Structure of Saccharomyces cerevisiae chitinase 1 and screening-based discovery of potent inhibitors.

Chitinases hydrolyse the beta(1,4)-glycosidic bonds of chitin, an essential fungal cell wall component. Genetic data on a subclass of fungal family 18 chitinases have suggested a role in cell wall morphology. Specific inhibitors of these enzymes would be useful as tools to study their role in cell wall morphogenesis and could possess antifungal properties. Here, we describe the crystallographic structure of a fungal "plant-type" family 18 chitinase, that of Saccharomyces cerevisiae CTS1. The enzyme is active against 4-methylumbelliferyl chitooligosaccharides and displays an unusually low pH op

Synthesis, Antifungal Activity, and Potential Mechanism of Natural Product Pogostone Derivatives Containing an Acylhydrazone Scaffold.

The current study designed and synthesized a series of acylhydrazone derivatives based on the natural product pogostone. Among them, compound 2a15 exhibited potent fungicidal activity against Valsa mali (V. mali) (EC50 = 0.34 μg/mL) and Sclerotinia sclerotiorum (S. sclerotiorum) (EC50 = 0.45 μg/mL). At 200 μg/mL, 2a15 exhibited 81.26 and 82.08% efficacies against V. mali and S. sclerotiorum, respectively. Qualitative structure-activity analysis showed that when the benzene ring was substituted by a bromine atom, chlorine atom, or electron-withdrawing group, compounds with the substituent group

Genetic Manipulation of Cryptococcus neoformans.

Cryptococcus neoformans is an opportunistic fungal pathogen, which causes life-threatening meningoencephalitis in immunocompromised individuals and is responsible for more than 1,000,000 infections and 600,000 deaths annually worldwide. Nevertheless, anti-cryptococcal therapeutic options are limited, mainly because of the similarity between fungal and human cellular structures. Owing to advances in genetic and molecular techniques and bioinformatics in the past decade, C. neoformans, belonging to the phylum basidiomycota, is now a major pathogenic fungal model system. In particular, genetic ma

Renal fungal ball: an unusual sonographic finding.

Fungal infections of the urinary tract tend to occur in the drainage structures instead of the renal parenchyma. In patients with systemic candidiasis, the kidney is vulnerable to the formation of cortical abscesses or obstructive intrarenal masses ("fungal balls"), usually at the ureteropelvic junction. We describe the case of a boy who presented with dysuria, fever, and chills. Sonographic examination showed mild enlargement of both kidneys and moderate dilatation of the pelvicaliceal system bilaterally. A well-defined, echogenic, oval, mobile mass measuring 2.5 x 2.0 cm, without posterior a

Human Brain Organoids: A New Model to Study Cryptococcus neoformans Neurotropism.

With the rise in immunocompromised individuals and patients with immune-related comorbidities such as COVID-19, the rate of fungal infections is growing. This increase, along with the current plateau in antifungal drug development, has made understanding the pathogenesis and dissemination of these organisms more pertinent than ever. The mouse model of fungal infection, while informative on a basic scientific level, has severe limitations in terms of translation to the human disease. Here we present data supporting the implementation of the human cerebral organoid model, which is generated from

Effect of anti-glycosphingolipid monoclonal antibodies in pathogenic fungal growth and differentiation. Characterization of monoclonal antibody MEST-3 directed to Manpalpha1-->3Manpalpha1-->2IPC.

BACKGROUND: Studies carried out during the 1990's demonstrated the presence of fungal glycoinositol phosphorylceramides (GIPCs) with unique structures, some of them showed reactivity with sera of patients with histoplasmosis, paracoccidioidomycosis or aspergillosis. It was also observed that fungal GIPCs were able to inhibit T lymphocyte proliferation "in vitro", and studies regarding the importance of these molecules to fungal survival showed that many species of fungi are vulnerable to inhibitors of sphingolipid biosynthesis. RESULTS: In this paper, we describe a detailed characterization o

Comparing the Folds of Prions and Other Pathogenic Amyloids.

Pathogenic amyloids are the main feature of several neurodegenerative disorders, such as Creutzfeldt⁻Jakob disease, Alzheimer’s disease, and Parkinson’s disease. High resolution structures of tau paired helical filaments (PHFs), amyloid-β(1-42) (Aβ(1-42)) fibrils, and α-synuclein fibrils were recently reported using cryo-electron microscopy. A high-resolution structure for the infectious prion protein, PrPSc, is not yet available due to its insolubility and its propensity to aggregate, but cryo-electron microscopy, X-ray fiber diffraction, and other approaches have

Infection with a novel derogenid trematode in a Flap-necked chameleon (Chamaeleo dilepis).

A 2-year-old female Flap-necked chameleon (Chamaeleo dilepis) was presented with a history of anorexia. On physical examination, three 11×4×2 mm, light tan-pink, raised, gelatinous, freely movable, poorly demarcated, raised nodules were identified within the oral cavity. An impression smear contained multiple, light blue to green structures exhibiting an 18×35 μm, elliptical-shaped anterior portion and a single 1×150 μm filament with a tapered end. Initial differential etiologies included trematode ova, flagellate protozoa, fungal sporangia, plant material, and sperm. Fungal culture yielded no

Protostane and fusidane triterpenes: a mini-review.

Protostane triterpenes belong to a group of tetracyclic triterpene that exhibit unique structural characteristics. Their natural distribution is primarily limited to the genus Alisma of the Alismataceae family, but they have also been occasionally found in other plant genera such as Lobelia, Garcinia, and Leucas. To date, there are 59 known protostane structures. Many of them have been reported to possess biological properties such as improving lipotropism, hepatoprotection, anti-viral activity against hepatitis B and HIV-I virus, anti-cancer activity, as well as reversal of multidrug resistan

Effective Neutrophil Phagocytosis of Aspergillus fumigatus Is Mediated by Classical Pathway Complement Activation.

