The complexity of the GABAA receptor shapes unique pharmacological profiles.

Gamma-amino butyric acid (GABA) is the most abundant inhibitory neurotransmitter in the central nervous system (CNS) and many physiological actions are modulated by GABA(A) receptors. These chloride channels can be opened by GABA and are a target for a variety of important drugs such as benzodiazepines, barbiturates, neuroactive steroids, convulsants and anaesthetics. GABA(A) receptors are involved in anxiety, feeding and drinking behaviour, circadian rhythm, cognition, vigilance, and learning and memory. Moreover, deficits in the functional expression of GABA(A) receptors have been implicated

Self-Grounded Vision: Hand Ownership Modulates Visual Location through Cortical β and γ Oscillations.

UNLABELLED: Vision is known to be shaped by context, defined by environmental and bodily signals. In the Taylor illusion, the size of an afterimage projected on one's hand changes according to proprioceptive signals conveying hand position. Here, we assessed whether the Taylor illusion does not just depend on the physical hand position, but also on bodily self-consciousness as quantified through illusory hand ownership. Relying on the somatic rubber hand illusion, we manipulated hand ownership, such that participants embodied a rubber hand placed next to their own hand. We found that an afteri

Theta-Gamma Subthalamic Stimulation for Verbal Fluency in Parkinson's Disease: A Randomized, Crossover Trial.

BACKGROUND: High-frequency deep brain stimulation (DBS) of the subthalamic nucleus (STN) improves Parkinson's disease (PD) motor symptoms but may deteriorate verbal fluency (VF). Theta stimulation showed potential cognitive benefits associated with motor worsening. OBJECTIVES: This randomized, double-blind, crossover study evaluated the efficacy and safety of combined theta-gamma frequency stimulation on VF in PD patients with STN-DBS. METHODS: Patients were randomized 1:1 for standard or theta-gamma stimulation. VF, motor, and non-motor symptoms were assessed at baseline, 1 h, and 1 month a

Noise-induced neurophysiological alterations in the rat medial geniculate body and thalamocortical desynchronization by deep brain stimulation.

The thalamic medial geniculate body (MGB) is uniquely positioned within the neural tinnitus networks. Deep brain stimulation (DBS) of the MGB has been proposed as a possible novel treatment for tinnitus, yet mechanisms remain elusive. The aim of this study was to characterize neurophysiologic hallmarks in the MGB after noise exposure and to assess the neurophysiological effects of electrical stimulation of the MGB. Fourteen male Sprague-Dawley rats were included. Nine subjects were unilaterally exposed to a 16-kHz octave-band noise at 115 dB for 90 min, five received sham exposure. Single unit

Modulation of auditory gamma-band responses using transcranial electrical stimulation.

Auditory gamma-band (>30 Hz) activity is a biomarker of cortical excitation/inhibition (E/I) balance in autism, schizophrenia, and bipolar disorder. We provide a comprehensive account of the effects of transcranial alternating current stimulation (tACS) and transcranial direct current stimulation (tDCS) on gamma responses. Forty-five healthy young adults listened to 40-Hz auditory click trains while electroencephalography (EEG) data were collected to measure stimulus-related gamma activity immediately before and after 10 min of 1 mA tACS (40 Hz), tDCS, or sham stimulation to left auditory cort

Discordant Alpha-Band Transcranial Alternating Current Stimulation Affects Cortico-Cortical and Cortico-Cerebellar Connectivity.

Synchronization of oscillatory brain activity is believed to play a critical role in linking distributed neuronal populations into transient functional networks. Alpha-band alternating current stimulation (tACS) was applied over bilateral parietal cortex in a double-blind sham-controlled study to test the notion that widespread alpha mediates causal relationships in the gamma-band both within local neuronal populations and also across distant brain regions. Causal relationships of oscillatory alpha- and gamma-band activity were characterized during performance of a visual global/local attentio

Dysregulated oscillatory connectivity in the visual system in autism spectrum disorder.

