Galectin-3 Activation and Inhibition in Heart Failure and Cardiovascular Disease: An Update.

Galectin-3 is a versatile protein orchestrating several physiological and pathophysiological processes in the human body. In the last decade, considerable interest in galectin-3 has emerged because of its potential role as a biotarget. Galectin-3 is differentially expressed depending on the tissue type, however its expression can be induced under conditions of tissue injury or stress. Galectin-3 overexpression and secretion is associated with several diseases and is extensively studied in the context of fibrosis, heart failure, atherosclerosis and diabetes mellitus. Monomeric (extracellular) g

Galectin-3 as a novel regulator of osteoblast-osteoclast interaction and bone homeostasis.

Bone tissue undergoes permanent and lifelong remodeling with a concerted action of bone-building osteoblasts and bone-resorbing osteoclasts. A precise cooperation between those two cell types is critical in the complex process of bone renewal. Galectin-3 is a member of the β-galactoside-binding lectin family playing multiple roles in cell growth, differentiation and aggregation. As it has been described to be expressed in bone, galectin-3 might influence bone homeostasis by regulating the function and/or interplay of osteoblasts and osteoclasts. Here, we investigated the role of galectin-3 in

The impact of galectin-3 inhibition on aldosterone-induced cardiac and renal injuries.

OBJECTIVES: This study investigated whether galectin (Gal)-3 inhibition could block aldosterone-induced cardiac and renal fibrosis and improve cardiorenal dysfunction. BACKGROUND: Aldosterone is involved in cardiac and renal fibrosis that is associated with the development of cardiorenal injury. However, the mechanisms of these interactions remain unclear. Gal-3, a β-galactoside-binding lectin, is increased in heart failure and kidney injury. METHODS: Rats were treated with aldosterone-salt combined with spironolactone (a mineralocorticoid receptor antagonist) or modified citrus pectin (a Ga

Is galectin-3 antibody a useful marker in the diagnosis of malignant pleural mesothelioma?

Malignant pleural mesothelioma (MPM) is a challenging diagnosis characterized by the absence of real specific diagnostic markers. Positivity with the galectin-3 antibody was assessed by a cytoplasmic expression in 17 MPM. Fourteen cases expressed the galectin-3 antibody. The three negative cases consisted of epithelioid, biphasic, and sarcomatoid MPM. The 14 positive cases consisted of epithelioid MPM in 12 cases, sarcomatoid MPM in one case, and biphasic MPM in one case. In spite of our inability to prove the real diagnostic value of the galectin-3 antibody, our findings make us wonder about

Immunohistochemical study of galectin-3 in mature and immature bull testis and epididymis.

Galectin-3, a member of the beta-galactoside-binding protein family, has been implicated in mammalian sperm maturation. We examined galectin-3 expression in the testis and epididymis of sexually mature and immature bulls. Western blot analysis showed varying levels of galectin-3 in the bull testis and epididymis, and galectin-3 immunoreactivity was higher in the mature testis and epididymis than in immature organs. Galectin-3 was primarily localized in interstitial cells of the immature bull testis and in the peritubular myoid and interstitial cells of the mature testis. In the immature epidid

Value of pentraxin-3 and galectin-3 in acute coronary syndrome: a short-term prospective cohort study.

BACKGROUND: Acute coronary syndrome (ACS) continues to be a leading cause of morbidity and mortality worldwide. Galectin-3 and pentraxin-3 are two prognostic biomarkers that have been studied in heart failure (HF). However, there are limited data on these biomarkers in the ACS population. The objective of the study was to determine the variables that are most affected by high concentrations of pentraxin-3 and galectin-3, and the influence they have on outcomes of all-cause mortality in patients with ACS. METHODS: We included a total of 160 patients [ST elevation myocardial infarction (STEMI),

Galectin-1 and galectin-3 expression in equine mesenchymal stromal cells (MSCs), synovial fibroblasts and chondrocytes, and the effect of inflammation on MSC motility.