Aspergillus fumigatus is an important airborne fungal pathogen and a major cause of invasive fungal infections. Susceptible individuals become infected via the inhalation of dormant conidia. If the immune system fails to clear these conidia, they will swell, germinate and grow into large hyphal structures. Neutrophils are essential effector cells for controlling A. fumigatus infection. In general, opsonization of microbial particles is crucial for efficient phagocytosis and killing by neutrophils. Although the antibodies present in human serum do bind to all fungal morphotypes, we observed no

Chemoenzymatic formal synthesis of (+)-brazilin.

Herein, the manuscript presents a chemoenzymatic formal synthetic route of (+)-brazilin, a homoisoflavonoid natural product with a chroman skeleton cis-fused with a 2,3-dihydro-1H-indene unit, which is isolated from the traditional Chinese medicine, Caesalpinia sappan L. The key feature of the synthetic strategy includes an enzyme-mediated desymmetrization by employing lipase from Candida antarctica type B (CALB) and a one-pot SN2/hydrolysis reaction.

Backbone resonance assignment of mucoricin: a step towards structural insights into mucor pathogenesis.

Mucoricin is a key virulence factor in mucormycosis, as it targets host endothelial cells and inhibits protein translation through its N-glycosylase activity, leading to vascular leak and increased vascular permeability, thereby contributing to further disease progression. Its structure is quite unique, being structurally homologous to the Ricin B chain as confirmed by cross-reactivity with monoclonal anti-Ricin B chain antibodies, while exhibiting enzymatic activity similar to the Ricin A chain. Here, we report near-complete backbone (Soliman in Nat Microbiol 6(3):313–326, 2021) 1H, 15N, and

A novel starch-binding laccase from the wheat pathogen Zymoseptoria tritici highlights the functional diversity of ascomycete laccases.

BACKGROUND: Laccases are multicopper oxidases, which are assigned into auxiliary activity family 1 (AA1) in the CAZy database. These enzymes, catalyzing the oxidation of phenolic and nonphenolic substrates coupled to reduction of O2 to H2O, are increasingly attractive as eco-friendly oxidation biocatalysts. Basidiomycota laccases are well characterized due to their potential in de-lignification of lignocellulose. By contrast, insight into the biochemical diversity of Ascomycota counterparts from saprophytes and plant pathogens is scarce. RESULTS: Here, we report the properties of the laccase

Cryo-Electron Tomography of Candida glabrata Plasma Membrane Proteins.

Fungal plasma membrane proteins have long been recognized as targets for the development of antifungal agents. Despite recent progress in experimental approaches and computational structural predictions, our knowledge of the structural dynamics and spatial distribution of these membrane proteins in the context of their native lipid environment remains limited. By applying cryo-electron tomography (cryoET) and subtomogram analysis, we aim to characterize the structural characteristics and spatial distribution of membrane proteins present in Candida glabrata plasma membranes. This study has resu

High microbial diversity in the rhizosphere and soil improves carrot (Daucus carota L.) postharvest storability.

Postharvest diseases can cause significant losses in carrot (Daucus carota) yield during storage. In this study, the effects of soil quality and microbial diversity in bulk field soil and the rhizosphere on carrot postharvest storability were investigated. Microbial genera in bulk soil and rhizosphere samples were identified by Illumina MiSeq amplicon sequencing of the bacterial 16S rRNA gene and fungal ITS regions. Disease symptoms in the stored carrots were monitored after 6 months of cold storage, and the main pathogens were identified. Field sites with mineral soils, which had higher pH va

Deep Fungal Stromal Keratitis After Cataract Surgery: A Case Report.

Fungal keratitis is a vision-threatening infection that typically originates at the ocular surface and gradually invades the deeper layers of the cornea. We report a rare fungal keratitis confined to the deep stromal layer that developed following cataract surgery. An 81-year-old woman presented with a creeping white opacity on the posterior corneal surface in the right eye. Anterior segment optical coherence tomography revealed a hyper-reflective lesion at the posterior cornea, and in vivo confocal microscopy showed filamentous structures in the deep stroma, supporting the diagnosis of fungal

Histology-Correlated FTIR Chemical Imaging of Fungal Infection-Associated Tissue Compartments in Human Skin: A Proof-of-Concept Study.

Background/Objectives: Fungal skin infections are commonly assessed using clinical examination and conventional histopathology, including hematoxylin-eosin (HE), periodic acid-Schiff (PAS), and Grocott methenamine silver (GMS) staining. However, these methods provide limited spatially resolved biochemical information. This proof-of-concept study investigated whether Fourier transform infrared (FTIR) chemical imaging can identify biochemical patterns associated with histologically defined fungal infection-associated tissue compartments in human skin. Methods: Archived formalin-fixed, paraffin-e

Fine-needle aspiration biopsy of disseminated sporotrichosis: a case report.

In this report, we describe a case of disseminated sporotrichosis that was diagnosed by fine-needle aspiration biopsy (FNAB). The cytologic smears exhibited a large number of macrophages, few polymorphonuclear neutrophils and numerous round or oval, sometimes elongated, isolated and scattered yeast-like structures localized extracellularly or inside macrophages. These structures were clearly visualized by Giemsa and Papanicolaou methods. Cultures from skin biopsy material revealed fungal colonies which were subsequently identified as Sporothrix schenckii. The cytologic aspects, the correlation