Autism spectrum disorder is increasingly associated with atypical perceptual and sensory symptoms. Here we explore the hypothesis that aberrant sensory processing in autism spectrum disorder could be linked to atypical intra- (local) and interregional (global) brain connectivity. To elucidate oscillatory dynamics and connectivity in the visual domain we used magnetoencephalography and a simple visual grating paradigm with a group of 18 adolescent autistic participants and 18 typically developing control subjects. Both groups showed similar increases in gamma (40-80 Hz) and decreases in alpha (

Endogenously generated gamma-band oscillations in early visual cortex: A neurofeedback study.

Human subjects were trained with neurofeedback (NFB) to enhance the power of narrow-band gamma oscillations in circumscribed regions of early visual cortex. To select the region and the oscillation frequency for NFB training, gamma oscillations were induced with locally presented drifting gratings. The source and frequency of these induced oscillations were determined using beamforming methods. During NFB training the power of narrow band gamma oscillations was continuously extracted from this source with online beamforming and converted into the pitch of a tone signal. We found that seven out

Simultaneous analysis of the LFP and spiking activity reveals essential components of a visuomotor transformation in the frontal eye field.

The frontal eye field (FEF) is a key brain region to study visuomotor transformations because the primary input to FEF is visual in nature, whereas its output reflects the planning of behaviorally relevant saccadic eye movements. In this study, we used a memory-guided saccade task to temporally dissociate the visual epoch from the saccadic epoch through a delay epoch, and used the local field potential (LFP) along with simultaneously recorded spike data to study the visuomotor transformation process. We showed that visual latency of the LFP preceded spiking activity in the visual epoch, wherea

Beta to low-gamma rhythmic brain stimulation over primary motor cortex and supplementary motor area differentially modulates oscillatory neural drives during gait.

Non-invasive brain stimulation such as transcranial direct current stimulation over the primary motor cortex (M1) and supplementary motor area (SMA) during gait can positively affect gait ability in patients after stroke; however, the frequency-specific modulatory effects of rhythmic brain stimulation over the M1 and SMA on the oscillatory neural drives during gait remain unclear. Therefore, we investigated the effects of the alpha and beta to low-gamma oscillatory transcranial direct current stimulation (otDCS) over the M1 and SMA on the oscillatory neural drives to lower limb muscles during

Frontal theta-gamma ratio is a sensitive index of concussion history in athletes on tasks of visuo-motor control.

Patients with mTBI often show deficits in executive function and changes in neural activity. Similar changes in those with a history of mTBI (i.e. concussion), however, have not been consistently reported. Frontal theta-to-gamma frequency ratio has shown promise in EEG research in predicting performance on working memory tasks. In the present study we explored the sensitivity of the frontal theta-to-gamma relative power spectral density (PSD) ratio to the history of concussion in 81 youth athletes (18 with a history of concussion, ages 13-18) during the tests of the Nike Sensory Training Stati

Children with Autism Spectrum Disorder Demonstrate Regionally Specific Altered Resting-State Phase-Amplitude Coupling.

Studies suggest that individuals with autism spectrum disorder (ASD) exhibit altered electrophysiological alpha to gamma phase-amplitude coupling (PAC). Preliminary reports with small samples report conflicting findings regarding the directionality of the alpha to gamma PAC alterations in ASD. The present study examined resting-state activity throughout the brain in a relatively large sample of 119 children with ASD and 47 typically developing children. Children with ASD demonstrated regionally specific abnormalities in alpha to low-gamma PAC, with increased alpha to low-gamma PAC for a centra

Zolpidem restores sleep and decreases amyloid in a mouse model.

INTRODUCTION: Deficits in non-rapid eye movement (NREM) sleep facilitate Alzheimer's disease (AD) progression. Enhancing gamma-aminobutyric acid-ergic (GABAergic) signaling can restore sleep. Unbiased computational analysis identified zolpidem as high-affinity GABA receptor modulator facilitating chloride transport that could slow AD. METHODS: Zolpidem's effects on sleep and Alzheimer's progression were evaluated in young APP/PS1 (amyloid precursor protein/presenilin 1) mice. Sleep was monitored with electroencephalography/electromyography (EEG/EMG) telemetry. Wide-field imaging with voltage-

Inter-hemispheric functional coupling of eyes-closed resting EEG rhythms in adolescents with Down syndrome.

