Sulfur dioxide (SO(2)) and benzo(a)pyrene (B(a)P) are common industrial and environmental contaminants. However, few data are available on the effects of SO(2) on proto-oncogenes and tumor suppressor genes, as well as the interactions between SO(2) and other xenobiotics regulating proto-oncogenes or tumor suppressor genes expression. To investigate the interactions between SO(2) and B(a)P, male Wistar rats were exposed to intratracheally instilled with B(a)P or SO(2) inhalation alone or together. We detected mRNA expression of CYP1A1 and 1A2, 7-ethoxyresorufin O-deethylase (EROD), and methoxyr

PURPOSE: High levels of visible light induce apoptotic cell death of photoreceptors, a process depending on the activation of the transcription factor AP-1. This suggests that regulation of gene expression might be important for light-induced photoreceptor cell death. We measured expression of AP-1 family members and of several apoptosis-related genes to test their potential involvement in photoreceptor apoptosis.
METHODS: Wildtype and c-fos-/- mice were exposed to low (roomlight) or high levels of visible light for up to two hours. Total RNA was prepared from isolated retinas during and afte

Constitutive expression of the cellular proto-oncogenes c-fos and c-jun, and in a lesser extent ras, was demonstrated in the glioma cell line C-6 by flow cytometry analysis using specific mono and polyclonal antibodies. Basal expression of the products of the early response genes c-fos and c-jun were increased 66 and 50% when Theiler's murine encephalomyelitis virus (TMEV) infected these cells. No increase in ras transcription could be demonstrated after infection. This activation follows a kinetic reaching maximum values after 60 min and was proportional to the multiplicity of infection used.

Macrophages, which serve as a bridge between innate and adaptive immunity, play an important role in sporotrichosis. Sporothrix schenckii infections can produce immune responses such as macrophage polarization and inflammatory factor secretion. In the early stages of inflammation, the expression of DAB2 in macrophages is increased, which controls the secretion of inflammatory factors and affects the polarization of macrophages. However, the expressions and mechanisms of DAB2 in sporotrichosis are not clear. In this study, we examined the expression of DAB2 and its regulation of inflammatory fa

Apoptotic injury participates in hepatic fibrosis, but the molecular mechanisms are not well understood. The present study aimed to investigate the role of inducible TIMP1 in the pathogenesis of hepatic apoptosis-fibrosis. Apoptosis was induced with GCDC, LPS, and alcohol in precision-cut liver slices or bile duct ligation (BDL) in rats, as reflected by caspase-3 activity, TUNEL assay, and apoptosis-related gene profiles. The hepatic fibrosis was detected with Picrosirius staining, hydroxyproline determination, and expression profiling of fibrosis-related genes. Levels of TIMP1 were upregulate

Acute myeloid leukemia (AML) is a biologically heterogeneous disease originating from the clonal expansion of hematopoietic stem cells (HSCs). Clonal expansion of hematopoietic stem cell progenitors (HSC-Prog), along with a block in differentiation, are hallmark features of AML. The disease is characterized by poor clinical outcomes, highlighting the urgent need for effective therapeutic strategies and suitable drug targets. We conducted multi-omics analyses, including single-cell RNA sequencing (scRNA-seq), Mendelian randomization (MR), and bulk RNA-seq, to investigate HDAC1's oncogenic role

Spinal cord injury (SCI) initiates a cascade of complex secondary damage processes, prominently involving programmed cell death (PCD). Although apoptosis and necroptosis have been extensively characterized, the role of oncosis in SCI remains inadequately understood. In this study, we examined the expression dynamics and cellular localization of oncosis-related genes (ORGs) following SCI. We conducted an analysis of bulk RNA-seq data to identify differentially expressed ORGs at five distinct time points post-injury. Six candidate genes (Trp53, Casp3, Jun, Tmem123, Chmp6, Map2) were identified b

BACKGROUND: Proliferation and IL-2 production in response to T-cell receptor ligation are impaired in patients with Wiskott-Aldrich syndrome (WAS). The transcription factors nuclear factor-kappaB (NF-kappaB), nuclear factor of activated T cells (NF-AT), and activating protein-1 (AP-1) play a critical role in IL-2 gene expression.
OBJECTIVE: To investigate the mechanisms of impaired IL-2 production after T-cell receptor ligation in T cells deficient in WAS protein (WASP).
METHODS: T cells from WASP-/- mice were stimulated with anti-CD3 and anti-CD28. Nuclear NF-kappaB, NF-AT, and AP-1 DNA-bin

