly sensitive and specific technique of time-resolved fluoroimmunoassay (TR-FIA). A collection of 8 monoclonal antibodies to the Austrian strain. Neudörfl, was used in this study. The findings demonstrate the uniformity of the antigenic structure of TBE viruses from different geographic regions of the USSR. In addition, an epitope was detected which is characteristic of western variants of TBE virus, and another epitope was detected which permits the differe

[Survival of tick-borne encephalitis complex viruses in a brain cell culture from suckling mice].

The evidence of long-term survival of the tick-borne encephalitis complex viruses in primarily trypsinized brain cells of suckling mice inoculated in vivo is presented. The viability of the brain cell cultures from suckling mice inoculated with an attenuated strain was shown to be higher than that of a similar culture of the brain cells from suckling mice infected with a virulent strain. The infectious virus could be recovered for over 200 days of the existence of these cultures. The experimental results appear to be another confirmation of the capacity of viruses of the tick-borne encephaliti