OBJECTIVE: We tested the hypothesis that inter-hemispheric directional functional coupling of eyes-closed resting EEG rhythms is abnormal in adolescents with Down syndrome (DS). METHODS: Eyes-closed resting EEG data were recorded in 38 DS adolescents (18.7 years +/-0.67 SE, IQ=49+/-1.9 SE) and in 17 matched normal control subjects (NYoung=19.1 years +/-0.39 SE). The EEG data were recorded from 8 electrodes (Fp1, Fp2, C3, C4, T3, T4, O1, O2) referenced to vertex. EEG rhythms of interest were delta (2-4 Hz), theta (4-8 Hz), alpha 1 (8-10.5 Hz), alpha 2 (10.5-13 Hz), beta 1 (13-20 Hz), and beta

The corticothalamic system in sleep.

The transition from wakefulness to NREM sleep is associated with typical signs of brain electrical activity, characterized by prolonged periods of hyperpolarization and increased membrane conductance in thalamocortical (TC) neurons, with the consequence that incoming messages are inhibited and the cerebral cortex is deprived of signals from the outside world. There are three major oscillations during NREM sleep. Spindles are generated within the thalamus, due to thalamic reticular (RE) neurons that impose rhythmic inhibitory sequences onto TC neurons, but the widespread synchronization of this

Can antiarrhythmic drugs save lives in patients with congestive heart failure?

Although arrhythmic death is a common cause of death in patients with congestive heart failure (CHF), numerous trials involving prophylactic antiarrhythmic drug treatment have yielded few gains. To date, only beta-blockers have shown a distinct mortality-reducing effect and despite the antiarrythmic effect of gamma-blockers, results point towards causes other than the antiarrhythmic effect in obtaining this beneficial effect. Atrial fibrillation is an often-encountered arrhythmia in patients with CHF and recent trials have cast doubt on the present treatment strategy of persistently striving t

Enhancing GABAergic signaling ameliorates aberrant gamma oscillations of olfactory bulb in AD mouse models.

BACKGROUND: Before the deposition of amyloid-beta plaques and the onset of learning memory deficits, patients with Alzheimer's disease (AD) experience olfactory dysfunction, typified by a reduced ability to detect, discriminate, and identify odors. Rodent models of AD, such as the Tg2576 and APP/PS1 mice, also display impaired olfaction, accompanied by aberrant in vivo or in vitro gamma rhythms in the olfactory pathway. However, the mechanistic relationships between the electrophysiological, biochemical and behavioral phenomena remain unclear. METHODS: To address the above issues in AD models

State-Dependent Effects of Transcranial Oscillatory Currents on the Motor System during Action Observation.

We applied transcranial alternating current stimulation (tACS) to the primary motor cortex (M1) at different frequencies during an index-thumb pinch-grip observation task. To estimate changes in the corticospinal output, we used the size of motor evoked potentials (MEPs) obtained by transcranial magnetic stimulation (TMS) of M1 using an online MRI-guided simultaneous TMS-tACS approach. The results of the beta-tACS confirm a non-selective increase in corticospinal excitability in subjects at rest; an increase was observed for both of the tested hand muscles, the first dorsal interosseous (FDI)

Distinct laminar origins of sensory-evoked high-gamma and low-frequency ECoG signals revealed by optogenetics.

Electrocorticography (ECoG) provides a high-spatiotemporal-resolution measure of cortical activity (cortical surface electrical potentials, CSEPs) in humans and animals. The CSEP high-gamma band (Hγ, 65 to 170 Hz) correlates with neuronal firing rates at the columnar spatial scale and is widely used as a biomarker of local activity. Whether Hγ reports all stages of columnar processing, intermediate processing in L2/3 (close to the ECoG electrode), or the main columnar output in L5, is unknown. We disentangled the laminar origins of Hγ and other ECoG bands by optogenetically suppressing L2/3 or
