CD8+ cytotoxic T cell responses to dominant tumor-associated antigens are profoundly weakened by aging yet subdominant responses retain functionality and expand in response to chemotherapy.

Increasing life expectancy is associated with increased cancer incidence, yet the effect of cancer and anti-cancer treatment on elderly patients and their immune systems is not well understood. Declining T cell function with aging in response to infection and vaccination is well documented, however little is known about aged T cell responses to tumor antigens during cancer progression or how these responses are modulated by standard chemotherapy. We examined T cell responses to cancer in aged mice using AE17sOVA mesothelioma in which ovalbumin (OVA) becomes a 'spy' tumor antigen containing one

Targeting of non-dominant antigens as a vaccine strategy to broaden T-cell responses during chronic viral infection.

In this study, we compared adenoviral vaccine vectors with the capacity to induce equally potent immune responses against non-dominant and immunodominant epitopes of murine lymphocytic choriomeningitis virus (LCMV). Our results demonstrate that vaccination targeting non-dominant epitopes facilitates potent virus-induced T-cell responses against immunodominant epitopes during subsequent challenge with highly invasive virus. In contrast, when an immunodominant epitope was included in the vaccine, the T-cell response associated with viral challenge remained focussed on that epitope. Early after c

Development a novel in silico mRNA vaccine against Clostridium difficile.

Clostridium difficile, a toxin-producing bacterium, can lead to conditions ranging from mild diarrhea to severe pseudomembranous colitis. Our research introduces an innovative strategy for developing a multi-epitope mRNA vaccine using in-silico methods targeting proteins involved in Clostridium difficile infection (CDI). Through computational models, we identified epitopes for B cells, helper T lymphocytes (HTL), and cytotoxic T lymphocytes (CTL) from protein sequences, assessing their immune response potential, allergenicity, toxicity, and autoimmune risk. The constructed 3D model underwent i

The Immunogenicity of an Artificial Tick-Borne Encephalitis Virus Polyepitope Immunogen May Depend on Spacers Flanking Epitope.

The development of artificial polyepitope immunogens based on the conserved CD4+ and CD8+ epitopes from different virus proteins combined into a single molecule is a promising approach in the development of vaccines for activating a specific T-cell response. Selection of spacer sequences flanking the epitopes within the construct and providing the optimal processing of such immunogens within the cell is an important step in the design of such vaccines. We report the results of designing polyepitope T-cell immunogens of tick-borne encephalitis virus that differ in the presence or absence of opt

Selection and fine-tuning of the autoimmune T-cell repertoire.

The immune system must avoid aggressive T-cell responses against self-antigens. But, paradoxically, exposure to self-peptides seems to have an important role in positive selection in the thymus and the maintenance of a broad T-cell repertoire in the periphery. Recent experiments have highlighted situations that allow high-avidity self-reactive T cells to avoid negative selection in the thymus. Accumulating evidence indicates that other, non-deleting mechanisms control the avidity with which T cells recognize self-antigens--a phenomenon that is known as 'tuning'. This might maximize the periphe

Immunoinformatics Design of a Multiepitope Vaccine (MEV) Targeting Streptococcus mutans: A Novel Computational Approach.

Dental caries, a persistent oral health challenge primarily linked to Streptococcus mutans, extends its implications beyond dental decay, affecting over 4 billion individuals globally. Despite its historical association with childhood, dental caries often persists into adulthood with prevalence rates ranging from 60 to 90% in children and 26 to 85% in adults. Currently, there is a dearth of multiepitope vaccines (MEVs) specifically designed to combat S. mutans. To address this gap, we employed an immunoinformatics approach for MEV design, identifying five promising vaccine candidates (PBP2X, P

Rational design of chimeric Multiepitope Based Vaccine (MEBV) against human T-cell lymphotropic virus type 1: An integrated vaccine informatics and molecular docking based approach.

Human T-cell lymphotropic virus type 1 (HTLV-1) is an infectious virus that has been linked to adult T cell leukemia /lymphoma, aggressive CD4-T cell malignancy and many other immune-related medical illnesses. So far, no effective vaccine is known to combat HTLV-1, hence, the current research work was performed to design a potential multi-epitope-based subunit vaccine (MEBV) by adopting the latest methodology of reverse vaccinology. Briefly, three highly antigenic proteins (Glycoprotein, Accessory protein, and Tax protein) with no or minimal (<37%) similarity with human proteome were sorted ou

Bile duct epithelia as target cells in primary biliary cirrhosis and primary sclerosing cholangitis.