Intrauterine growth-restricted (IUGR) fetal sheep have increased hepatic glucose production (HGP) that is resistant to suppression during a hyperinsulinemic-isoglycemic clamp (insulin clamp). We hypothesized that the IUGR fetal liver would have activation of metabolic and signaling pathways that support HGP and inhibition of insulin-signaling pathways. To test this, we used transcriptomic profiling with liver samples from control (CON) and IUGR fetuses receiving saline or an insulin clamp. The IUGR liver had upregulation of genes associated with gluconeogenesis/glycolysis, transcription factor

OBJECTIVE: Rheumatoid arthritis (RA) is a chronic autoimmune joint disease driven by dysregulated immune cells and transcription factors. Despite known molecular alterations, systematic screening of key biomarkers and their link to the immune microenvironment remains lacking, particularly regarding extensive multi-algorithm cross-validation across multiple independent cohorts. This study employs bioinformatics and machine learning to identify potential RA biomarkers, aiming to support diagnosis and targeted therapy.
METHODS: Multiple RA-related transcriptomic datasets derived from synovial ti

The expression of galanin (GAL) in L5 dorsal root ganglia (DRG) and calcitonin gene-related peptide (CGRP) in motoneurons (MN) of lumbar spinal cord and their colocalisation with the nuclear c-JUN protein was investigated by immunocytochemistry following transection of rat sciatic nerve. Expression of c-JUN in L5 DRG neurons increased 10 h following transection. Between 24 h and 10 days 64%-72% of all neurons were labelled. After 50 and 150 days, the end of the observation period, 62% and 27%, respectively, of neurons were labelled by c-JUN. Expression of GAL started after 24 h, reached a maxi

Light therapy, using red and near-infrared (NIR) irradiation, is currently applied for the treatment of various neurodegenerative diseases, such as Alzheimer's disease (AD). Transcranial photobiomodulation therapy (tPBMT) can alleviate neurodegeneration, neuronal loss, and β-amyloid peptide plaque burden. Alternatively, potential early inhibition of oxidative stress, neuroinflammation, apoptosis, and amyloidogenic cellular pathways may constrain pathological changes with aging. In this research, we conduct an 808-nm tPBMT with a 30-day course of daily 1-hour sessions for mice and assess its in

Heme oxygenase type 2 (HO-2) is an enzyme that uses heme as a substrate to produce iron, biliverdin, and carbon monoxide (CO). This enzyme participates in regulation of nociceptive signal transmission in spinal cord tissue. We set out to identify genes undergoing alterations in expression in a model of inflammatory pain and to determine whether HO-2 participates in that regulation. After the hindpaw injection of formalin in mice, we measured changes in expression of immediate early genes including c-fos, c-jun, jun B, nerve growth factor induced genes (NGFI-A and NGFI-B) and activity-related c

AIMS: Depression significantly increases the risk of cardiovascular diseases (CVD) by exacerbating hypothalamic-pituitary-adrenal (HPA) axis hyperactivity and oxidative stress damage, common risk factors for both conditions. Our study aims to elucidate how elevated corticosterone levels in mice induced by chronic unpredictable mild stress (CUMS), an experimental model showing depression-like behaviors, contribute to the cardiac oxidative damage and dysfunction, and to assess the protective role of tauroursodeoxycholic acid (TUDCA) in this process.
RESULTS: CUMS mice show electrocardiogram abn

An early and immediate response of cells upon irradiation with UV light and various other forms of genotoxic stress is the induction of the proto-oncogenes c-fos and c-jun. To address the questions of whether (a) methylating agents that are powerful carcinogens are effective in induction of fos and jun mRNAs, (b) induction is affected by the repair capacity of the cells, and (c) induction is accompanied by genotoxic effects, the levels of c-fos, c-jun, junB and junD mRNA were analysed in isogenic Chinese hamster cell lines deficient (phenotypically Mex-) and proficient (Mex+) for the DNA repai

Widespread aberrant gene expression is a pathological hallmark of polycystic kidney disease (PKD). Numerous pathogenic signaling cascades, including c-Myc, Fos, and Jun, are transactivated. However, the underlying epigenetic regulators are poorly defined. Here we show that H3K27ac, an acetylated modification of DNA packing protein histone H3 that marks active enhancers, is elevated in mouse and human samples of autosomal dominant PKD. Using comparative H3K27ac ChIP-Seq analysis, we mapped over 16000 active intronic and intergenic enhancer elements in Pkd1-mutant mouse kidneys. We found that th