BACKGROUND: Mesenchymal stromal cells (MSCs) can be used intra-articularly to quell inflammation and promote cartilage healing; however, mechanisms by which MSCs mitigate joint disease remain poorly understood. Galectins, a family of β-galactoside binding proteins, regulate inflammation, adhesion and cell migration in diverse cell types. Galectin-1 and galectin-3 are proposed to be important intra-articular modulators of inflammation in both osteoarthritis and rheumatoid arthritis. Here, we asked whether equine bone marrow-derived MSCs (BMSCs) express higher levels of galectin-1 and -3 relativ

Macadamia oil alleviates brain injury induced by potassium dichromate via modulation of galectin-3/caspase-3 and Sitruin1.

The protein galectin-3 is essential for cell migration, activation, and proliferation. It also functions as an inflammatory mediator and triggers caspase-3, which causes brain damage. The purpose of this study was to unveil the phytochemical richness of macadamia nut oil (MO) and assess its potential as a treatment for brain damage caused by potassium dichromate (PDC), as well as its impact on galectin-3/caspase-3 and Sitruin1. The active ingredients of MO were determined by thorough analyses using GC-MS, HPLC, and spectrophotometry. These included oleic acid (47.53%), palmitoleic acid (16.48%

Mapping galectin-3 ligands in human tear fluid establishes spliceoform-dependent lacritin binding.

Galectin-3 (Gal-3) is a carbohydrate-binding protein which plays crucial roles in inflammation, immune response, and cell signaling. At the ocular surface, Gal-3 is also known to crosslink transmembrane mucins to support barrier function. However, the biological role of Gal-3 in circulating tear fluid remains largely unexplored. Similarly, whether Gal-3 engages extracellular glycoprotein ligands in tears to affect biological processes is unknown. Herein, we combine lectin ELISA, affinity enrichment, mass spectrometry (MS)-based glycoproteomics, and lectin blotting to uncover Gal-3 interactors

Timosaponin AIII alleviates type 2 diabetes-induced cardiomyopathy by targeting galectin-3 (LGALS3).

Timosaponin AIII, the main active saponin derived from Anemarrhena asphodeloides Bunge, exerts anti-diabetic effects. However, its underlying mechanism in diabetic cardiomyopathy (DCM) remains unclear. In this study, we explored the pharmacological effects of timosaponin AIII on DCM using a mouse model and H9c2 cells. We found that timosaponin AIII treatment attenuated cardiac remodeling, myocardial fibrosis, and cardiomyocyte apoptosis in high-fat diet (HFD) combined with streptozotocin (STZ)-induced diabetic mice. Additionally, it significantly reduced galectin-3 expression in HFD/STZ-induce

Galectin-3 gene deletion alters lipid profile in healthy and cardiomyopathy mouse hearts.

Galectin-3 (Gal3) is known to interact with glycans of proteins and lipids. In cardiovascular disease, the elevated expression of Gal3 mediates inflammation, hypertrophy and fibrosis. We explored the effect of Gal3 on the cardiac lipid profile in healthy mice or in mice with dilated cardiomyopathy (DCM). Using lipidomics and gene-targeted mouse models, we studied the influence of Gal3 gene deletion on cardiac lipid profiles in the healthy mice or mice with DCM. Cardiac-restricted transgenic activation of Hippo pathway led to DCM phenotype and Gal3 upregulation. DCM mice were cross-bred with Ga

Plasma galectin-3 is associated with decreased glomerular filtration rate in chronic HIV.

BACKGROUND: People living with HIV (PLWH) have higher rates of chronic kidney disease (CKD) compared with HIV-uninfected individuals. The pathogenesis of CKD in HIV remains poorly understood but is likely from a combination of various factors, such as traditional comorbidities, prolonged antiretroviral therapy, immune dysregulation, and direct HIV effect on the kidneys. We evaluated plasma galectin-3 (Gal-3), a circulating marker of fibrosis, and its association with renal function. METHODS: Estimated glomerular filtration rate (eGFR) was assessed by CKD-EPI. Plasma galectin-3 was obtained fr

Galectin-3, a rising star in modulating microglia activation under conditions of neurodegeneration.

The advent of high-throughput single-cell transcriptomic analysis of microglia has revealed different phenotypes that are inherently associated with disease conditions. A common feature of some of these activated phenotypes is the upregulation of galectin-3. Representative examples of these phenotypes include disease-associated microglia (DAM) and white-associated microglia (WAM), whose role(s) in neuroprotection/neurotoxicity is a matter of high interest in the microglia community. In this review, we summarise the main findings that demonstrate the ability of galectin-3 to interact with key p