Primary biliary cirrhosis (PBC) and primary sclerosing cholangitis (PSC) are chronic autoimmune-mediated diseases of the biliary tree, resulting in a loss of bile ducts. There are morphological features that clearly distinguish them from each other: in PBC, there is overt destruction of the bile ducts with disruption of the basement membrane; in PSC there is abundant periductular fibrosis with shrinkage and subsequent loss of the bile ducts. In order to see if the disparate histopathology is paralleled by different immunohistology we looked at a panel of epitopes on bile duct epithelia especia

Identification and selection of immunodominant B and T cell epitopes for dengue multi-epitope-based vaccine.

Dengue virus (DENV) comprises four serotypes (DENV1-4) which cause 390 million global infections with 500,000 hospitalizations and 25,000 fatalities annually. Currently, the only FDA approved DENV vaccine is the chimeric live-attenuated vaccine, Dengvaxia®, which is based on the yellow fever virus (YFV) genome that carries the prM and E genes of the respective DENV 1, 2, 3, and 4 serotypes. However, it has lower efficacies against serotypes DENV1 (51%) and DENV2 (34%) when compared with DENV3 (75%) and DENV4 (77%). The absence of T cell epitopes from non-structural (NS) and capsid (C) proteins

Trivalent multi-epitope mRNA vaccine against norovirus, rotavirus, and adenovirus 40/41: epitope screening, molecular docking, and molecular dynamics simulation with in silico validation guided by immunoinformatics.

BACKGROUND: Diarrheal diseases constitute a major global public health threat, particularly endangering young children, the elderly, and immunocompromised individuals. Three key pathogens-norovirus, rotavirus, and adenovirus 40/41-can induce dehydration, electrolyte imbalances, and severe complications, resulting in tens of thousands of deaths annually. Conventional vaccines have inherent limitations, including relatively long development cycles and high production costs. With the deep integration of bioinformatics and immunology, immunoinformatic techniques driven by high-throughput analysis

In-silico design and evaluation of a novel mRNA vaccine against human bocavirus 1: A neglected viral pathogen.

Human Bocavirus 1 (HBoV1) majorly affects infants and children, resulting severe lower respiratory tract infections. Since its first identification in 2005, HBoV1 has been detected in up to 18 % of respiratory infection cases, with a chronicity rate ranging from 2 % to 19 % in pediatric populations. Despite its considerable clinical impact, no specific vaccines or targeted therapies are currently available. This study aimed to design a chimera vaccine via reverse vaccinology and immunoinformatics approaches. T-cell and B-cell epitopes were derived and subsequently screened for non-allergenicit

Epitope-based peptide vaccine design and target site depiction against Middle East Respiratory Syndrome Coronavirus: an immune-informatics study.

BACKGROUND: Middle East Respiratory Syndrome Coronavirus (MERS-COV) is the main cause of lung and kidney infections in developing countries such as Saudi Arabia and South Korea. This infectious single-stranded, positive (+) sense RNA virus enters the host by binding to dipeptidyl-peptide receptors. Since no vaccine is yet available for MERS-COV, rapid case identification, isolation, and infection prevention strategies must be used to combat the spreading of MERS-COV infection. Additionally, there is a desperate need for vaccines and antiviral strategies. METHODS: The present study used immuno

Reverse vaccinology-based multi-epitope vaccine design against Indian group A rotavirus targeting VP7, VP4, and VP6 proteins.

Rotavirus, a primary contributor to severe cases of infantile gastroenteritis on a global scale, results in significant morbidity and mortality in the under-five population, particularly in middle to low-income countries, including India. WHO-approved live-attenuated vaccines are linked to a heightened susceptibility to intussusception and exhibit low efficacy, primarily attributed to the high genetic diversity of rotavirus, varying over time and across different geographic regions. Herein, molecular data on Indian rotavirus A (RVA) has been reviewed through phylogenetic analysis, revealing G1

Efficient induction of CD8 T-associated immune protection by vaccination with mRNA transfected dendritic cells.

Dendritic cells are excellent targets for antigen-specific immune intervention. Here we attempted to introduce a CD8 T cell-dependent epitope into dendritic cells for presentation on major histocompatibility complex class I and induction of immunity. Murine bone-marrow-derived dendritic cells were subjected to electroporation with mRNA transcribed in vitro from a plasmid encoding lymphocytic choriomeningitis virus glycoprotein or enhanced green fluorescent protein under the control of a T7 promotor. The transfection efficiency of dendritic cells was 22 to 40%. Maturation was not inhibited by t
